试验选用荷斯坦泌乳早期奶牛15头,采用3X3复拉丁方试验设计,研究日粮DCAB水平分别为+367、+514、+663mmol/kgDM对奶牛生产性能、血液酸碱平衡和血清矿物质含量的影响。试验结果,不同DCAB水平日粮对奶牛奶产量差异不显著(P>0.05),高DCAB水平日粮可增加奶牛血液标准HCO-3含量(P<0.05),不同DCAB水平对奶牛各种血清常量矿物质含量没有影响(P>0.05)。试验表明,日粮DCAB水平超过+514mmol/kgDM可对奶牛奶产量产生不利影响。  相似文献

During the summers of 2001 and 2002, hourly average ozone concentrations were measured at three sites of differing elevation (188, 588, and 1221 m.a.s.l.) on the forested south-facing slopes of the Lower Fraser Valley (LFV), British Columbia. Sites experienced ozone concentrations ranging from 0 to 88 ppb in 2001, and 0 to 96 ppb in 2002. Daily patterns were in agreement with previous studies showing morning increases and late afternoon peaks. Reduced diurnal variation increased the exposure of higher-elevation forested sites. An upper-level ridge coinciding with a thermal coastal trough caused above-average ozone concentrations, and the ‘maximum acceptable’ 1-hour National Ambient Air Quality Objective (AQO) of 82 ppb to be exceeded. Maximum ozone concentrations and AQO exceedance frequency both increased with distance eastward in the valley. A preliminary survey of ozone-like injury symptoms on native shrubs suggested that the elevated ozone levels occurring in the LFV may cause injury to forest plants.  相似文献

OBJECTIVE: To isolate bovine mammary gland cells with stem cell characteristics. SAMPLE POPULATION: Monolayers of bovine mammary gland cells. PROCEDURE: Mammary gland cell populations were separated by use of selected media supplements. Phenotypic characteristics were examined via light and transmission electron microscopy. Cellular expression of casein and connexin 43 was identified immunohistochemically. A scrape-loading and dye transfer assay was used to examine the mammary gland cell populations for homogenous gap junctional intercellular communication (GJIC). RESULTS: Subpopulations of mammary gland cells grown in vitro are classified on the basis of their distinct morphologic features and ability to communicate via gap junctions. Ultrastructurally, 2 morphologically distinct cell types were classified as type I and II cells. Type I cells were small light undiffertiated cells and large light undifferentiated cells that were deficient in functional gap junctions (as is characteristic of stem cells). Type II cells included large light differentiated cells and terminally differentiated cells; GJIC was functional in type II cells. Type II cells had cytoplasmic expression of connexin 43, whereas, type I cells did not. All cells expressed casein. CONCLUSIONS AND CLINICAL RELEVANCE: Subpopulations of bovine mammary gland cells with stem cell characteristics were identified. Phenotypic differences are observed among type I bovine mammary gland cells with stem cell characteristics. Gap junctional intercellular communication may be necessary for the differentiation of stem cells. Characterization of bovine mammary gland stem cells and their progeny may provide a new tool with which to study mammary gland health.  相似文献

A Pyrenophora teres f. teres population in North Dakota was analyzed for virulence variation and genetic diversity using 75 monospore isolates that were collected across a 4-year period (2004 to 2007) from two North Dakota State University agricultural experiment stations at Fargo and Langdon. Pathogenicity tests by inoculation onto 22 barley differential lines at seedling stage revealed 49 pathotypes, indicating a wide range of pathogenic diversity. Two-way analysis of variance of disease ratings revealed a significant difference in the virulence among isolates and in the resistance among barley lines, as well as in the interactions between the two. 'CI5791', 'Algerian', and 'Heartland' were three barley lines showing a high level of seedling resistance to all North Dakota isolates tested; however, many previously reported resistance genes have been overcome. Forty multilocus genotypes were identified from this set of isolates by genotyping at 13 simple-sequence repeat loci. High percentages of clonal cultures were detected in the samplings from 2005 and 2007 in Fargo and 2005 in Langdon. Using a clone-corrected sample set, the mean gene diversity (h) was estimated to be 0.58, approximately the same for both locations. The calculated Wright's F(ST) value is small (0.11) but was significantly >0, indicating a significant differentiation between the Fargo and Langdon populations. In the gametic disequilibrium test, only 3 of 78 possible pairwise comparisons over all isolates showed significant (P < 0.05) nonrandom association, suggesting a random mating mode. Our results suggest that the populations from the two locations are derived from a common source and undergo frequent recombination. This research provides important information for barley breeders regarding development and deployment of cultivars with resistance to net form net blotch in this region.  相似文献

Research has documented immobilization of rodents, rabbits, guinea pigs and dogs by mechanical means, typically using neck clips or inversion ('animal hypnosis'). In contrast, only a few studies of mechanical immobilization of cats are available, although some success has been reported in the literature. Domestic cats may be effectively immobilized by clips placed along the animal's dorsum. We use the term 'pinch-induced behavioral inhibition' (PIBI) for this behavior because it describes both the method and the response, while avoiding the more anthropomorphic term 'hypnosis'. We investigated the effectiveness of PIBI and its neurological and habituation effects in healthy cats and cats with idiopathic cystitis (IC). Although not all cats were susceptible to PIBI and effectiveness varied among individuals, PIBI was useful for gentle restraint in most cats.  相似文献

We examined runoff and sediment transport associated with large-scale construction projects in Fujian Province, China. Six experimental plots, comprised of four plot types, were designed to mimic typical conditions immediately following disturbance. It was found that natural vegetative cover reduces both runoff and erosion by approximately 36 and 7457 times over bare ground, and 16 and 1801 times over typically planted grasses, respectively. The increase in erosion associated with the replacement of native vegetation with bare ground or grasses due to large-scale infrastructure projects in Fujian from 1999 to 2004, amounted to an estimated loss of 1.76 × 107 tonnes of top soil and 3.04 × 108 m3 of surface runoff from the province during the bare soil construction phase, and an additional 4.25 × 106 tonnes of top soil and 1.35 × 108 m3 of surface runoff from the province associated with the first year of operation for each project. This has implications for frequency and occurrence of landslides and other geographic hazards, the transport of chemicals into waterways, the transport of goods through shipping passages, and the fertility of land in Fujian.  相似文献

A flock of Indian Ringneck parakeets (Psittacula krameri manillensis) was imported to the United States from Australia. Soon after, 1 parakeet suddenly died, and a second parakeet died after a 2-day course of illness, which consisted of anorexia, lethargy, emaciation, and dyspnea. At necropsy, the affected birds had diffuse consolidation and red discoloration of the lungs, as well as thickened, congested air sacs. The microscopic examination revealed multifocal, necrotizing bronchitis, parabronchitis, and interstitial pneumonia. The lumen of the affected airways contained numerous, large syncytial cells with up to 15 nuclei. The nuclei of these syncytial cells often contained large, eosinophilic inclusion bodies, consistent with herpesvirus. The epithelium of the trachea and air sacs was hypertrophied and contained syncytial cells with intranuclear inclusion bodies similar to the bronchi. In addition, a few intranuclear inclusion bodies were also present in the epithelial cells that line the air capillaries. On ultrastructural examination, the nuclei of degenerating epithelial cells contained clusters of viral nucleocapsid proteins and unenveloped, icosahedral, viral particles that were approximately 90 nm in diameter. In addition, some epithelial cells contained clusters of enveloped viral particles approximately 105 nm in diameter, within the cytocavitary network. These lesions are characteristic of those caused by respiratory herpesvirus of parakeets.  相似文献
