In a flock of sheep to which food with excess copper (= Cu) had been fed for some months 39 animals died of chronic copper toxicosis. In 5 sheep of this flock the plasma concentrations of the enzymes glutamate dehydrogenase (GLDH), sorbitol dehydrogenase (SDH), aspartate-amino-transferase (ASAT) and copper were measured several times over a period of 100 days. At day 23, 58 and 97 needle biopsies of the liver were taken. The biopsies were used for histological examination and Cu-analysis. After 100 days the sheep were slaughtered and chronic copper poisoning was diagnosed by raised Cu concentration and typical histological alterations in the liver. The activities of GLDH and SDH in the plasma of the 5 sheep were consistently above normal, ASAT activity and the plasma Cu level were raised only occasionally. The results of Cu determination in biopsy samples were of little predictive value compared with post mortem Cu analysis of the liver.  相似文献

Background, Aim and Scope   Part 1: Behaviour of Polycyclic Musks in Sewage Sludge of Different Treatment Plants in Summer and Winter Part 2: Investigation of Polycyclic Musks in Soils and Plants -  Preamble. In Part 1 of the study, screening tests were performed to investigate the occurrence of PCMs in sewage sludges. For a preliminary risk assessment, further information is needed about their behaviour in the terrestrial environment. Hence, Part 2 examined the adsorption of PCMs to soil, their dissipation and leaching in soil and their uptake by plants. Background, Aim and Scope   Polycyclic Musks (PCMs) enter the environment via the waste water system. Because of their persistence, they can accumulate in different matrices like sewage sludge or biota. By the use of sewage sludge as a fertilizer, PCMs are transferred to agricultural soils. Therefore, in Part 1 of the study, screening tests were performed to investigate the occurrence of PCMs in sewage sludge. For a preliminary risk assessment, further information is needed about their behaviour in the terrestrial environment. Hence, Part 2 of the study examined the adsorption of PCMs to soil, their dissipation and leaching in soil, and their uptake by plants. Materials and Methods: In the screening study, samples of activated sewage sludge were taken both in summer and in winter at 21 treatment plants. In order to get an overview of the contamination situation, sampling covered different types of treatment plants (in rural, urban, industrial areas). Analytical methods for the determination of HHCB, AHTN, ADBI, ATTN, AHDI and ATII in the sludge samples were developed and applied. Results: The analytical screening of PCMs showed their presence in activated and dried sewage sludge samples. HHCB and AHTN represented about 95% of the PCMs investigated. Their concentrations in the activated sludge samples varied between 2.9 and 10.4 mg/kg dry mass (dm) and 1.1 to 4.2 mg/kg dm, respectively. Although different types of sewage treatment plants were investigated, similar PCM levels were found, showing the widespread input of these compounds into domestic waste water. Discussion: PCM concentrations in activated sludge varied widely. The variation drops substantially when concentrations are related to the varying dry mass. In dehydrated sludge, PCM concentrations were up to 24 mg/kg dm for HHCB and up to 6.9 mg/kg dm for AHTN. These high values are comparable to those obtained in other investigations analysing PCMs. If the degradation of organic mass during anaerobic decomposition is included in the evaluation, the figures obtained are comparable to those for activated sludge. Elimination in sewage sludge was higher in summer than in winter. Therefore, the contamination of the sludges in winter reached higher levels compared to the summer. Conclusions: The results show that PCMs are widespread contaminants in sewage sludge. Recommendations and Perspectives: PCM should be considered in a risk assessment as potential contaminants of sewage sludge destined for agricultural use. Due to the high PCM levels in sewage sludge, further investigations into the degradation and elimination behaviour in sewage sludge have to be carried out, including that involving PCM metabolites such as lactone derivatives.  相似文献

Cereals contaminated by Aspergillus spp., Penicillium spp., and Fusarium spp. and their mycotoxins, for example, ochratoxin A (OTA) and deoxynivalenol (DON), are not only a risk to human and animal health but can also show poor technological properties and baking quality. The influence of these genera on the sulfur speciation of low molecular weight (LMW) subunits of glutenin was characterized by investigating suboptimally stored wheat samples in situ by X-ray absorption near edge structure (XANES) spectroscopy and baking tests. Field fungi of the genus Fusarium have hardly any influence on both the sulfur speciation of wheat gluten proteins and the baking properties, whereas storage fungi of the genera Aspergillus and Penicillium have a direct influence. An increased amount of sulfur in sulfonic acid state was found, which is not available for thiol/disulfide exchange reactions in the gluten network, and thus leads to a considerably reduced baking volume. From changes of the composition of the mould flora during suboptimal storage of wheat and from the mycotoxin contents, it can be concluded that microbial competitive interactions play an important role in the development of the mould flora and the mycotoxin concentrations during (suboptimal) storage of wheat.  相似文献

The occurrence of BSE cases in Germany after the ban of meat and bone meal for ruminant feed in 1994 requires a detailed investigation of animal derived feedstuffs regarding their specific risks as vectors for the disease. Accepting the theory that BSE is a prion transmitted disease, the theoretical infectious potential was calculated for animal derived feedstuffs. This calculation was based on the assumption, that risk material (brain, spinal cord) of one clinically diseased cattle was rendered in the process as established in Germany (133 degrees C, 3 bar, 20 min) or, alternatively, that one diseased animal was slaughtered resulting in normal processing of the by-products for human food production. From this risk assessment it became obvious that meat and bone meal was one, but probably not the most important source for the spreading of BSE. Taking into account the high sensitivity of calves it can be speculated that certain products, e.g. from bone processing (bone meal) and fat melting (mixed animal fats), commonly used for the formulation of milk replacers, might have been more important as pathways. As it can't be excluded retrospectively that infected meat and bone meal was imported from the UK, this non-calculable influence may have been related to the significance of the other products. The calculation model underlines that efficient removal of specified risk material (brain, spinal cord) and adequate processing (133 degrees C, 3 bar, 20 min) or alternatively other equivalent treatments of fats are prerequisites for minimising the risk of feed borne transmission of BSE by animal derived feedstuffs. The spatial distribution of olive fruit flyBactrocera (Dacus) oleae (Gmelin) (Diptera: Tephritidae) field captures with McPhail traps within an experimental orchard was evaluated. Contour maps were constructed to examine the patterns in the 3-year trapping data. Captures varied widely inside the olive orchard, with traps suspended on wild olive trees exhibiting the poorest performance. Favorable microclimate, created by a standing water pool, appeared to be responsible for increased trap captures during the hot summer months. In reared piglets (n = 10), fattening pigs (n = 20) and sows (n = 8) the effects of lactulose as feed additive on the quality and composition of chyme and faeces were tested to find out potential dietetic advantages of its use. The lactulose concentration in the diets varied between 27-29 g/kg dry matter in piglets and fattening pigs and 55-140 g/kg dry matter in the diet of sows. In piglets and fattening pigs the lactulose intake did not alter the dry matter content and pH in faeces; in spite of the higher concentration lactulose did not result in diarrhea in sows or adult minipigs, there was only a slight decrease in the dry matter content of faeces. In contrast to findings in human beings only traces of lactulose reached the hind gut, although higher lactulose concentrations were measured at the end of the small intestine (in piglets: 10-40 g, in fattening pigs 40-70 g lactulose per kg dry matter of chyme). The oral intake of lactulose did not increase the concentration of lactic acid or volatile fatty acids in the content of the small intestine and did not reduce the pH in the chyme, but in the contents of cecum and colon ascendens higher concentrations of volatile fatty acids were measured (in piglets: significantly higher values, in fattening pigs slightly increased concentrations). In piglets as well as in fattening pigs lactulose resulted in reduced concentrations of ammonia in the content of small and large intestine, further more the concentrations of lipopolysaccharides were increased in the colonal chyme, when pigs ingested the lactulose containing diet. Due to the fact that only small amounts of lactulose reached the hind gut there are doubts, whether lactulose has comparable dietetic effects in pigs as known from human beings (for example laxative effects, forced elimination of bacteria like salmonella). The results obtained here in pigs underline the necessity of experimental studies in the target species (including parameters of digestive physiology, chemical/microbial composition of the chyme) before substances are used and recommended for dietetic purposes in feeding practice. In spite of generally adopted similarity of the alimentary tract in pigs and human beings there are eventually some important differences within the intestine, its digestive capacity or in reactions on ingested substances leading to unexpected results.  相似文献
