Achieving a balance between vegetative growth and spore production is essential for successful biocontrol by fungi. Low sporulation rates in the field can result in poor establishment and survival,whereas failure of conidia to recognise hosts can lead to persistence without efficacy. Commercial biocontrol products involve bulk preparations of conidia, however considerable variability in conidiation rates exists between biocontrol agents, which can restrict choice of strain for production. The majority of studies on Trichoderma conidiation have focused on the species T. viride and T. atroviride.These species form conidia in response to blue and near-UV light and/or nutrient deprivation and conidiation proceeds in a highly co-ordinated fashion, however relatively little is known on the genetic basis of Trichoderrma conidiation. In addition, whilst photoconidiation appears to be a general response detailed studies in other Trichoderma species are absent. In this study, conidiation in the lesser known biocontrol species T. hamatum is being investigated using a combined morphological and molecular approach. In contrast to T. atroviride, conidiation in response to blue-light was weaker and variable and suggested that additional triggers may be required for the T. hamatum photoresponse. A series of comparative photoconidiation assays are currently being undertaken investigating the effect of inoculum type and abiotic factors on timing and intensity of the response.Results will be discussed in relation to the current knowledge on conidial morphogenesis in Trichoderma. In addition to these morphological assays, a selection of genes implicated in sporulation and the blue-light responses are currently being isolated and characterised from T. hamatum. Two genes, phr1 and cmp1 , which were isolated previously from T. atroviride will be used as early and late markers of gene expression during the photoresponse in T. hamatum in order to define time points for harvesting comparable stage-specific RNA from T. hamatum and T. atroviride. Using degenerate PCR putative sporulation gene orthologues have also been identified in T. hamatum.Work is currently underway to isolate genomic clones of these genes from T. hamatum and T.atroviride. Sequence and expression analysis of orthologues, including expression in response to abiotic factors will be presented and discussed in relation to the current knowledge of the molecular basis of conidiation in Trichoderma and other filamentous fungi.……  相似文献

Discards are the portion of animal and plant material in the catch that is dumped back at sea. The Common Fisheries Policy plan proposed by the European Commission for 2014–2020 presents a controversial goal: to enforce the landing of fishing discards as a measure to encourage their reduction. This historical and political decision will shape the future of the fishing exploitation in European Seas. Discards generated by European fleets are not negligible, and its reduction is an ecological, socioeconomical and moral imperative. However, it must be achieved through the reduction in discards at source and the promotion of selective and non‐destructive gears. We argue it is doubtful that this discard ban will result in an effective reduction of discards. The proposed measure may, in fact, negatively affect ecosystems at all levels of biological hierarchy by disregarding the Ecosystem‐Based Approach to Fisheries and the Precautionary Principle. It could negatively impact several species by increasing fishing mortality, also commercial species if discards are not accounted in the total allowable catch. Communities preying on discards will likely be affected. The role discards currently play in the energy turnover of current ecosystems will be modified and should be fully evaluated. The landing of discards will likely generate new markets of fishmeal due to the growing demands for marine living resources. The ban will require substantial public investment to deal with technical problems on board and to control and enforce. Therefore, this measure should be only implemented after rigorous scientific and technical studies have been developed.  相似文献

Irish Sea fisheries have undergone considerable change in recent years following the decline of commercially important finfish stocks and their slow response to management's recovery plans. In 2015, the fishing industry called for a holistic exploration into the impact of environmental change and food web effects to identify the drivers underpinning stock dynamics. In this study, we identify correlations between large‐scale climatic indicators, temperature, primary and secondary productivity, and fish recruitment in the Irish Sea and incorporate them into an Ecopath with Ecosim food web model co‐created by scientists and fishers. Negative correlations were found between the North Atlantic Oscillation winter index (NAOw) and large zooplankton abundance and between the Atlantic Multidecadal Oscillation (AMO) and the recruitment of cod (Gadus morhua) and whiting (Merlangius merlangus). Using correlation analyses to direct the addition of environmental drivers to the Irish Sea ecosystem model improved the models fit against observed biomass and catch data and revealed the indirect impacts of environmental change as mitigated through trophic interactions. Model simulations suggest that historic environmental change suppressed the overall production of commercial finfish, limiting opportunities for the fishing industry, whilst also dampening the rate of stock recovery despite marked reductions in fishing effort. These results suggest that failure to account for ecosystem information may lead to misconceived expectations and flawed fisheries management; therefore, there is a need to operationalize ecosystem information through management procedures to support fisheries advice.  相似文献

Intact soil cores containing plants of Paspalum notatum or Digitaria decumbens were selected with the acetylene reduction method, and then exposed to 15N15 to confirm nitrogen fixation in tropical grass-bacteria associations. In a preliminary experiment with P. notatum15N2 incorporation was slow but progressive during 24 h in roots but translocation to rhizomes and leaves ceased after 17h. With improved assay chambers, enrichments of 0.151 and 0.563 15N atom % excess were obtained in roots of D. decumbens cv transvala and P. notatum systems respectively, after 3 days. Enrichments in rhizomes were similar to those of roots; however in the leaves only 8% of root enrichment was observed. The addition of sucrose to the soil doubled N2-fixation in roots in both grass species studied, but did not result in increased incorporation into the leaves of P. notatum.  相似文献

During the winter time in Finland, sunlight is inadequate for vitamin D synthesis. Many pet rabbits live as house rabbits with limited outdoor access even during summer and may therefore be dependent on dietary sources of vitamin D. The aims of this study were to report the serum 25‐hydroxyvitamin D concentrations in Finnish pet rabbits and to identify factors that influence vitamin D status. Serum 25‐hydroxyvitamin D concentrations from 140 pet rabbits were determined using a vitamin D enzyme immunoassay (EIA) kit. Eleven rabbits were excluded from the statistical analysis because of unclear dietary data. The remaining 129 rabbits were divided into groups depending on outdoor access during summer (no access n = 26, periodic n = 57, regular n = 46) as well as daily diet: little or no hay and commercial rabbit food ≤1/2 dl (n = 12); a lot of hay and no commercial food daily (n = 23); a lot of hay and commercial food <1 dl (n = 59); a lot of hay and commercial food ≥1 dl (n = 35). The range of serum 25‐hydroxyvitamin D concentration was from 4.5 to 67.5 ng/ml with a mean of 26.1 ng/ml. Statistical general linear model adjusted for weight, age and season indicated that diet was associated with vitamin D concentrations (p = 0.001), but outdoor access during summer was not (p = 0.41). Mean 25‐hydroxyvitamin D concentration was significantly higher in the rabbits receiving a lot of hay and commercial food ≥1 dl (33.9 ± 13.2 ng/ml) than in rabbits in other diet groups (24.0 ± 8.5 ng/ml, 21.7 ± 8.1 ng/ml, and 22.2 ± 18.0 ng/ml, respectively). This investigation showed wide variation in 25‐hydroxyvitamin D concentrations among Finnish pet rabbits. Diet remains a main source since outdoor access seems to be too limited to provide adequate vitamin D synthesis for most of them, and the use of vitamin D supplements is rare.  相似文献

This article describes the Finnish meat-inspection curriculum and presents an expert-panel evaluation of meat-inspection education. The work tasks of the meat-inspection veterinarian are challenging and include classical meat inspection, meat hygiene, hygiene control, and animal disease and welfare. The meat-inspection veterinarian is not only an inspector, which by itself is very demanding, but also an expert or "consultant" on food safety. The significant role of the meat-inspection veterinarian in society puts high demands on meat-inspection education, which should provide veterinary students with sufficient tools to perform meat inspection and hygiene control in slaughterhouses, cutting premises, and further processing plants. To be of high quality, such education must be evaluated from time to time. An expert panel evaluated Finnish undergraduate meat-inspection education and found that it provides veterinary students with good knowledge of meat inspection. The structure of the curriculum, with theoretical studies followed by four weeks of practice in a slaughterhouse, was considered vital for learning and for creating interest in meat inspection. The evaluation also revealed that certain subjects should receive greater emphasis and some new subjects should be introduced. Hygiene-control tasks, in particular, have increased and should receive more emphasis in education. Personnel management and interaction skills should be introduced into the curriculum as these skills influence all the duties of the meat-inspection veterinarian. This article outlines the subjects to be included in the modern, high-quality meat-inspection curriculum recommended by the expert panel.  相似文献

Finnish isolates of Beauveria bassiana (8 isolates), Metarhizium anisopliae (7), Tolypocladium sp. (2), Paecilomyces farinosus (2), P. fumosoroseus (1), Steinernema feltiae (3) and Bacillus thuringiensis ('Muscabac') were tested for efficacy against mixed populations of Delia radicum and D. floralis under field conditions in 1986–90. All pathogens were applied preventatively, the fungi as aqueous conidial or mycelial suspensions, dry conidia or dry mycelial powder. In only two of the nine experiments did B. bassiana or M. anisopliae give some control. In 1986, B. bassiana SF85–2 and Tolypocladium sp. SF85–4 (both at rate 1.2 x 109 conidia plant‐ 1), and ‘Muscabac’ (25 g 1?1, 1 dl plant?1) reduced the number of pupae by 80%, 60% and 50%, respectively, as compared with untreated and chemical (isophenphos) controls. In 1990, M. anisopliae SF86–39 at rate 1.6 x 109conidia plant?1 and 1.5 x 108 CFU plant?1 and S. feltiae SFS‐22 (35000 plant?1), increased the yield of cauliflower 2.2, 1.8, and 2.3‐fold, respectively, as compared with the untreated control, but these yields were only 19%, 15% and 19% of those of the chemical (diazinon) control. Paecilomyces isolates were ineffective in the 1986 experiment in which they were included. Our results suggest that it is difficult to find efficient control agents among the fungal and nematode species tested for use as biopesticides against cabbage root flies, but that the potential of M. anisopliae against these pests deserves further study.  相似文献
