In the mouse trigeminal pathway, sensory inputs from distinct facial structures, such as whiskers or lower jaw and lip, are topographically mapped onto the somatosensory cortex through relay stations in the thalamus and hindbrain. In the developing hindbrain, the mechanisms generating such maps remain elusive. We found that in the principal sensory nucleus, the whisker-related map is contributed by rhombomere 3-derived neurons, whereas the rhombomere 2-derived progeny supply the lower jaw and lip representation. Moreover, early Hoxa2 expression in neuroepithelium prevents the trigeminal nerve from ectopically projecting to the cerebellum, whereas late expression in the principal sensory nucleus promotes selective arborization of whisker-related afferents and topographic connectivity to the thalamus. Hoxa2 inactivation further results in the absence of whisker-related maps in the postnatal brain. Thus, Hoxa2- and rhombomere 3-dependent cues determine the whisker area map and are required for the assembly of the whisker-to-barrel somatosensory circuit.  相似文献

This multi-institutional study was designed to determine the clinical pharmacokinetics of fluconazole and outcomes in client-owned dogs (n = 37) and cats (n = 35) with fungal disease. Fluconazole serum concentrations were measured. Pharmacokinetic analysis was limited to animals at steady state (≥72 hr of treatment). The mean (range) body weight in 31 dogs was 25.6 (2.8–58.2) kg and in 31 cats was 3.9 (2.4–6.1) kg included in pharmacokinetic analyses. The dose, average steady-state serum concentrations (CSS), and oral clearance in dogs were 14.2 (4.5–21.3) mg/kg/d, 26.8 (3.8–61.5) µg/mL, and 0.63 ml min−1 kg−1, respectively, and in cats were 18.6 (8.2–40.0) mg/kg/d, 32.1 (1.9–103.5) µg/mL, and 0.61 ml min−1 kg−1, respectively. Random inter-animal pharmacokinetic variability was high in both species. Two dogs had near twofold increases in serum fluconazole when generic formulations were changed, suggesting lack of bioequivalence. Median CSS for dogs and cats achieving clinical remission was 19.4 and 35.8 µg/ml, respectively. Starting oral doses of 10 mg/kg q12h in dogs and 50–100 mg total daily dose in cats are recommended to achieve median CSS associated with clinical remission. Due to the large pharmacokinetic variability, individualized dose adjustments based on CSS (therapeutic drug monitoring) and treatment failure should be considered.  相似文献

Understanding the transmission of plant pathogen inoculum during the periods when the host plants are not present is crucial for predicting the initiation of epidemics and optimizing mitigation strategies. However, inoculum production at the end of the cropping season, survival during the intercrop period, and the emergence or release of inoculum can be highly variable, difficult to assess, and generally inferred indirectly from symptom data. As a result, a lack of large datasets hampers the study of these epidemiological processes. Here, inoculum production was studied in Leptosphaeria maculans, the cause of phoma stem canker of oilseed rape. The fungus survives on stubble left in the field, from which ascospores are released at the beginning of the next cropping season. An image processing framework was developed to estimate the density of fruiting bodies produced on stem pieces following incubation in field conditions, and a quality assessment of the processing chain was performed. A total of 2540 standardized RGB digital images of stems were then analysed, collected from 27 oilseed rape fields in Brittany over four cropping seasons. Manual post-processing removed 16% of the pictures, e.g. when moisture-induced darkening of the oilseed rape stems caused overestimation of the area covered with fruiting bodies. The potential level of inoculum increased with increasing phoma stem canker severity at harvest, and depended on the source field and the cropping season. This work shows how image-based phenotyping generates high-throughput disease data, opening up the prospect of substantially increased precision in epidemiological studies.  相似文献
