Abstract

In general, according to previous studies, pioneer species do not require arbuscular mycorrhizal fungi (AMF) to increase their growth and survival in tropical systems. The aim of this study was to determine the dependence response to AMF of Heliocarpus appendiculatus, a pioneer species, at different phosphorus (P) levels. In a greenhouse experiment, H. appendiculatus seedlings were grown in pots with a sterile vermiculite-sand mixture (1:1). Two sets of pots were set up: One set was inoculated (150 spores per pot) with indigenous AMF from a tropical rain forest at “Los Tuxtlas” (Veracruz, Mexico); the other set was not inoculated. To each set, 0, 0.02, 0.2, and 2 g L?1P was added. All pots were watered with 250 mL of nutrient solution. Mycorrhizal plants showed a higher total dry weight and relative growth rate in 0.02 g L?1P concentration, while nonmycorrhizal plants responded positively at 0.2 g L?1P; a decrease in plant responses at higher P levels was observed in both treatments. H. appendiculatus showed to have higher relative dependence at lower P concentration (≈50%). As levels of P increased, mycorrhizal colonization decreased. Successful growth of pioneer species during succession process may be improved if there is AMF content in soils, prior to disturbance.  相似文献

Current biogeographic models hypothesize that brown bears migrated from Asia to the New World ~100 to 50 thousand years ago but did not reach areas south of Beringia until ~13 to 12 thousand years ago, after the opening of a mid-continental ice-free corridor. We report a 26-thousand-year-old brown bear fossil from central Alberta, well south of Beringia. Mitochondrial DNA recovered from the specimen shows that it belongs to the same clade of bears inhabiting southern Canada and the northern United States today and that modern brown bears in this region are probably descended from populations that persisted south of the southern glacial margin during the Last Glacial Maximum.  相似文献

Background, Aims and Scope   Sulfate-reducing bacteria (SRB) are known for their capacity to reduce and precipitate heavy metals (HM) as metal sulfides, offering the opportunity to create an in situ reactive zone for the treatment of heavy metal-contaminated groundwater, a process called in situ metal precipitation (ISMP). The applicability of the ISMP technology first has to be investigated at a laboratory scale before going into an on site application. The evaluation and optimization of the ISMP process is facilitated when physical/chemical analysis techniques are combined with molecular tools that specifically monitor the abundance, diversity and dynamics of the indigenous sulfate reducing microbial community. In this study, batch experiments were conducted in order to investigate the feasibility of ISMP as a groundwater remediation strategy for an industrial site contaminated with elevated levels of Zn, Cd, Co and Ni. Methods   The potential of different types of carbon source/ electron donor (lactate, acetate, methanol, ethanol, Hydrogen Release Compound?, molasses) to stimulate the sulfate reduction and metal precipitation activity of the naturally present (or indigenous) SRB community was explored. In addition, the effect of amending vitamin B12 and yeast extract was evaluated. The ISMP process was monitored by combining analytical analyzes of process parameters (SO42-concentration, heavy metal concentrations, pH, Eh) with molecular tools such as SRB subgroup and genus specific PCR, denaturing gradient gel electrophoresis (DGGE), and phylogenetic analysis of clone sequences, based on either the 16S rRNA or the dsr (dissimilatory sulfite reductase) gene. Results and Discussion   The efficiency of different carbon-sources to stimulate the ISMP process followed the order HRC 〉 molasses 〉 methanol 〉 lactate 〉 ethanol 〉 acetate. Within 10 weeks, the highest sulfate and metal removal efficiencies ranged from 85% to 99%. Addition of yeast extract boosted the ISMP process, whereas vitamin B12 negligibly affected SRB activity. Analysis of the sulfate reducing population by SRB subgroup and genus specific PCR demonstrated that members of the genus Desulfosporosinus dominated in all batch tests, while 16S rDNA DGGE profiles additionally revealed the presence in the microbial communities of non-sulfate reducing bacteria within the family Clostridium and the -proteobacteria. The dsrB-based DGGE profiles allowed us to assess the diversity and dynamics of the sulfate reducing community and added to a better understanding of the effects of different batch conditions on the ISMP process. Remarkably, all dsrB sequences affiliated with the dsrB gene sequence cluster found in Desulfotomaculum, which received their xenologous dsrB gene from the -proteobacteria. Conclusions   The batch experiments, which aimed at stimulating the activities of the indigenous SRB communities, demonstrated that these communities were present and that their activities could be used to obtain efficient in situ precipitation of the contaminating heavy metals. This opens the possibility to test this concept in the future as an on site demonstration as part of the groundwater strategy for the heavy metal contaminated site. Although batch setups are suitable for preliminary feasibility studies for ISMP, they do not reflect the in situ situation where sulfate and heavy metal and metalloid polluted groundwater are supplied continuously. A sulfate reducing strain JG32A was isolated from whose 16S rRNA gene affiliated with the genus Desulfosporosinus, while its dsrB gene sequence clustered with Desulfotomaculum dsrB gene sequences, which received their xenologous dsr genes from -proteobacteria. Therefore we hypothesize that the batch experiments enrich members of the Desulfosporosinus genus that possess a non-orthologous dsrB gene. Recommendation and Perspective   The next step towards an on site pilot test for ISMP will be the setup of a series of column experiments, with process conditions that are selected based on the above mentioned results. This will allow to define optimal ISMP process conditions and to test its long-term efficacy and sustainability before going into an on site bioremediation application. By applying the described molecular tools together with physical-chemical analyzes, it can be investigated whether the same SRB community is enriched and which type of C-source is most effective in promoting and sustaining its growth and sulfate-reduction activity.  相似文献

The objective of the present experiment was to investigate the effects of transportation, lairage, and preslaughter stressor treatment on glycolytic potential and pork quality of the glycolytic longissimus and the oxidative supraspinatus (SSP) or serratus ventralis (SV) muscles. In a 2 x 2 x 2 factorial design, 384 pigs were assigned randomly either to short (50 min) and smooth or long (3 h) and rough transport, long (3 h) or short (< 45 min) lairage, and minimal or high preslaughter stress. Muscle samples were taken from the LM at 135 min and from the SSP at 160 min postmortem for determination of the glycolytic potential and rate of glycolysis. At 23 h postmortem, pork quality was assessed in the LM and the SV. Effects of transport and lairage conditions were similar in both muscle types. Long transport increased (P < 0.01) the glycolytic potential and muscle lactate concentrations compared with short transport. Both long transportation and short lairage decreased (P < 0.01) redness (a* values) and yellowness (b* values) of the LM and SV. In combination with short lairage, long transport decreased (P < 0.05) pork lightness (lower L* values), and electrical conductivity was increased (P < 0.05) after long transport. Several interactions between stress level and muscle type (P < 0.001) were observed. High preslaughter stress decreased (P < 0.001) muscle glycogen in both the LM and SSP, but this decrease was greater in the LM. Lactate concentrations were increased (P < 0.001) only in the LM by high preslaughter stress. Increases in ultimate pH (P < 0.001) and decreases in a* values (P < 0.01) were greatest in the SV, whereas increases in electrical conductivity (P < 0.001) were greatest in the LM. The lack of interactions among transportation, lairage, and muscle type was attributed to the relatively minor differences in stress among treatments. It was concluded that, in glycolytic muscle types such as the LM, the high physical and psychological stress levels associated with stress in the immediate preslaughter period have a greater effect on the water-holding capacity of the meat and may promote PSE development. Conversely, oxidative muscle types tend to have higher ultimate pH values and produce DFD pork in response to intense physical activity and/or high psychological stress levels preslaughter.  相似文献

Woody cuttings from two Salix genotypes (genotype I—clone LUC-31, Salix alba and genotype II- clone STOTT, Salix viminalis) were grown hydroponically for 14 days at increasing concentrations of Zn: control, 50, 100 or 150 μ M Zn. Genotype tolerance to excess zinc (Zn) was evaluated using a root elongation test. The changes in growth, Zn, iron (Fe), copper (Cu) and manganese (Mn) concentrations as well as photosynthetic performance were used as additional evaluation criteria. Photosynthetic pigments concentrations in Zn-exposed cuttings of genotype II decreased as compared with genotype I, which corresponded well with the higher leaf Zn accumulation, decreased Fe concentrations as well as lowered photosynthetic rate. Based on the indicators used, genotype I (Salix alba) was classified as more tolerant to excess Zn than genotype II (Salix viminalis).  相似文献

BACKGROUND: The cotton whitefly, Bemisia tabaci (Gennadius), is a cryptic species complex, and members of the complex have become serious pests in Pakistan because of their feeding and their ability to transmit cotton leaf curl virus (CLCuV). Here, an analysis was made of the identity of B. tabaci collected from cotton and a range of non‐cotton hosts in the cotton‐growing zones in Punjab and Sindh, the main cotton‐producing provinces of Pakistan, using a portion of the mitochondrial cytochrome oxidase 1 gene. The geographic distribution of the different members of the complex was then compared with the incidence of CLCuD. RESULTS: Using the Dinsdale nomenclature, the results revealed three putative species, Asia 1, Asia II 1 and Middle East‐Asia Minor 1. Asia II 1 (also referred to in the literature as biotypes K, P, PCG‐1, PK1, SY and ZHJ2) was only recorded from Punjab cotton plants, whereas Asia 1 (also referred to in the literature as biotypes H, M, NA and PCG‐2) was found in both Sindh and Punjab. Middle East‐Asia Minor 1 (commonly known as biotype B and B2) was found only in Sindh. Moreover, Asia II 1 was associated with high incidences of CLCuD, whereas regions where Middle East‐Asia Minor 1 was present had a lower incidence. Phylogenetic analysis showed that the Middle East‐Asia Minor 1 population in Sindh formed a distinct genetic subgroup within the putative species, suggesting that the Sindh province of Pakistan may form part of its home range. So far, no individuals from the putative species Mediterranean (commonly known as biotypes Q, J and L) have been found in Pakistan. CONCLUSIONS: The capacity to manage pests and disease effectively relies on knowledge of the identity of the agents causing the damage. In the case of CLCuD in Pakistan, this knowledge has been obscured to some extent because of the inconsistent approach to identifying and distinguishing the different B. tabaci associated with CLCuD. The situation has now been clarified, and a strong association between disease incidence and vector identity and abundance has been shown. Given this advance, future research can now focus on factors that influence the capacity of different vector species to transmit the viruses that cause CLCuD, the reason for differences in vector abundance and the lack of geographic overlap between the cryptic vector species. The objectives of the present experiment were 1) to study the effects of transport conditions and lairage duration on stress level, muscle glycolytic potential, and pork quality; and 2) to investigate whether the negative effects of high stress immediately preslaughter are affected by preceding handling factors (transport and lairage). In a 2 x 2 x 2 factorial design, halothane-free pigs (n = 384) were assigned to either short (50 min) and smooth or long (3 h) and rough transport; long (3 h) or short (<45 min) lairage; and minimal or high preslaughter stress. Pigs were processed in eight groups (48 pigs per group) on various days at a commercial plant. Blood samples were taken at exsanguination to measure plasma cortisol and lactate concentrations. Muscle pH and temperature were measured at 30 and 40 min, respectively, and both were measured at 3 h, postmortem. A LM sample was taken 135 min postmortem to estimate glycogen content and rate of glycolysis. Pork quality attributes were assessed 23 h postmortem. Short transport increased cortisol when followed by short lairage (transport x lairage; P < 0.01). Long transport, but not lairage (P > 0.30), tended to increase (P = 0.06) muscle glycolytic potential. Long transport tended to increase (P = 0.08) electrical conductivity, and decreased a* (P < 0.01) and b* (P < 0.02) values. Decreasing lairage from 3 h to <45 min decreased (P < 0.05) the L* value, but it did not (P > 0.10) affect other pork quality traits. High stress decreased (P < 0.001) muscle glycolytic potential, and increased (P < 0.001) plasma lactate, cortisol, muscle temperature, rate of pH decline, and ultimate pH. Except for decreased (P < 0.001) b* values, pork color was not (P > 0.40) affected by high stress, but water-holding properties (measured by electrical conductivity, filter paper moisture, and drip loss) were impaired (P < 0.001) by high stress. Fiber optic-measured light scattering and Warner-Bratzler shear force were not (P > 0.12) affected by any treatment. Comparisons with the "optimal" handling (short transport, long lairage, and minimal stress) revealed that, with regard to water-holding properties, the negative effects of high stress were aggravated by suboptimal transport and lairage conditions. High stress alone increased electrical conductivity by 56%, whereas high stress in combination with short lairage led to an 88% increase. However, high preslaughter stress contributed most and was the major factor responsible for reductions in pork quality.  相似文献

The objective of the present experiment was to study physiological changes elicited in response to stress in the immediate preslaughter period and to link them to pork quality characteristics. Crossbred, halothane-free pigs (n = 192) were processed in eight groups (24 pigs per group) on various days at one of two commercial processing plants operating different stunning systems (electrical and CO2 stunning in Plants A and B, respectively). In each group, half the pigs were exposed to either minimal or high preslaughter stress. Blood samples were taken at exsanguination, and lactate, cortisol, and catecholamines, as well as blood pH and temperature, were assessed and linked to various longissimus muscle quality attributes. Additionally, muscle pH and temperature were measured 30 min postmortem, and muscle glycolytic potential was determined 22 h postmortem. At both processing plants, high preslaughter stress resulted in higher (P < 0.05) blood cortisol and lactate; however, the effects of preslaughter stress on catecholamines and blood pH were believed to be biased by the different stunning methods employed at the plants. High preslaughter stress increased (P < 0.05) blood temperature at Plant A but not at Plant B. At both plants, high stress increased (P < 0.05) 30-min muscle temperature and decreased (P < 0.05) 30-min muscle pH. Ultimate pH was increased (P < 0.05) and muscle glycolytic potential was decreased (P < 0.05) by high preslaughter stress. At both plants, high stress resulted in inferior pork quality attributes (P < 0.05), including reflectance, electrical conductivity, filter paper moisture, drip loss, and L* value. The effect of stress was greater on water-holding capacity than on pork color, with drip losses increased by 56%. Of all stress indicators measured at exsanguination, only blood lactate was strongly correlated with pork quality attributes. Regression analyses revealed that blood lactate and glycolytic potential accounted for 52 and 48% of the variation in drip loss and L* value, respectively. In combination with high preslaughter stress, high glycolytic potentials were related to increased drip losses. We conclude that high preslaughter stress leads to impaired pork quality, with high muscle energy levels aggravating the negative effects of preslaughter stress. Monitoring stress level by blood lactate measurement in combination with strategies to control muscle energy present at slaughter may help to improve meat quality.  相似文献

Economic weights are obtained for feed intake using a growth model and an economic model. The underlying concept of the growth model is the linear plateau model. Parameters of this model are the marginal ratio (MR) of extra fat and extra protein deposition with increasing feed intake (FI) and the maximum protein deposition (Pd(max)). The optimum feed intake (FI0) is defined as the minimum feed intake that meets energy requirements for Pd(max). The effect of varying FI and MR on performance traits was determined. An increase in FI results in a larger increase in growth rate with lower MR. For a given MR, feed conversion ratio is lowest when FI equals FI0. Lean meat percentage (LMP) is largest for a low MR in combination with a low FI. The decrease in LMP with higher FI islargest when FI exceeds FI0. Economic weights for FI, MR and Pd(max) depend on FI in relation to FI0. Economic weights for FI are positive when FI is less than FI0 and negative when FI is larger than FI0. The MR has only then a negative economic weight, when FI is below FI0. Economic weights of FI and MR have a larger magnitude with lower MR and lower Fl. In contrast, economic weights for growth rate and FI derived from the economic model only change in magnitude and not in sign with different levels of these traits. The economic model always puts a negative economic weight on FI since it expresses profit due to a decrease in FI with constant growth rate and LMP. This holds the risk of continuous decrease in FI in pig breeding programs. In contrast, the use of growth models for genetic improvement allows direct selection for an optimum feed intake which maximizes feed efficiency in combination with maximum lean meat growth. It is concluded that recording procedures have to be adapted to collect the data necessary to implement growth models in practical pig breeding applications.  相似文献

The stress by air pollution at the systematicPan-European 16 × 16 km2 forest (crown) condition monitoring network, is discussed by comparingsite-specific estimates of critical and presentconcentration and deposition levels for S and Ncompounds and ozone. Results indicate that theexceedance of critical levels, related to directabove-ground impacts, decrease going from O3 >SO2 > N compounds. Critical N loads related toeffects on the forest understorey are exceeded atapproximately 25% of the plots, located mainly inWestern and Central Europe. Critical N loads relatedto effects on trees are hardly ever exceeded, but mostlikely, this is an under estimate. Critical aciddeposition levels are exceeded at approximately 30%of the plots with a low base saturation, where acidinputs may release toxic Al. This is especially thecase in Central and Eastern Europe, where presentloads are high and in boreal forest in SouthernScandinavia where critical loads are low. Although theuncertainties in the calculated exceedances is large,the spatial pattern, which is most important for acorrelative study, seems reliable, implying that thecritical load concept is suitable for regional risk assessments.  相似文献
