Flow cytometric sorting technology was used to measure the difference in DNA content between X- and Y-chromosome-bearing spermatozoa in bucks. Spermatozoa were analysed by flow cytometry to characterize X- and Y-chromosome-bearing sperm populations and to quantify the DNA difference between them. Two symmetrical, overlapping and clearly separated peaks, corresponding to X- and Y-bearing spermatozoa, were detected. The difference in fluorescence intensity between the peaks was 4.4 +/- 0.03% without any significant inter- or intra-animal variations. Therefore, the identification and selection of high-purity samples of sperm populations for sex sorting is easier in bucks compared with other domestic species.  相似文献

The purpose of this study was to investigate the effect of seminal plasma (SP) from bulls of known fertility on bovine endometrial epithelial cells (bEEC) in culture. The bEEC from passage 5, approximately 5.0–13 × 105 cells per flask, were challenged with SP from bulls of high or low fertility (n = 3 and 2, respectively) or PBS (control), at 1% (75 μl) or 4% (300 μl) and were incubated for 72 hr (n = 13 per challenge). Total cell number and viability of bEEC after challenge with 1% SP from either high‐ or low‐fertility bulls (75H or 75L, respectively) did not differ from controls. In contrast, challenge with 4% of SP from high‐ or low‐fertility bulls (300H or 300L) negatively affected bEEC cell number and viability. Challenge with 300 L had a greater adverse effect than 300H. These results suggest that the negative effect of bovine SP on bEEC is both dose‐dependent and fertility‐dependent.  相似文献

Numerous culture-based diagnostics are available on the Australian and international markets for on-farm detection of bacterial pathogens in milk. Use of such diagnostics may provide an opportunity to improve the prudent use of antimicrobials in udder health management. Farms are low-resource settings in terms of diagnostic microbiology capacity. The World Health Organisation has identified criteria for the evaluation of diagnostic tests in low resource settings based on Accuracy, Sensitivity, Specificity, User-friendliness, being Rapid or Robust, Equipment-free and being Deliverable (ASSURED). Here, we review how those criteria can be interpreted in the context of microbiological diagnosis of mastitis pathogens, and how on-farm diagnostics that are currently available in Australia perform relative to ASSURED criteria. This evaluation identifies multiple trade-offs, both with regard to scientific criteria and with regards to convenience criteria. More importantly, the purpose of testing may differ between farms, and test performance should be evaluated relative to its intended use. The ability of on-farm mastitis diagnostics to inform mastitis treatment decision-making in a timely and cost-effective manner depends not just on test characteristics but also on farm-specific pathogen prevalence, and on the farm enterprise's priorities and the farm manager's potential courses of action. With most assay evaluations to date conducted in professional laboratories, there is a surprising dearth of information on how well any of the diagnostic tests perform on-farm and, indeed, of the on-farm decision-making processes that they aim to inform.  相似文献

Production and water use in lettuces under variable water supply   总被引:3,自引:0,他引:3  
The effects of a variable water supply on the water use, growth and yield of two crisphead and one romaine (i.e., Cos) lettuce cultivar were examined in a field experiment using a line source sprinkler system that produced a range of water regimes that occur in growers fields. Four locations at increasing distances from the main line were monitored through the season (i.e., from thinning to harvest, 28–63 days after planting (DAP)). These locations at the end of the season corresponded to: (1) rewatering to field capacity (FC); (2) watering with a volume 13% below that required in the field capacity treatment (0.87*FC); (3) 30% below FC (0.70*FC); and (4) 55% below FC (0.45*FC). A linear production function for dry matter accumulation and fresh weight vs. crop evapotranspiration (ETc) was determined for lettuce during this period, giving a water use efficiency for dry matter of 1.86 g m–2 mm–1 and for fresh weight of 48 g m–2 mm–1 . For lettuce irrigated to field capacity, ETc between thinning and harvest was 146 mm; maximum crop coefficients of 0.81–1.02 were obtained at maturity (55–63 DAP). For the three irrigation treatments receiving the largest water application, ETc was higher in the Cos culivar than in the two crisphead lettuce cultivars which had similar ETc. Plant fresh weight was more sensitive than dry weight to reduction in water supply. In the FC treatment, root length density and soil water extraction were greatest in the top 0–45 cm, and decreased rapidly below 45 cm depth. Soil water extraction by roots increased at lower depths when irrigation was reduced. Instantaneous rates of leaf photosynthesis and leaf water potential showed no response to the irrigation treatments in this study, despite differences in biomass production. Evaporation was determined to be the major component of ETc for 45 of the 63 days of the growing season. The large loss of water by evaporation during mid-season and the apparent insensitivity of lettuce to the volume of irrigation during this period may provide an opportunity for reducing irrigation applications.  相似文献

A method of marking abalone (Haliotis asinina Linne) for sea ranching and stock enhancement purposes was developed. Three‐month‐old abalone juveniles (11.8‐mm shell length, 0.28 g) were fed artificial diets for 1, 2, or 3 weeks. The width of the bluish‐green shell band produced by abalone juveniles was 1.7, 2.6, and 4.2 mm after 1, 2, or 3 weeks of feeding respectively. The growth and survival of juveniles fed artificial diets did not differ from that of juveniles fed the seaweed Gracilariopsis bailinae (control). Feeding the diet‐fed juveniles with the seaweed thereafter produced the natural brownish shell, thus forming a sandwiched bluish‐green band. An experimental release in outdoor tanks with natural growth of seaweeds and diatoms, and in a marine reserve showed that the shell band remained clear and distinct, indicating the usefulness of this shell marking method in sea ranching and stock enhancement of abalone.  相似文献

Free-drainage or “open” substrate system used for vegetable production in greenhouses is associated with appreciable NO3 leaching losses and drainage volumes. Simulation models of crop N uptake, N leaching, water use and drainage of crops in these systems will be useful for crop and water resource management, and environmental assessment. This work (i) modified the TOMGRO model to simulate N uptake for tomato grown in greenhouses in SE Spain, (ii) modified the PrHo model to simulate transpiration of tomato grown in substrate and (iii) developed an aggregated model combining TOMGRO and PrHo to calculate N uptake concentrations and drainage NO3 concentration. The component models simulate NO3-N leached by subtracting simulated N uptake from measured applied N, and drainage by subtracting simulated transpiration from measured irrigation. Three tomato crops grown sequentially in free-draining rock wool in a plastic greenhouse were used for calibration and validation. Measured daily transpiration was determined by the water balance method from daily measurements of irrigation and drainage. Measured N uptake was determined by N balance, using data of volumes and of concentrations of NO3 and NH4+ in applied nutrient solution and drainage. Accuracy of the two modified component models and aggregated model was assessed by comparing simulated to measured values using linear regression analysis, comparison of slope and intercept values of regression equations, and root mean squared error (RMSE) values. For the three crops, the modified TOMGRO provided accurate simulations of cumulative crop N uptake, (RMSE = 6.4, 1.9 and 2.6% of total N uptake) and NO3-N leached (RMSE = 11.0, 10.3, and 6.1% of total NO3-N leached). The modified PrHo provided accurate simulation of cumulative transpiration (RMSE = 4.3, 1.7 and 2.4% of total transpiration) and cumulative drainage (RMSE = 13.8, 6.9, 7.4% of total drainage). For the four cumulative parameters, slopes and intercepts of the linear regressions were mostly not statistically significant (P < 0.05) from one and zero, respectively, and coefficient of determination (r2) values were 0.96-0.98. Simulated values of total drainage volumes for the three crops were +21, +1 and −13% of measured total drainage volumes. The aggregated TOMGRO-PrHo model generally provided accurate simulation of crop N uptake concentration after 30-40 days of transplanting, with an average RMSE of approximately 2 mmol L−1. Simulated values of average NO3 concentration in drainage, obtained with the aggregated model, were −7, +18 and +31% of measured values.  相似文献
