To evaluate the impact of temporal variation of primary productivity on the recruitment of Japanese sardine (Sardinops melanostictus) in the Sea of Japan, the phenology of sea surface phytoplankton abundance was estimated from 8 day multiple satellite (SeaWiFS, MODIS‐Aqua, MERIS, and VIIRS) derived sea surface chlorophyll (SSChl) a concentrations from January 1998 to December 2015. Because relationships between SSChl a and in situ chlorophyll a concentrations were significantly different among periods based on the satellite combinations used, maximum and minimum SSChl a concentrations of 1 year were relativized as 1 and 0, respectively. Spatio‐temporal variation of relativized SSChl a concentrations was determined by using empirical orthogonal function (EOF) analysis. Scores in the first EOF mode denoted the basin‐scale variations of SSChl a concentrations in the Sea of Japan, and the major peak from the end of February to the end of May displayed the spring bloom. The logarithm of recruitment per spawner (LNRPS) for sardine was positively affected by delays in the start and end dates of the spring phytoplankton bloom. The delay of the date of the lowest sea surface temperature contributed to the delay of the end of the spring bloom during the period 1998–2015 and elevated the LNRPS during the period 1982–2015. Sardine spawns in the southern Sea of Japan from April to May, hence, delays of the spring bloom prolonged its overlap with sardine larval periods, and thus improved the recruitment of Japanese sardine in the Sea of Japan. 相似文献
Surface properties of fibrous and ground cotton and linen were investigated by inverse gas chromatography (IGC) and the contact angle with different liquids was also measured on fabrics composed of both fibers. Results proved that dispersion component of surface tension (γsd) determined by IGC depends not only on the surface energy, but also on several factors influencing the adsorbability of probe molecules on the cellulosic substrates. For cotton samples, the trapping of n-alkanes among waxy molecules in the outer layer of fibers can be presumed. This effect results in larger γsd for cotton fibers than for linen in spite of the higher wettability of the linen fabrics. Besides the surface energy and trapping effects, the grinding also influences the γsd values. Specific enthalpy of adsorption (ΔHAab) of polar probes could be determined on all linen samples, but only on the ground cotton sample. Lewis acid-base character calculated for linen and ground cotton samples depends on the same effects as the γsd does. The similar ΔHAab values of chloroform (acidic) and THF (basic) measured on each of the samples support the conclusion that the surface character is amphoteric, which is also proved by the high ΔHAab values of the amphoteric ethyl acetate and acetone probes. 相似文献
Using RT-PCR and semi-quantitative PCR, mRNA expression for canine interferon (IFN)-gamma, interleukin (IL)-2, IL-4, IL-5, IL-10, tumor necrosis factor (TNF)-alpha and transforming growth factor (TGF)-beta in peripheral blood mononuclear cells (PBMCs) was examined in dogs with or without demodicosis. mRNA expression for IFN-gamma as well as TNF-alpha in dogs with demodicosis (localized (LD) and generalized (GD)) was slightly lower than those in dogs without demodicosis (healthy controls). Expression of IL-5 mRNA in dogs with demodicosis was higher than that in control dogs, but there were no significant differences in IL-4 and IL-10 mRNA expression levels among the three groups. On the other hand, expression levels of TGF-beta mRNA in dogs with GD were higher than those in control dogs and dogs with LD. The expression levels of IL-5 and TGF-beta mRNA decreased in all three dogs with GD which showed resolution of the clinical signs. Taken together, these results suggest that the Th2-like response in PBMCs from dogs with demodicosis is up-regulated, and that subsequent increased expression of IL-5 and TGF-beta mRNA in dogs with GD is reversible after treatment. Therefore, these cytokines, particularly IL-5, might be a useful clinical index of the clinical course in demodicosis. Also, increased TGF-beta mRNA expression might be a key factor for revealing the difference in the mechanism of onset between LD and GD. 相似文献
MicroRNAs (miRNAs) are a class of small non-coding RNAs with a critical role in development and environmental responses. Efficient
and reliable detection of miRNAs is an essential step towards understanding their roles in specific cells and tissues. However,
gel-based assays currently used to detect miRNAs are very limited in terms of throughput, sensitivity and specificity. Here
we provide protocols for detection and quantification of miRNAs by RT-PCR. We describe an end-point and real-time looped RT-PCR
procedure and demonstrate detection of miRNAs from as little as 20 pg of plant tissue total RNA and from total RNA isolated
from as little as 0.1 μl of phloem sap. In addition, we have developed an alternative real-time PCR assay that can further
improve specificity when detecting low abundant miRNAs. Using this assay, we have demonstrated that miRNAs are differentially
expressed in the phloem sap and the surrounding vascular tissue. This method enables fast, sensitive and specific miRNA expression
profiling and is suitable for facilitation of high-throughput detection and quantification of miRNA expression. 相似文献

The ionic composition of culture water may be a more important limiting factor than the salinity itself and may lead to osmotic stress which may influence growth and survival of shrimp culture. The uptake rate and the effect of magnesium chloride (MgCl2), calcium chloride (CaCl2), and potassium chloride (KCl) salts in juveniles of whiteleg shrimp (Litopenaeus vannamei) cultured with biofloc technology (BFT) was evaluated for 62 days in seawater (30 practical salinity unity). Five treatments were analyzed in triplicate: T1; control (water exchange rate of 5% daily), T2; adding CaCl2?+?MgCl2?+?KCl, T3; adding MgCl2?+?KCl, T4; adding CaCl2?+?KCl, and T5; adding CaCl2?+?MgCl2. Mineral salts were added to water and the response of experimental parameters: physicochemical variables of water quality, osmotic pressure, total hemocyte count, glutathione peroxidase gene expression, superoxide dismutase, and zootechnical variables of the shrimp was assessed. The uptake of single chloride salts (CaCl2, MgCl2, and KCl) by shrimp varied as a function of the concentrations and the ratio of these three ions. Magnesium deficiencies in the culture medium increased CaCl2 and KCl uptake without showing gene expression of SOD and GPx. The best survival rate was obtained by adding the three ions (T2) and the control (T1, water exchange of 5%). We concluded that L. vannamei can be maintained by the addition of essential chloride ions in BFT without water replacement.
