Abstract. Recent developments in in situγ ray spectrometry offer a new approach to measuring the activity of radionuclides such as 137Cs and 40K in soils, and thus estimating erosion or deposition rates and field moist bulk density (ρm). Such estimates would be rapid and involve minimal site disturbance, especially important where archaeological remains are present. This paper presents the results of a pilot investigation of an eroded field in Scotland in which a portable hyper pure germanium (HPGe) detector was used to measure γ ray spectra in situ. The gamma (γ) photon flux observed at the soil surface is a function of the 137Cs inventory, its depth distribution characteristics and ρm. A coefficient, QCs, derived from the forward scattering of 137Cs γ ray photons within the soil profile relative to the 137Cs full energy peak (662 keV), was used to correct the in situ calibration for changes in the 137Cs vertical distribution in the ploughed field, a function of tillage, soil accumulation and ρm. Based on only 8 measurements, the agreement between in situγ ray spectrometry and soil sample measurements of 137Cs inventories improved from a non significant r2=0.05 to a significant r2=0.62 (P<0.05). Erosion and deposition rates calculated from the corrected in situ137Cs measurements had a similarly good agreement with those calculated from soil cores. Mean soil bulk density was also calculated using a separate coefficient, QK, derived from the forward scattering γ photons from 40K within the soil relative to the 40K full energy peak (1460 keV). Again there was good agreement with soil core measurements (r2=0.64; P<0.05). The precision of the in situ137Cs measurement was limited by the precision with which QCs can be estimated, a function of the low 137Cs deposition levels associated with the weapons testing fallout and relatively low detector efficiency (35%). In contrast, the precision of the in situ ρm determination was only limited by the spatial variability associated with soil sampling.  相似文献

1. In a series of experiments with laying hens, attempts were made to eliminate the anti‐nutritive factor which reduces egg production when feeding field beans (Vicia faba L.) as a source of supplementary protein in cereal‐based diets.

2. Neither pelleting diets nor heat processing the beans by autoclaving proved beneficial. A continuous‐flow process for infra‐red heating of the seeds at 150 °C for 70 s followed immediately by flaking (micronisation) gave inconsistent results.

3. Beans having a white skin with associated low tannin content, did not support significantly higher egg production than did a normal brown‐skinned bean, thus indicating that tannin was not the main anti‐nutritive factor.

4. A cotyledon fraction, obtained by mechanical removal of the tannin‐containing skin from brown beans, did not support higher egg production than did the whole bean, and the corresponding brown‐skin fraction did not reduce egg production when substituted for cereal in the control diet.   • 1. Shallow lakes excavated for ornamental purposes during the 18th and 19th centuries are abundant in lowland Europe. However, relative to older man‐made and/or natural lakes, these lakes may have been undervalued from the perspective of nature conservation.
  • 2. To evaluate this idea a comparison was made between the aquatic macrophyte communities (submerged and floating‐leaved vegetation) of 66 shallow, English lakes including 34 ornamental lakes and 32 flooded medieval peat workings (the Norfolk and Suffolk Broads system), the latter being widely protected by conservation legislation.
  • 3. Some 47%, 38% and 15% of the lakes studied were phytoplankton‐dominated, macrophyte‐dominated or deemed too shallow (<50 cm water depth), respectively, to support a macrophyte vegetation. A higher proportion of the ornamental lakes were macrophyte‐dominated (51%) by comparison with the broads (34%). In addition, many of the ornamental lakes contained diverse plant communities including abundant populations of Characeae, a common feature of lakes in the region before the major onset of eutrophication.

From the perspectives of macrophyte species richness, charophyte communities and indeed ‘reference condition macrophyte assemblages’, many of the studied ornamental lakes can be considered to be of high conservation value. Yet, in contrast to the broads, the vast majority of ornamental lakes have little conservation protection and are rarely subject to biological monitoring and/or surveying, thus leaving them vulnerable to eutrophication and inappropriate management. This study suggests that ornamental lakes are worthy of much greater attention from conservation organizations.

The study describes three polymerase chain reaction (PCR) systems for the CVI988 vaccine virus: the meq gene, the MDV BamHI-D/H 132 bp tandem repeat fragment and the MDV-gB gene. Whereas the PCR product of virulent MDV strains and of the CVI988 virus strain with the meq and the 132 bp primer sets differed for the two templates, the MDV-gB PCR products were similar. The sensitivity of the three PCRs was determined for the two templates: the CVI988 DNA was detected up to 2.48 plaque forming units, and a MDV-1 DNA, was amplified with the 132 bp primers up to the 10(-3) DNA dilution, and up to the 10(-2) with the MDV-gB and meq gene primers. As conventional detection for the CVI988 vaccine virus is by tissue culture, the aim was to analyse the feasibility of the molecular detection of the vaccine virus in the vaccinated chick. In two experimental trials employing specific pathogen free and commercial Lohmann chicks, respectively, the vaccine virus replicated to a limited extent; it was detected only in the spleen of up to 60% chicks at 2-4 weeks and in one chick at 3 weeks, respectively. The survey of three commercial Lohmann flocks, kept in biosecurity conditions, revealed the vaccine virus only in the spleen of 40% of 30-day-old chicks. The present study shows that CV1988 DNA is present in vaccinated chicks in a low quantity and it is difficult to detect directly from the chick, probably because vaccine viruses are latent in vivo. For an efficient detection it is pertinent to cultivate the vaccine virus on chicken embryo fibroblasts (CEF), as then the virus escapes the latent state, enters into the productive mode of replication, and a high viral copy number is produced.  相似文献

The aim of this study was to evaluate the presence of Helicobacter (H.) spp. in swine affected by gastric ulceration. Stomachs from 400 regularly slaughtered swine were subjected to gross pathological examination to evaluate the presence of gastric ulcers. Sixty-five samples collected from ulcerated pars esophagea and 15 samples from non-ulcerated pyloric portions were submitted to histopathological and molecular analyses, to detect Helicobacter spp., H. suis and H. pylori by PCR. Feces and saliva swabs were also collected from 25 animals in order to detect in vivo the presence of Helicobacter spp.. Gastric ulcers were detected in 373 cases (93%). The presence of ulcers in association with inflammatory processes was further confirmed by histological examination. Forty-nine percent (32/65) of the ulcerated esophageal portions as well as 53% (8/15) of the non-ulcerated pyloric portions were positive for Helicobacter spp. by PCR. The Helicobacter spp. positive samples were also positive for H. suis, while H. pylori was not detected. These results were confirmed by restriction enzyme analysis. With regard to feces and saliva samples, 15/25 (60%) and 16/25 (64%) were positive for Helicobacter spp. PCR, respectively but all were negative in H. suis and H. pylori specific PCR.  相似文献
