Phagocytosis is an important function of both invertebrate and vertebrate blood cells. In this study, the phagocytic activity of haemocyte subpopulations of penaeid shrimp, Litopenaeus vannamei, (Boone), against pathogenic and non‐pathogenic particles was investigated in vitro. The haemocytes of penaeid shrimp were firstly separated by centrifugation on a continuous density gradient of iodixanol into four fractions with five subpopulations (sub), of which sub 1 (hyalinocytes) and sub 4 (semi‐granulocytes) have the main function in phagocytosis of both pathogenic and non‐pathogenic bacteria as well as fluorescent polystyrene beads. It was found that these haemocyte subpopulations engulfed virulent Vibrio campbellii and Vibrio harveyi at a higher rate than non‐virulent Escherichia coli and polystyrene beads. When these bacteria were mixed with shrimp haemocyte subpopulations and incubated for 180 min, the percentage of viable intracellular V. campbellii (25.5 ± 6.0%) recovered was significantly higher than the percentage recovered from V. harveyi (13.5 ± 1.1%). No viable intracellular E. coli was observed in this study. In contrast to V. harveyi and E. coli, V. campbellii containing endosomes did not acidify in time. Incubation of haemocyte subpopulations with the most virulent V. campbellii strain resulted in a significant drop in haemocyte viability (41.4 ± 6.3% in sub 1 and 30.2 ± 15.1% in sub 4) after 180 min post‐inoculation in comparison with the less virulent V. harveyi (84.1 ± 5.6% in sub 1 and 83.4 ± 4.1% in sub 4) and non‐virulent E. coli (92.7 ± 2.8% in sub 1 and 92.3 ± 5.6% in sub 4) and polystyrene beads (91.9 ± 1.6% in sub 1 and 84.4 ± 3.4% in sub 4). The South American tomato pinworm, Tuta absoluta (Meyrick, 1917) (Lepidoptera: Gelechiidae), is heavily targeted by insecticide applications. Methoxyfenozide is highly effective against T. absoluta with reduced side effects to natural enemies. This work aimed to (1) genetically and biochemically characterize resistance of the T. absoluta GBN population to methoxyfenozide, (2) establish cross resistance profiles with other insecticide groups and (3) monitor resistance in populations with the goal of improving T. absoluta insecticide resistance management (IRM). Methoxyfenozide resistance was completely recessive, polyfactorial and autosomal. Effective dominance revealed that 10 mg methoxyfenozide/L would be enough to eliminate susceptible homozygotes and heterozygotes, thus used to diagnose resistance in field populations. The synergism of methoxyfenozide toxicity in the resistant population for PBO (SR?=?95×), DEF (SR?=?51×) and DEM (SR?=?45×), suggested monooxygenases, esterases and glutathione S-transferases as resistance mechanisms. However, only monooxygenase activity appeared to be involved in methoxyfenozide resistance. Resistance ratio for methoxyfenozide (2352-fold) after selection and cross-resistance ratios of a lab-selected GBN strain (“GBN-Sel”) were significant relative to a susceptible strain “JDR1-Sus” for tebufenozide (656-fold), cartap hydrochloride (10.68-fold), deltamethrin (4.70-fold), abamectin (2.65-fold), lufenuron (2.22-fold) and indoxacarb (1.92-fold), with negative cross-resistance to spinetoram (0.32-fold). Evidence of control failures was observed in 10 field populations of T. absoluta (mortalities between 13 and 76%), and all populations showed frequencies of resistant phenotypes (percentage survivorship ranging between 4 and 96%). A rational basis for managing resistance to bisacylhydrazines is discussed, along with details of recommended T. absoluta resistance management tactics.

implications to both breeding strategies and orchard management for fruit production. The purpose of this study was to identify
and to characterize the S-RNase alleles of the gametophytic incompatibility among apple cultivars developed in Brazil, seeking
to give support for choosing right combinations of parent in the apple breeding programs. It also sought to identify correct
combinations of scion/pollinator cultivars of commercial apple orchards. A total of 16 specific S-RNase alleles primers were
tested against DNA extracted from 12 Brazilian cultivars and their parents. The molecular analysis confronted to the reference
cultivars, showed that the cultivars Daiane, Imperatriz and Princesa have the same incompatibility S3 and S5 alleles, while
‘Lisgala’ showed the alleles S2 and S5; ‘Suprema’, S1 and S9; ‘Catarina’, S1 and S19; ‘Joaquina’ and ‘Fred Hough’, S5 and
S19; ‘Baronesa’, S3 and S9; ‘Duquesa’, S2 and S3. For ‘Primícia’ and ‘Condessa’ it was only possible to identify one of the
S-alleles, namely S24 and S2, respectively, with the second allele remaining to be identified. Progeny test indicated the
Mendelian inheritance for RNase alleles. Results of this study will be helpful to judiciously choose parents in apple breeding

Most materials shrink laterally like a rubber band when stretched, so their Poisson's ratios are positive. Likewise, most materials contract in all directions when hydrostatically compressed and decrease density when stretched, so they have positive linear compressibilities. We found that the in-plane Poisson's ratio of carbon nanotube sheets (buckypaper) can be tuned from positive to negative by mixing single-walled and multiwalled nanotubes. Density-normalized sheet toughness, strength, and modulus were substantially increased by this mixing. A simple model predicts the sign and magnitude of Poisson's ratio for buckypaper from the relative ease of nanofiber bending and stretch, and explains why the Poisson's ratios of ordinary writing paper are positive and much larger. Theory also explains why the negative in-plane Poisson's ratio is associated with a large positive Poisson's ratio for the sheet thickness, and predicts that hydrostatic compression can produce biaxial sheet expansion. This tunability of Poisson's ratio can be exploited in the design of sheet-derived composites, artificial muscles, gaskets, and chemical and mechanical sensors. 相似文献
Abstract CASE HISTORY: A 15-mm diameter mass developed in the S/C tissue overlying the right lateral gluteal muscle of a 6½-year-old female Flat-coated Retriever. PATHOLOGICAL FINDINGS: Cytological preparations following aspiration of the mass were highly cellular and consisted of a population of large polygonal cells containing single to multiple nuclei, large prominent nucleoli, and intracytoplasmic vacuoles. Histologically, the neoplasm consisted of similar large cells surrounded by thick fibrous connective tissue trabeculae. The large polygonal cells reacted positively with antibodies against vimentin, low- and high-molecular-weight variants of cytokeratin (AE1/AE3), but not with antibodies to desmin or glial fibrillary acidic protein (GFAP). DIAGNOSIS: The clinical, gross, histological and immunohistochemical findings are similar to those reported for parachordomas in humans. Neither recurrence nor metastases were noted 18 months after surgical excision of the mass. CLINICAL RELEVANCE: This is the first reported case of a possible parachordoma in a dog, a benign tumour with cytological features of malignancy. 相似文献
The aim of the present study was to describe the strategies of the control of an outbreak of leptospiral infection in dairy cattle in Maranhão State, Northeastern Brazil. In the period from January to July 2015, 18 (17%) out of 106 cows presented abortion, six (5.7%) stillbirth, and 12 (11.3%) repeated estrus, totaling 24 animals with reproductive problems. The diagnosis of leptospirosis was based on serology (microscopic agglutination test—MAT), bacteriological culture, and polymerase chain reaction (PCR). Antibiotic therapy, vaccination protocols, and changes in management practices were suggested as control measures. Of all animals on the farm (n?=?280), 136 (48.6%) were seropositive for at least one serovar of Leptospira sp. No pure leptospiral culture was obtained. Eight of the animals with reproductive problems yielded positive PCR results (vaginal fluid of seven animals and urine and vaginal fluid of one animal). Genetic sequencing of a vaginal fluid/urine PCR-positive sample revealed Leptospira borgpetersenii. One year after the adoption of control measures, no reproductive problems were observed. Thus, leptospirosis probably caused the reproductive failures in the herd, and the control and prevention measures implemented were efficient in controlling the disease.

The objective of this experiment was to test the probiotic Efinol®L during transportation of cardinal tetra, Paracheirodon axelrodi (Schultz). For the transportation, fish were distributed in 18 plastic tanks, of which nine contained the Efinol®L (10 mg L−1; probiotic treatment) and the remaining had no probiotic (control treatment). Transport lasted 24 h and three different boxes of each treatment were sampled at 3, 12 and 24 h. Up to the 12-h sampling period, no significant difference in the survival was observed; on the other hand, survival was higher at the end of the transport (24 h) in the probiotic treatment. No significant difference was found in dissolved oxygen and temperature between treatments. Conductivity, pH and alkalinity increased along the transport, but without a difference between treatments. Ammonia increased in all treatments, although it was significantly lower in the probiotic group at 24 h. Cortisol levels were significantly higher in all transported fish when compared with the basal values. At 24 h, cortisol levels in control fish were significantly higher than those in the probiotic treatment. With the observed results, we are able to conclude that the probiotic Efinol®L is efficient during cardinal transport, lowering the mortality and helping maintain water quality possibly by lowering metabolic wastes. 相似文献
