Fourteen adult dogs of mixed breeding were given intraoperative irradiation (25 Gy) after surgical incisions were made into the greater curvature of the stomach and the ventral surface of the urinary bladder. Sequential biopsy samples were obtained 10 days to 180 days after surgical operation. All irradiated stomachs developed gastritis and persistent ulceration of the irradiated field. Microscopic changes induced by irradiation of both the bladder and stomach progressed from severe submucosal edema to severe submucosal fibrosis. A parallel progression of fibrinoid degeneration of the small blood vessels was seen in both organs. Severe gastric ulceration persisted up to 180 days after irradiation, although a degree of mucous neck cell and gastric gland regeneration did occur. Pathologic changes were less severe in the bladder than in the stomach. The bladder had greater resiliency and capability for healing and, in contrast to the stomach, showed a capability to reepithelialize the radiation-induced ulcers. Conclusions of this study are as follows: (a) the canine urinary bladder tolerated intraoperative radiation therapy after tissue resection better than did the canine stomach, (b) the combination of surgical operation and irradiation resulted in a more prolonged and complicated healing pattern than did either procedure alone, and (c) the introduction of a surgical procedure upon irradiated tissue within an undetermined time span relative to irradiation resulted in a similar pattern of disturbed healing.  相似文献

An isomorphous osmium derivative of crystalline yeast initiator transfer RNA has been prepared and interpreted to 6-angstrom resolution. The coordinates of the heavy atoms have been determined by Patterson and "direct" methods applied to the difference coefficients of the centric projections, followed by least-squares refinement. There is one dominant site per asymmetric unit, consistent with the finding by neutron-activation analysis that there is approximately one osmium atom per molecule of transfer RNA. The osmium derivative appears to be a normal substrate for enzymatic aminoacylation.  相似文献

The 2.0 angstroms crystal structure of a complex containing bee-venom phospholipase A2 (PLA2) and a phosphonate transition-state analogue was solved by multiple isomorphous replacement. The electron-density map is sufficiently detailed to visualize the proximal sugars of the enzyme's N-linked carbohydrate and a single molecule of the transition-state analogue bound ot its active center. Although bee-venom PLA2 does not belong to the large homologous Class I/II family that encompasses most other well-studied PLA2s, there is segmental sequence similarity and conservation of many functional substructures. Comparison of the bee-venom enzyme with other phospholipase structures provides compelling evidence for a common catalytic mechanism.  相似文献

Pulsed laser deposition (PLD) is a conceptually and experimentally simple yet highly versatile tool for thin-film and multilayer research. Its advantages for the film growth of oxides and other chemically complex materials include stoichiometric transfer, growth from an energetic beam, reactive deposition, and inherent simplicity for the growth of multilayered structures. With the use of PLD, artificially layered materials and metastable phases have been created and their properties varied by control of the layer thicknesses. In situ monitoring techniques have provided information about the role of energetic species in the formation of ultrahard phases and in the doping of semiconductors. Cluster-assembled nanocrystalline and composite films offer opportunities to control and produce new combinations of properties with PLD.  相似文献

Microbial degradation of fungicides on leaf surfaces after repeated applications to turfgrass was investigated. Prior andcurrent work in our laboratory has identified two characteristicsof the turfgrass leaf system that may contribute to the enhanceddegradation of fungicides after repeated application to turfgrass:(1) The leaf surface is rich in microorganisms (~108 g-1 dr wt leaf), and (2) Leaf surface microorganisms may respond to repeated fungicide applications in a manner consistentwith the phenomena of enhanced biodegradation. Field studies wereconducted on ‘Penncross' creeping bentgrass with four fungicidesrepresenting three chemical families applied either two or eight times in one growing season. Biodegradation was estimated using data from both a field study and a parallel laboratory study thatfollowed the fate of 14C-labelled fungicides. For the laboratory incubations, the locations of the residual 14C fungicides were estimated using a sequential extraction protocolthat fractionated the materials into three pools: available,retained and bound. Data from both the field and laboratory studyrefuted our hypothesis that enhanced biodegradation would developfollowing repeated applications of the fungicides onto the leafsurface. Our studies support a conclusion that a two-stagephysical sorption process leads to plant incorporation and thiscontrols most of the fungicide's fate. Thus, our data suggestthat microbial activity plays a less important part in theprocess than would be indicated by considering the size of themicrobial population on the leaves.  相似文献

Thirty-five dogs with ocular lesions associated with coccidioidomycosis were examined. Serologic and/or histologic evaluation confirmed the diagnoses of coccidioidomycosis. The disease most frequently associated with signs referable to the anterior segment of the eye, such as iritis and granulomatous uveitis. Histologic evaluation of the eye revealed a primary posterior segment disease, such as chorioretinitis or retinal separation, with an extension into the anterior segment of the eye.  相似文献
