CASE DESCRIPTION: A 9-year-old dog was evaluated for traumatic cervical myelopathy after a surgical attempt to realign and stabilize the C2 and C3 vertebrae. CLINICAL FINDINGS: The dog could not ventilate spontaneously and was tetraplegic; positive-pressure ventilation (PPV) was maintained. Myelography and computed tomography revealed spinal cord compression with subluxation of the C2 and C3 vertebrae and extrusion of the C2-3 intervertebral disk. TREATMENT AND OUTCOME: Surgically, the protruding disk material was removed and the vertebrae were realigned with screws and wire. For PPV, assist control ventilation in volume control mode and then in pressure control mode was used in the first 6 days; this was followed by synchronized intermittent mandatory ventilation until 33 days after the injury; then only continuous positive airway pressure was provided until the dog could breathe unassisted, 37 days after the injury. Physical therapy that included passive range of motion exercises, neuromuscular electrical stimulation, and functional weight-bearing positions was administered until the dog was discharged 46 days after injury; the dog was severely ataxic and tetraparetic but could walk. Therapy was continued at home, and 1 year later, the dog could run and had moderate ataxia and tetraparesis. CLINICAL RELEVANCE: Hypoventilation with tetraparesis in traumatic spinal cord injury can be successfully treated with PPV exceeding 30 days, surgery, and physical therapy.  相似文献

The SAFE dynamic model was applied to a ”clean rain” roof experiment performed within the EXMAN project. In the experiment ambient throughfall was removed and replaced with artificial ”clean throughfall”. Input of S, N and H to the forest ecosystem was reduced by 75–100%. The results of the modelling show, that the time scales of model predictions and experiments are the same. The change in base cation flux was well reproduced, while the simulation of changes in aluminum flux was less successful. pH stayed constant in the experiment as well as in the calculations.  相似文献

A comparative study of a 20-h, non-commercial, open-formula PCR method and the standard culture-based method NMKL 187, for detection of Salmonella, was performed according to the validation protocol from the Nordic organisation for validation of alternative microbiological methods (NordVal) on 81 artificially or naturally contaminated animal feed samples. The PCR method is based on culture enrichment in buffered peptone water for 16 ± 2 h followed by a magnetic beads based semi automated DNA extraction and real-time PCR analysis, including an internal amplification control. The limit of detection (LOD50) was found to be 7.19 and 7.24 CFU/sample for the PCR method and NMKL187, respectively. A very good correlation between results obtained by the two methods was found (Cohen's kappa=0.92). The relative accuracy, relative sensitivity and relative specificity were found to be 97.5%, 102.0% and 96.6%, respectively. This method is the fastest open PCR based analysis protocol for detection of Salmonella in feed samples. Implementing rapid methods such as the one validated in this study can speed up Salmonella testing of feed for food-producing animals.  相似文献

A 6-year-old Labrador retriever-cross was evaluated for an abnormal gait and head carriage 6 weeks after suffering trauma. The dog was presented with an ambulatory tetraparesis and was reluctant to move his head. Myelography and computed tomography demonstrated a subluxation of the atlanto-occipital joint with compression of the spinomedullary junction and the brain stem by the occipital bone. Removal of the compressive part of the occipital bone resulted in improvement of the clinical signs within 6 weeks, and resolution of clinical signs occurred 8 months after surgery.  相似文献

The PROFILE model is a steady state soil chemistry model which is used to calculate soil weathering rate. The model has also been used to calculate critical loads of acidity and N to forest soils, using the ratio of Ca+Mg+K to total inorganic aluminium in the soil solution as criterion, and to surface waters, using the ANC leached from the soil column as criterion. An uncertainty analysis of the PROFILE model was performed by Monte Carlo analysis, varying input parameter errors individually and simultaneously in ranges of ±10–100%, depending on parameter. The uncretainty in calculation of weathering rate, ANC leaching and ratio of Ca+Mg+K to inorganic Al in the soil solution was studied for three Nordic sites. Furthermore, the effect of uncertainty in estimates of critical load for forest soils was assessed. The analysis shows that the weathering rate can be calculated with high precision, provided that the errors of input parameter are within the range that has been reported in the literature. The model tend to be less sensitive to errors in input parameters for the range of conditions where forest damage is most likely to occur. Critical loads of acid deposition for one site calculated on the basis of the model varies within a largest range of ±40%. A study of one geographical grid included in the Swedish critical loads assessment shows that with the number of calculation points in the grid, the distribution of critical loads will stay stable independently of stochastic errors.  相似文献

Fossil rodent middens and wetland deposits from the central Atacama Desert (22 degrees to 24 degrees S) indicate increasing summer precipitation, grass cover, and groundwater levels from 16.2 to 10.5 calendar kiloyears before present (ky B.P.). Higher elevation shrubs and summer-flowering grasses expanded downslope across what is now the edge of Absolute Desert, a broad expanse now largely devoid of rainfall and vegetation. Paradoxically, this pluvial period coincided with the summer insolation minimum and reduced adiabatic heating over the central Andes. Summer precipitation over the central Andes and central Atacama may depend on remote teleconnections between seasonal insolation forcing in both hemispheres, the Asian monsoon, and Pacific sea surface temperature gradients. A less pronounced episode of higher groundwater levels in the central Atacama from 8 to 3 ky B.P. conflicts with an extreme lowstand of Lake Titicaca, indicating either different climatic forcing or different response times and sensitivities to climatic change.  相似文献

Sugar beet root rot, caused by the oomycete Aphanomyces cochlioides, is a serious and economically important disease of sugar beets world-wide. Today, disease risk assessment consists of a time-consuming greenhouse bioassay using bait plants. In the present study, a real-time quantitative PCR (qPCR) assay for determination of A. cochlioides DNA in field-infested soil samples was developed and validated using the standard bioassay. The qPCR assay proved to be species-specific and was optimized to give high amplification efficiency suitable for target copy quantification. A high correlation (R2 > 0.98, p < 0.001) with pathogen inoculum density was shown, demonstrating the suitability for monitoring soil samples. The limit of detection (LOD) was evaluated in several different soil types and varied between 1 and 50 oospores/g soil, depending on clay content. Soils with a high LOD were characterised as having a low clay content and high content of sand. Varying levels of the A. cochlioides target sequence were detected in 20 of the 61 naturally infested soil samples. Discrepancies between the bioassay and the qPCR assay were found in soils from low- and medium-risk fields. However, the qPCR diagnostic assay provides a potentially valuable new tool in disease risk assessment, enabling sugar beet growers to identify high-risk fields.  相似文献

The aim of this work was to optimize a supercritical fluid extraction (SFE)/enzymatic reaction process for the determination of the fatty acid composition of castor seeds. A lipase from Candida antarctica (Novozyme 435) was used to catalyze the methanolysis reaction in supercritical carbon dioxide (SC-CO(2)). A Box-Behnken statistical design was used to evaluate effects of various values of pressure (200-400 bar), temperature (40-80 degrees C), methanol concentration (1-5 vol %), and water concentration (0.02-0.18 vol %) on the yield of methylated castor oil. Response surfaces were plotted, and these together with results from some additional experiments produced optimal extraction/reaction conditions for SC-CO(2) at 300 bar and 80 degrees C, with 7 vol % methanol and 0.02 vol % water. These conditions were used for the determination of the castor oil content expressed as fatty acid methyl esters (FAMEs) in castor seeds. The results obtained were similar to those obtained using conventional methodology based on solvent extraction followed by chemical transmethylation. It was concluded that the methodology developed could be used for the determination of castor oil content as well as composition of individual FAMEs in castor seeds.  相似文献
