The aim was to verify the effect of follicle‐stimulating hormone (FSH) supplementation to α‐MEM+ or TCM199+ media on the in vitro development of ovarian preantral follicles (PFs) derived from collared peccaries. Ovaries (n = 5 pairs) were collected and divided into fragments destined to control group (non‐cultured) or treatments that were cultured for 7 days. The PFs morphology, growth and activation were evaluated by classical histology. The immunohistochemistry markers Ag‐NOR and PCNA were used for nuclear proliferation analysis, and the picrosirius red labelling was used for ovarian extracellular matrix (ECM) evaluation. After 7‐day culture, only the TCM199+ treatment maintained the proportion of intact PFs similar to day 1 (63.2%), but no differences were found among treatments (p > .05). In addition, a significant increase in the growing follicles proportion was verified for all the treatments, indicating follicular activation (p > .05). By the Ag‐NOR analysis, only the TCM199+/FSH maintained the nuclear proliferation similar to the first day (p > .05). The picrosirius red staining revealed that the ECM remained intact in all the treatments (p > .05). We suggest the use of TCM199+ medium supplemented of FSH for the in vitro development of peccaries PFs under 7‐day culturing conditions.  相似文献

This study aimed to evaluate various concentrations of egg yolk (5, 10, or 20%) in combination with different concentrations of glycerol (3% or 6%) added to a Tris‐based extender on the post‐thaw characteristics of sperm obtained from Tayassu tajacu. For this purpose, semen from 10 sexually male mature collared peccaries was collected by electroejaculation and evaluated for sperm motility, vigour, viability, morphology and functional membrane integrity. The ejaculates were initially extended in Tris‐fructose plus egg yolk (5%, 10% or 20%). After cooling, the semen was added to Tris‐egg yolk plus glycerol (6% or 12%), resulting in a final concentration of 3% or 6% glycerol of the extender. Straws were frozen using liquid nitrogen and thawed in a water bath at 37°C for 30 s. The frozen–thawed semen was evaluated as reported for fresh semen. After thawing, a significant decrease was verified for sperm motility and vigour, for all the samples in comparison with fresh semen. However, no differences were evidenced among treatments for any sperm characteristics evaluated (p > 0.05), except for the combination between 10% egg yolk and 6% glycerol, which provided the worst preservation of functional membrane integrity (p < 0.05). The interactions between higher concentrations of egg yolk (20%) and glycerol (6%) and also between lower concentrations of the same substances (5% egg yolk and 3% glycerol) added to the Tris‐based extender negatively affected the preservation of the normal sperm morphology after thawing (p < 0.05). In conclusion, the use of Tris‐based extender added to 10% or 20% egg yolk plus 3% glycerol is recommended for effective sperm cryopreservation in collared peccaries.  相似文献

Knowledge of the origin, organization and nature of the cassava (Manihot esculenta Crantz) germplasm collection in Brazil is incomplete due to lack of critical information on several aspects of the collection. This study verifies the utility of SSR-primed PCR markers for germplasm assessment and then utilizes these markers as well as RAPD's to characterize the Brazilian collection. We specifically address the following questions: 1)what is the relationship of morphologically closely related species to cultivated cassava? 2) What is the genetic diversity of cultivars within and between different habitats in Brazil? 3) Do agronomic traits and molecular markers reveal the same relationship among cassava accessions? 4) How complete is the Brazilian cassava collection and how well is it represented in the Word Core Collection of cassava, maintained by CIAT? Results of the interspecific studies of cassava and its wild relatives confirms the close relationship of cassava, Manihot esculenta ssp. esculenta to Manihot esculenta ssp. flabellifolia as well identifying several other closely related wild species. Next, PCR-based markers indicate a strong grouping of varieties related to the region of cultivation in Brazil. Moreover, important regions such as Cerrados and Amazon are relatively poorly represented in germplasm collections. Interestingly, the relationships of accessions based on agronomic traits are not fully congruent with relationships revealed with RAPD markers. Finally, the genetic diversity of the Brazilian cassava collection is not fully represented in the Core of the Word Core Collection of CIAT.  相似文献

Species of the genus Gracilaria are widely distributed around the world, and many are wild harvested and cultivated for phycocolloid production (agar) and human food. This study investigated the growth performance of three color strains of Gracilaria domingensis in an integrated organic system. The three chromatic variants (red, green, and brown) were cultured in a shrimp farm at two depths (surface and 20 cm). The highest relative growth rates were obtained in the first week of cultivation for all three strains studied. Although the growth rates and biomass were slightly higher at a depth of 20 cm, significant differences were not observed at different depths (p < 0.05). The growth of the three strains varied significantly over time (p < 0.001). The maximal growth values obtained were 11.25 ± 1.50% day−1 (red), 12.76 ± 3.96% day−1 (green), and 7.86 ± 1.75% day−1 (brown). The mean growth rates for the whole period were 4.63 ± 6.97, 3.38 ± 9.09, and 2.16 ± 6.61% day−1 for the red, green, and brown strains, respectively. The pigment content (chlorophyll a, phycocyanin, and phycoerythrin) of the three strains studied was higher at a depth of 20 cm, while the maximum electron transport rate (ETRmax) and photosynthetic efficiency were higher at the surface. The high growth rates obtained in this study demonstrate the great potential of this species for integrated organic farming. Moreover, the ecological success shown by the red strain suggests that this strain is the most suitable for integrated cultivation.  相似文献

The behavior in competitive adsorption-desorption reactions of Cu and Zn was studied in four calcareous soils. Cu and Zn were added to the soil by Cu, Zn, and Cu+Zn sulfate solutions in a CaSO4 background. Soil sorption of these cations was described by equilibrium isotherms that fitted either Freundlich- or Langmuir type equations, although Cu desorption data fitted only Freundlich isotherms. Cu and Zn competition was quantified by distribution coefficients, Kd, relating cation distribution between soil and solute and by the competitive Langmuir equation. The competitive Langmuir equation was the better suited to describe the Cu-Zn competitive adsorption in these soils. Distribution coefficients presented lower values when both cations were present, decreasing when the Cu and Zn concentration in solution increased (decreasing soil affinity for these cations), thereby increasing their mobility through the soil. However, the distribution coefficient of specifically adsorbed Cu in equilibrium with cations extracted by a Mg (NO) solution increased with Cu concentration. Cu adsorption was more depres 3 se 2 d by Zn than Zn adsorption by Cu. The different behavior of Cu and Zn seems dependent on the percentage Ca (CO) and, to a lesser degree, on Cu and Zn organic matter complexes, free iro 3 n 2 content, and surface precipitation on oxides and carbonates.  相似文献
