Rice genes OsDjA2 and OsERF104, encoding a chaperone protein and an APETELA2/ ethylene-responsive factor, respectively, are strongly induced in a compatible interaction with blast fungus, and also have function in plant susceptibility validated through gene silencing. Here, we reported the CRISPR/Cas9 knockout of OsDjA2 and OsERF104 genes resulting in considerable improvement of blast resistance. A total of 15 OsDjA2 (62.5%) and 17 OsERF104 (70.8%) T0 transformed lines were identified from 24 regenerated plants for each target and used in downstream experiments. Phenotyping of homozygous T1 mutant lines revealed not only a significant decrease in the number of blast lesions but also a reduction in the percentage of diseased leaf area, compared with the infected control plants. Our results supported CRISPR/Cas9-mediated target mutation in rice susceptibility genes as a potential and alternative breeding strategy for building resistance to blast disease. 相似文献

Cold molecular clouds are the birthplaces of stars and planets, where dense cores of gas collapse to form protostars. The dust mixed in these clouds is thought to be made of grains of an average size of 0.1 micrometer. We report the widespread detection of the coreshine effect as a direct sign of the existence of grown, micrometer-sized dust grains. This effect is seen in half of the cores we have analyzed in our survey, spanning all Galactic longitudes, and is dominated by changes in the internal properties and local environment of the cores, implying that the coreshine effect can be used to constrain fundamental core properties such as the three-dimensional density structure and ages and also the grain characteristics themselves. 相似文献

The objective of this study was to evaluate the stability of 3 distinct preparations of ketamine and xylazine, with or without acepromazine, stored at room temperature or at 4°C for 1, 2, and 3 mo. Drug concentrations were compared to fresh solutions, using a high performance liquid chromatography-mass spectrometry/selected-ion monitoring (HPLC-MS/SIM) assay. The concentrations of ketamine and xylazine, diluted in physiological saline, did not change over time at room temperature or at 4°C. However, acepromazine concentrations decreased over time when stored at room temperature. In contrast, undiluted ketamine-xylazine preparations gradually decreased in concentration when stored at room temperature. All of the drug concentrations remained above 90% of their original concentration when stored at 4°C. In conclusion, when diluted in physiological saline, ketamine-xylazine cocktails can be stored for 3 mo, whereas undiluted cocktails can lose efficacy over 3 mo at room temperature. Context
Managing forests under climate change requires adaptation. The adaptive capacity of forest tree populations is huge but not limitless. Integrating evolutionary considerations into adaptive forestry practice will enhance the capacity of managed forests to respond to climate-driven changes.Aims
Focusing on natural regeneration systems, we propose a general framework that can be used in various and complex local situations by forest managers, in combination with their own expertise, to integrate evolutionary considerations into decision making for the emergence of an evolution-oriented forestry.Methods
We develop a simple process-based analytical grid, using few processes and parameters, to analyse the impact of forestry practice on the evolution and evolvability of tree populations.Results
We review qualitative and, whenever possible, quantitative expectations on the intensity of evolutionary drivers in forest trees. Then, we review the effects of actual and potential forestry practice on the evolutionary processes. We illustrate the complexity of interactions in two study cases: the evolutionary consequences for forest trees of biotic interactions and of highly heterogeneous environment.Conclusion
Evolution-oriented forestry may contribute adapting forests to climate change. It requires combining short-term and long-term objectives. We propose future lines of research and experimentation. 相似文献6.
