The Alabama Black Belt area is widespread of Vertisols that are generally unsuitable for conventional septic systems; nonetheless, systems of this type have been widely used in this region for decades. In order to explore alternatives for these conventional septic systems, a real-time soil moisture–controlled subsurface drip irrigation wastewater disposal system was integrated and field tested in a Houston Vertisol for 1 year. This automated disposal system effectively limited wastewater disposal during unfavorably wet drain field conditions. However, the resulting nutrient supply into the drain field was observed to be in surplus to crop growth requirements. Soil cores taken at the conclusion of the 1-year study indicated evidence of nitrate and phosphorus leaching. Available nitrates in the top 100 cm of soil showed a decreasing trend but were higher than all other parallel controls. Soil crop-available phosphorous in the soil increased below the drip line, as result that may be ascribed to soil cracking that was not properly controlled at the test site. Despite the demonstrated deficiencies, integrating timing of wastewater disposal with soil moisture conditions can supplement existing municipal or decentralized community wastewater treatment disposal systems.  相似文献

Species-specific primers for the polymerase chain reaction (PCR) for the detection of Rhodococcus equi were developed. These primers were based on unique DNA fragments produced from R. equi reference strains and field isolates. Following random amplification of polymorphic DNA from R. equi and R. rhodochrous with a set of 40 arbitrary 10-base pair (bp) primers, a pair of species-specific primers was designed to detect a unique 700-bp fragment of R. equi chromosomal DNA. This PCR product was limited to R. equi and was not detectable in other Rhodococcus species or in a panel of additional gram-positive and gram-negative bacteria.  相似文献

We evaluated by nested PCR reaction, different cow secretions from a herd with 48% of prevalence of bovine tuberculosis (BTB), seeking to determine niches where Mycobacterium bovis could be found. Postmortem examination of 18 (75%) tuberculin reacting cows allowed demonstrates BTB-compatible lesions in six, all of them PCR positives in milk and four in colostra samples. Our results showed that up to 62% of the colostra analysed contained M. bovis DNA, whereas only 18% of milk gave a positive reaction. Moreover, in bronchoalveolar lavages from cattle with compatible lesions in lungs or lymph nodes, where macrophages account up to 90% of cells, we did not find evidences of M. bovis. Altogether, these results suggest that differences in the anti-bacterial capacity of bovine macrophages, dependent upon microenvironment and organ-specific factors, exist. Alternatively, we hypothesize that hypoxic conditions that are encountered in mammary glands macrophages could induce M. bovis entrance into a 'dormancy-like' state, and that the high number of colostra samples were M. bovis was detected, could be an indicator of reactivation during 'peripartum'.  相似文献

Estimating soil hydraulic properties, such as infiltration rate and hydraulic conductivity, is important for understanding hydrological processes such as rainfall and irrigation partitioning. Current infiltrometers can require considerable operator input to limit the number of readings that can be simultaneously performed. Therefore, the objective of this work was to develop a simple double-ring infiltrometer for automated data collection under falling head conditions. The design consisted of 15.2-cm tall inner- and outer-rings of 14.6 and 33.0 cm in diameter, respectively. The inner-ring was held in the centre of the outer-ring by a small pipe that also served as a handle. A small hole was drilled 3.8 cm from the bottom of each ring and a slightly larger hose passed through both holes. One hose end led into the inside of the inner-ring and the opposite end to the outside of the outer-ring. A pressure transducer was attached to the outside end of the hose. A datalogger was used to record the transducer output. This design was compared in-situ to a constant head method using a Mariotte bottle system on two distinct soils and tested on four different soil series ranging in textural class from a loamy sand to a clay. Soils had been fallow prior to this work for at least 2 years, except the loamy sand which had a 5-year-old Bahia grass ( Paspalum notatum ) stand. Although there were some differences between the two approaches, values estimated with the proposed method had less variability. This method allows a single user to collect multiple readings. Collected data can be used to estimate quasi-steady state and cumulative infiltration, and in situ hydraulic conductivity of saturated soil. The proposed procedure could be beneficial when multiple readings of soil hydraulic properties are required, such as when characterizing soil spatial variability.  相似文献

Schizaphis graminum (Rondani) is a serious pest of cultivated wheat (Triticum aestivum L.) and resistance is only available in other related species such as Hordeum chilense. Amphiploids between H. chilense and Triticum spp. have been obtained, and addition lines of H. chilense in wheat have been developed. Thirty-five accessions of H. chilense were screened to identify greenbug antixenosis, antibiosis and tolerance. Antixenosis was determined in a conventional host free choice test; antibiosis was measured by aphid life cycle and fecundity rate, and tolerance was tested in a conventional infestation test of 4 weeks. Two commercial barley cultivars were used as susceptible and resistant controls. Eight H. chilense accessions showed higher degrees of antixenosis than the resistant check, 19 were similar and the rest were lower. All accessions were more resistant than the susceptible check. Measured by aphid life cycle, 22 H. chilense accessions showed higher antibiosis than the resistant cv., and all exhibited a higher antibiotic effect on fecundity rates than the control. A similar degree of tolerance to that of the resistant control was observed in six accessions, the remaining entries ranged between the controls. The presence of one mechanism did not exclude the existence of other mechanisms in the same entry and therefore, independence of the different mechanisms is proposed. Most of the accessions showed higher variability than both controls for the three mechanisms, and it appears to be genetic variability within entries for the three mechanisms.  相似文献

As hill shape significantly influences water infiltration into potato hills, modification of hill shape may be an opportunity for improving fertilizer nitrogen use efficiency on sandy soils. The interactive effect of different hill shapes and rate of nitrogen (N) fertilizer application on N use efficiency was assessed in a 3-year potato (Solanum tuberosum L. cv, Russet Burbank) field experiment on Plainfield loamy sand soil at Hancock, Wisconsin, USA. A split-plot design was used with hill shape (shaped-plateau, pointed, or standard) as the main plots and in-season N rates (0, 135, 202, 269 kg?N ha?1) as the split plots with four replications in randomized complete blocks. In 1 of 3 years, potato yield and quality were increased and less N was needed to optimize yield and quality where the hills were shaped. In the other 2 years, the more blocky hills (shaped-plateau and standard) showed consistent tendencies (p?=?0.02 to 0.19) toward better crop performance; however, time of hill formation was influencing these results with root pruning likely the influencing factor. Results of this study show more blocky hills with only one hilling operation at emergence can significantly improve potato yield and quality and N use efficiency on these sandy soils.  相似文献

The dot enzyme-linked immunosorbent assay (Dot-ELISA) was compared with the passive haemagglutination test (PHT) and thin layer immunoassay (TIA) for the detection of antibodies against Fasciola hepatica in naturally and experimentally infected sheep. The infected animals gave titres from 1:25,000 to 1:204,000, while control animals gave titres of from 1:100 to 1:800. The titres of the infected sheep obtained by Dot-ELISA were 1000-2000 times higher than the ones obtained using TIA or PHT. Due to its high sensitivity, this technique could be very useful for the diagnosis of ovine fascioliasis.  相似文献

In this study, the effect of deforestation and cultivation of maize (Zea mays L.) on the physicochemical characteristics and the bacterial community structure in soil were studied at the national park Área de Protección de Flora y Fauna Nevado de Toluca in Mexico. Soil was sampled from three forested areas in the national park, from three deforested areas grazed by animals and from three areas cultivated with maize. The soil was characterized chemically and biologically, whilst the bacterial community structure was investigated through 454 pyrosequencing of the 16S rRNA gene. The pH in the forest soil decreased from 6·1 to 5·3 in the maize‐cultivated soil, whilst the soil organic C content decreased 1·4 times in the arable soil compared with the forest soil. The microbial biomass C decreased 2·9 times in the arable soil compared with the forest soil, but the metabolic quotient qCO2 (ratio basal respiration to microbial biomass C) nearly doubled. Deforestation and maize cultivation reduced the abundance of Proteobacteria, Actinobacteria and Bacteroidetes, whereas Acidobacteria, Chloroflexi, Gemmatimonadetes and Firmicutes were resistant to these changes. It was found that soil characteristics were affected negatively by deforestation and nearly half of the organic matter was lost, and on these sloped fields, erosion will be high, further decreasing soil fertility.
