Previous studies have shown that soil fungal biomass increases towards more natural, mature systems. Shifts to a fungal-based soil food web have previously been observed with abandonment of agricultural fields and extensification of agriculture. In a previous field experiment we found increased fungal biomass with reduced N fertilisation. Here, we explore relationships between fungi, bacteria, N input and grassland age on real dairy farms in the Netherlands. We hypothesised that also in pastures that are still in production there is a negative relationship between fungal biomass and fertilisation, and that fungal biomass increases with grassland age in pastures that are still in production. We expected the fungal/bacterial biomass ratio to show the same responses, as this ratio has often been used as an indicator for management changes. We sampled 48 pastures from eight organic dairy farms. Sites differed in age and fertilisation rate. We determined fungal and bacterial biomass, as well as ergosterol (a fungal biomarker). Fungal and bacterial biomass and ergosterol, showed a negative relationship with N application rate, and correlated positively with organic matter percentage. In old pastures, fungal biomass and ergosterol were higher than in younger pastures. Because bacterial biomass responded in the same way as fungal biomass, the F/B ratio remained constant, and can therefore—in our data set—not be used as an indicator for changing management. We conclude that the changes in fungal and bacterial biomass were driven by changes in organic matter quality and quantity. The negative relationship we found between N application rate and fungal biomass adds to earlier work and confirms the presence of this relationship in pastures with relatively small differences in management intensities. Earlier studies on shifts in fungal biomass focused on ex-agricultural fields or restoration projects. Here we show that fungal biomass is also higher in older agricultural pastures.  相似文献

Here we report on the frequency of melanized fungal hyphae in 323 soils, covering different land use types. The proportion of total hyphae that was melanized averaged 61%. Arable fields with loamy sand, heathlands and city parks on sandy soils had the highest percentage of melanized hyphae. In addition to the frequency determinations, a microcosm study was performed on the role of melanized hyphae in two different ecosystems: an ex-arable field and a forest. Melanized hyphae appeared to be part of the active hyphae in the forest soil but not in the ex-arable soil. In conclusion, our results indicate that (1) melanized hyphae represent a large proportion of the total fungal biomass in soils and that (2) their function might differ between ecosystems.  相似文献

Helicobacter (H.) heilmannii sensu stricto (s.s.) is a zoonotic bacterium that naturally colonizes the stomach of dogs and cats. In humans, this microorganism has been associated with gastritis, peptic ulcer disease and mucosa associated lymphoid tissue (MALT) lymphoma. Little information is available about the pathogenesis of H. heilmannii s.s. infections in humans and it is unknown whether differences in virulence exist within this species. Therefore, a Mongolian gerbil model was used to study bacterium-host interactions of 9 H. heilmannii s.s. strains. The colonization ability of the strains, the intensity of gastritis and gene expression of various inflammatory cytokines in the stomach were determined at 9 weeks after experimental infection. The induction of an antrum-dominant chronic active gastritis with formation of lymphocytic aggregates was shown for 7 strains. High-level antral colonization was seen for 4 strains, while colonization of 4 other strains was more restricted and one strain was not detected in the stomach at 9 weeks post infection. All strains inducing a chronic active gastritis caused an up-regulation of the pro-inflammatory cytokine IL-1β in the antrum. A reduced antral expression of H+/K+ ATPase was seen in the stomach after infection with 3 highly colonizing strains and 2 highly colonizing strains caused an increased gastrin expression in the fundus. In none of the H. heilmannii s.s.-infected groups, IFN-γ expression was up-regulated. This study demonstrates diversity in bacterium-host interactions within the species H. heilmannii s.s. and that the pathogenesis of gastric infections with this microorganism is not identical to that of an H. pylori infection.  相似文献

Rhinoceros populations in Africa are under severe threat as a result of surging poaching rates and risk-mitigation strategies are continuously adapted in an attempt to ensure the survival of the species. This study compared faecal glucocorticoid metabolite (fGCM) levels of two age classes of limited free-ranging female white rhinos with fGCM levels of adult free-ranging female white rhinos. Subsequently, fGCM alterations in the limited free-ranging animals were monitored following routine dehorning as a measure of the animals’ short-term physiological stress response. Baseline fGCM levels differed significantly between tested groups, with both free-ranging and limited free-ranging adult animals showing significantly higher fGCM levels compared with limited free-ranging juvenile females. In contrast, baseline fGCM levels did not differ significantly between limited free-ranging and free-ranging adult individuals. Routine dehorning procedures resulted in a short-term stress response expressed by a significant increase in fGCM levels 48 h post-dehorning, with stress steroid levels returning to pre-dehorning concentrations 72 h after the procedure.  相似文献

In an earlier study we reported the apparent stabilization of a low fungal biomass in ex-arable lands during the first decades after abandonment. It was hypothesized that the lack of increase in fungal biomass was due to constraints on development of fungi with persistent hyphae such as lignocellulolytic basidiomycetes and ericoid mycorrhizal fungi. With respect to the former group, the slow increase of the pool of lignocellulose-rich organic matter was expected to be the major constraint for their development. To study this, we enriched soil samples of one arable land, of two recently abandoned arable lands, of one older abandoned arable land and of heathland with carbon substrates that differed in composition (glucose, cellulose and sawdust). In addition, we combined the effect of carbon addition on fungal biomass development in arable and recently abandoned lands with inoculation of 1% of soil from the older abandoned site and the heathland. All treatments induced a fast increase and a subsequent rapid decline in fungal biomass in the arable and ex-arable fields. Denaturing Gradient Gel Electrophoresis (DGGE) band patterns and enzyme activities did show differences between the carbon treatments but not between the recent and older abandoned field sites, indicating a similarly responding fungal community even after three decades of land abandonment and irrespective of soil inoculation. Identification of fungi by sequencing and culturing confirmed that decomposition processes were mostly dominated by opportunistic fungi in arable and ex-arable fields. In the heathland, only a very slow increase of microbial activity was observed after addition of carbon and sequencing of DGGE bands showed that ericoid mycorrhiza (ERM) fungi were responsible for carbon decomposition. We conclude that an increase of enduringly present fungal hyphae in ex-arable land may only be possible when a separate litter layer develops and/or when suitable host plants for ERM fungi become established.  相似文献

The most important active compound in garlic is alliin. Sulfur (S) fertilization was shown to significantly increase the alliin concentration in garlic cloves, while high nitrogen (N) levels had an adverse effect. The effect of graded N and S application on the storage life of garlic has been paid little attention so far. A bifactorial field trial with 4 levels of N and S was conducted in a randomized block design. At harvest, 40 bulbs per treatment were stored under terms comparable to the storage conditions in average households (20 °C, dry, and dim) for 83 days. Every 3 weeks, samples were analyzed for their alliin and water content. The alliin concentration in peeled garlic cloves increased during storage from on average 9.2 mg g(-1) dry weight at harvest to 21.4 mg g(-1) dry weight after 83 days of storage. S fertilization increased the alliin concentration by a factor of 2.3 from 11.4 mg g(-1) in the control treatment to 26.6 mg g(-1) dry weight at the highest S level of 45 kg ha(-1) after 83 days of storage. N fertilization decreased by a trend of the alliin content. Fertilizer rates had only a minor influence on water losses from bulbs at short-term storage. After 83 days of storage, water losses were by trend lower at higher S levels, and this relationship proved to be significant when no N was applied. Best quality in terms of high alliin contents was obtained during the entire storage time at an S level of at minimum 30 kg ha(-1) S if no N was applied. The results show that the physiological S demand of 15 kg ha(-1) S for optimum yield is lower than the S requirement of 30 kg ha(-1) S for a longer storage life.  相似文献

The use of antimicrobials in production animals has become a worldwide concern in the face of rising resistance levels in commensal, pathogenic and zoonotic bacteria. In the years 2007 and 2008 antimicrobial consumption records were collected during two non consecutive production cycles in 32 randomly selected Belgian broiler farms. Antimicrobials were used in 48 of the 64 monitored production cycles, 7 farms did not use any antimicrobials in both production cycles, 2 farms only administered antimicrobials in one of the two production cycles, the other 23 farms applied antimicrobial treatment in both production cycles. For the quantification of antimicrobial drug use, the treatment incidences (TI) based on the defined daily doses (the dose as it should be applied: DDD) and used daily doses (the actual dose applied: UDD) were calculated. A mean antimicrobial treatment incidence per 1000 animals of 131.8 (standard deviation 126.8) animals treated daily with one DDD and 121.4 (SD 106.7) animals treated daily with one UDD was found. The most frequently used compounds were amoxicillin, tylosin and trimethoprim-sulphonamide with a mean TI(UDD) of 37.9, 34.8, and 21.7, respectively. The ratio of the UDD/DDD gives an estimate on correctness of dosing. Tylosin was underdosed in most of the administrations whereas amoxicillin and trimethoprim-sulphonamide were slightly overdosed in the average flock.  相似文献

Due to the great year-round demand for forest products, off-road forestry traffic occurs even when the ground is susceptible to soil compaction and rutting. We investigated the impacts of repeated passes with a laden forwarder (34?Mg) on the soil physical properties of two clear-cuts on stony till soils in northern Sweden. Core samples (n?=?71) were collected from the top 5?cm of mineral soil in and beside wheel tracks, after six passes with the forwarder. Soil physical properties were quantified using classical soil physical analyses and X-ray tomography. The hydraulic conductivity was 70% lower in the wheel tracks than in the soil beside. The X-ray image analysis indicated that this was due to the smaller total volume and lower connectivity of structural pores (φ?>?60?µm). Total porosity was 24% and 12% lower in the tracks at the two sites respectively, and mean bulk density was 1.39?g?cm?3 in the tracks, compared to 1.13?g?cm?3 beside them. To conclude, traffic changed the soil physical properties in a way that may lead to longer periods of high water content in the wheel tracks, increased risk of surface runoff and insufficient aeration for optimal seedling growth.  相似文献

The emission of gaseous sulfur (S) compounds by plants is related to several factors, such as the plant S status or fungal infection. Hydrogen sulfide (H(2)S) is either released or taken up by the plant depending on the ambient air concentration and the plant demand for S. On the contrary, carbonyl sulfide (COS) is normally taken up by plants. In a greenhouse experiment, the dependence of H(2)S and COS exchange with ambient air on the S status of oilseed rape ( Brassica napus L.) and on fungal infection with Sclerotinia sclerotiorum was investigated. Thiol contents were determined to understand their influence on the exchange of gaseous S compounds. The experiment revealed that H(2)S emissions were closely related to pathogen infections as well as to S nutrition. S fertilization caused a change from H(2)S consumption by S-deficient oilseed rape plants to a H(2)S release of 41 pg g(-1) (dw) min(-1) after the addition of 250 mg of S per pot. Fungal infection caused an even stronger increase of H(2)S emissions with a maximum of 1842 pg g(-1) (dw) min(-1) 2 days after infection. Healthy oilseed rape plants acted as a sink for COS. Fungal infection caused a shift from COS uptake to COS releases. The release of S-containing gases thus seems to be part of the response to fungal infection. The roles the S-containing gases may play in this response are discussed.  相似文献
