Factors concerning aseptic culture establishment and hardening were studied in detail in choicest Silk Banana. Effect of size of sucker (small, medium and large), carbon concentration (10, 20 and 30 g/l), season of initiation (wet and dry) and pre-treatments such as segmentation and incision to the explants were studied during initiation. Further, hardening related factors such as substrates used for primary and secondary hardening, nursery nutrition (source and frequency of application) and pre-treatments for ex vitro rooting were also studied. Results revealed that small suckers were most suited for initiations with the least contamination, maximum establishment and higher percentage of greening. Lower concentration of sucrose, though delayed greening, resulted in 100?% establishment of explants. Initiations performed during the drier period were completely free from the fungal contamination and showed less bacterial contamination than those performed during the rainy season. Segmentation of explant into four parts during first subculture supported maximum shoot proliferation by overcoming apical dominance. Coir pith was observed to be the most congenial substrate during primary hardening, whereas coir pith alone or sand: red earth: coir pith (1:1:1) supported superior performance of plantlets during secondary hardening. Single application of mono ammonium phosphate improved growth of plantlets during secondary hardening. Pre-treatment with mono ammonium phosphate gave the best response in terms of rooting and hardening of un-rooted micro-shoots. Thus, the discussed methodology could help the industries to take up commercial scale propagation of Silk Banana.  相似文献

Mass multiplication of non-Cavendish group of bananas has been limited due to non availability of reliable multiplication protocol and higher cost of production. The process of root induction and hardening are considered to be the most capital and labour intensive activities in micropropagation of any species. Carbon sources being one of the prime components governing the ex vitro survival of the plants need to be studied to maximize the success rate. Present is an attempt to study the effect of kind and concentration of carbon source on concurrent ex vitro rooting and hardening (CEVRH) in Ney Poovan banana. The results revealed that kind and concentration of carbon source present in the nutrient pool at the time of multiplication had a significant effect on the ex vitro performance of the plantlets. Better ex vitro rooting and hardening performance was exhibited by the shoots obtained from glucose (2?%) supplemented medium. Though rooting percentage was reduced in this treatment, considering the superior performance of the plantlets, it could be recommended to the commercial units for large-scale propagation of Ney Poovan banana plants. The concept of simultaneous rooting and hardening could be a boon for the varieties showing poor multiplication rate resulting in their higher cost of production.  相似文献

Endoglucanase has been isolated from Aspergillus aculeatus. The purified enzyme showed a single band and had a molecular weight of 45,000 Da as indicated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, with a specific activity of 1.4 units/mg. The purified enzyme was identified as endoglucanase, showing a high specific activity toward CM-cellulose and low specific activity toward Avicel. The activity of the isolated enzyme was optimum at a pH of 5.0 and temperature of 40 degrees C, respectively. The isoelectric point of the enzyme was 4.3. T(m) was found to be 57 degrees C. The treatment of the endoglucanase with diethylpyrocarbonate resulted in the modification of the histidine residues present in the enzyme, with a concomitant loss of 70% of the original enzymatic activity. However, carbodiimide completely inactivated the endoglucanase. The results show that the enzyme is able to sustain 50% of its activity even when heated at 90 degrees C for a period of 5 h. Endoglucanase can be used in the controlled hydrolysis of cellulose and other cellulose-rich foods. It can be used in the development of targeted functional foods from agrimaterials for value addition in the food chain.  相似文献

The common carp (Cyprinus carpio L.) is one of the major aquaculture species, contributing nearly 35% to the inland fish production in Karnataka, India. Stocks collected from Hungary (2), Indonesia and Vietnam were assessed alongside two local stocks in a series of culture performance trials with the objective of setting up a base population for developing a breeding programme. The present study deals with the genetic divergence and polymorphism in these six stocks using random‐amplified polymorphic DNA (RAPD) markers. A total of 180 decamer random primers were screened for polymerase chain reaction (PCR) amplification (OPA 1‐20, OPB1‐20, OPC1‐20, OPD1‐20, OPE1‐20, OPF1‐20, OPG1‐20, OPP1‐20 and OPM1‐20). Eight primers were selected for analysis of common carp genotypes (OPA‐7, OPA‐20, OPB‐17, OPF‐10, OP F‐9, OPG‐4, OPG‐9 and OPP‐16). Out of 492 bands recorded, 57.1% were polymorphic. Stepwise regression analysis was carried out to find best combination markers affecting body weight (P<0.001). The results demonstrate major differences in the genetic structures between different stocks. Dendrogram data showed grouping of individuals according to stocks and corresponding data variables revealed the per cent homology within the stock and also found markers correlating to the body weight.  相似文献

The effect of the chelating agent ethylenediaminetetraacetic acid (EDTA) on the structure and function of endoglucanase is studied. In the presence of 2 mM EDTA, endoglucanase showed an enhanced enzymatic activity of 1.5-fold compared to control. No further change in activity was observed with increase in the concentration of EDTA to 5 mM. The K(m) values for control and in the presence of EDTA are 0.060 and 0.044%, respectively, and K(cat) was 1.9 min(-1) in the presence of EDTA. The kinetic parameters indicated a decrease in the K(m) with an increase in the K(cat). Far-ultraviolet circular dichroism (far-UV-CD) results showed a 20% decrease in ellipticity values at 217 nm in the presence of EDTA compared to native enzyme. The apparent T(m) shifted from a control value of 57 ± 1 to 76 ± 1 °C in the presence of EDTA (5 mM). The above results suggested that the enhanced activity in the presence of EDTA is due to an increase in the K(cat) and flexible conformation of the enzyme. The stability of endoglucanase increased in the presence of EDTA.  相似文献

Phenylalanine ammonia lyase (PAL) activity was studied in different genotypes of tomato with varying degrees of resistance and susceptibility to bacterial canker disease after inoculation withClavibacter michiganensis ssp.michiganensis. In resistant genotypes the enzyme activity increased significantly 21 h after bacterial inoculation, whereas in the susceptible genotypes the activity decreased. The increase or decrease in PAL activity correlated well with the degree of host resistance along with total phenol contents. Twenty tomato (Solanum lycopersicon) cultivars were screened for resistance against bacterial spot disease incited byXanthomonas axonopodis pv.vesicatoria under field conditions with and without pathogen infection. Screening was done by artificially inoculating aX. axonopodis pv.vesicatoria suspension to 4-week-old tomato seedlings and observing them for typical symptoms of the disease. Seedlings were categorized into highly resistant, resistant, susceptible and highly susceptible cultivars on the basis of disease incidence. Tomato cultivars were screened for defense-related enzymes, total phenols and lignin contents. The temporal patterns of all these enzymes were estimated with a moderately susceptible tomato cultivar. Native PAGE analysis of both peroxidase (POX) and polyphenol oxidase (PPO) was carried out for the time course of enzyme activities and also by selecting three different tomato cultivars, following infection with the pathogen. Based on the inducible amounts of these enzymes upon pathogen infection, the tomato cultivars were correlated with the disease incidence under field conditions. A significant (P≤0.05) correlation was observed between the degree of host resistance and the enzyme levels. In highly resistant tomato cultivars the enzyme levels, total phenols and lignin contents were increased in comparison with highly susceptible tomato cultivars. Isoform analysis of POX and PPO enzymes indicated a clear difference between resistant and susceptible tomato cultivars in the number of isoforms and also in the intensity of each isoform in the presence of pathogen infection. Ney Poovan banana, the most widely cultivated mixed diploid banana, has been reported to be susceptible to Fusarium wilt like many other varieties in the world. The identification of natural variants possessing disease tolerance or resistance is one strategy to prevent losses. The aim of this study was to identify resistant lines in Ney Poovan banana, through extensive field surveys and screening of putative variants using a detached leaf-based challenging technique. The selected lines were screened under field condition to determine their economic feasibility for commercial-scale use. A total of 26 lines were observed to be resistant to the disease, out of which 24 exhibited a commercially acceptable productivity index and five best lines possessing desirable attributes were obtained.  相似文献
