A gas chromatographic (GC) method is described to determine deoxynivalenol in wheat and corn at levels as low as 20 ppb. Ground samples are extracted with water, adsorbed onto a Clin Elut column, extracted with ethyl acetate, and passed through a silica gel Sep-Pak cartridge. The final extract is then derivatized with N-heptafluorobutyrylimidazole and quantitated by GC using an electron capture detector. Recoveries are greater than 85% for spiked samples at levels of 50-1000 ppb. Results for wheat, corn, and mixed feed samples are given as well as the results of an interlaboratory study on a naturally contaminated wheat sample. 相似文献

Fingerling channel catfish Ictaturus punctarus were stocked into eight 0.04-ha ponds at 12,500 fishlha (treatment 1) and 50,000 fish/ha (treatment 2) with four ponds per treatment. At the end of of phase I (59 d) 50% of the fish were removed from each of the ponds in treatment 2 and divided equally into two ponds, forming treatment 3 (eight ponds at a density of 12,500 fish/ha). The remaining fish in treatment 2 (25,000 fish/ha) were maintained in the original ponds until the end of phase II (36 d). At this time, the fish were removed and equally divided at a density of 12,500 fish/ha into separate ponds. These ponds were continued to be denoted as treatment 2. All fish were grown for a total of 188 d. Production characteristics between treatments were compared at phases and at the end of the 188 d. There was no significant difference in feed conversion ratios due to treatment. The individual weights of the fish were higher in treatment I, but the difference occurred only in phase I. Size variabilities in treatments 2 and 3 were also higher than in treatment 1, which may cause a decrease in the percentage of marketable fish. Although there were some adverse effects due to the initial high stocking densities, overall pond production was higher in treatments 2 and 3. Treatment 2 had a daily net production of 49.9 ± 3.43 kg/ha and treatment 3 had 44.6 ± 3.81 kg/ha per d, compared to treatment 1 with only 32.4 ± 1.06 kg/ha per d (mean ± SD). 相似文献

Channel catfish (lctalurus punctatus) fingerlings stocked at a rate of 450 fish/0.04 ha pond were simultaneously cultured with fingerlings stocked in 1.25 m3 cages (0, 250, 350, or 450 fishlcage; one cage/pond). The fish in the cages were cultured and harvested for a 90–330 g (whole fish) market. The fish in the open ponds were cultured and harvested for a 490–1,140 g market. Harvest weights of open pond fish in all treatments were similar indicating that the presence of the caged fish and the associated higher daily pond feeding rates did not affect open pond production. Ninety-five to 99% of the caged fish and 96 to 98% of the open pond fish were of marketable size at harvest. Survival and food conversion ratios were similar among treatments. Results of this study indicate that total pond production can be increased (in this case up to 19%) by using a combination of open pond and cage techniques and by simultaneously producing fish for two markets. 相似文献

Cathelicidins are a class of antimicrobial peptides (AMPs) known to possess rapid and direct antimicrobial activities against a variety of microorganisms. Recently identified cathelicidins derived from alligator and sea snake were found to be more effective in inhibiting microbial growth than other AMPs previously characterized. The ability of these two cathelicidins along with the peptides, cecropin and pleurocidin, to protect channel catfish (Ictalurus punctatus, Rafinesque) and hybrid catfish (I. punctatus ♀ × blue catfish, Ictalurus furcatus, Valenciennes ♂) against Edwardsiella ictaluri, one of the most prevalent pathogens affecting commercial catfish industry, was investigated. Cathelicidin-injected fish (50 µg ml−1 fish−1) that were simultaneously challenged with E. ictaluri through bath immersion at a concentration of ~1 × 106 CFU/ml had increased survival rates compared with other peptide treatments and the infected control. Bacterial numbers were also reduced in the liver and kidney of channel catfish and hybrid catfish in the cathelicidin treatments 24 hr post-infection. After 8 days of challenge, serum was collected to determine immune-related parameters such as bactericidal activity, lysozyme, serum protein, albumin and globulin. These immune-related parameters were significantly elevated in fish injected with the two cathelicidins as compared to other peptide treatments. These results indicate that cathelicidins derived from alligator and sea snake can stimulate immunity and enhance the resistance to E. ictaluri infection in channel catfish and hybrid catfish. 相似文献

Abstract.— Underwater acoustic harassment devices (AHDs) are designed to deter seals from attacking fin-fish aquaculture cages. They may also be unintentionally disturbing non-target marine mammals. As the distance from the sound source increases, the rate at which the sound pressure level (SPL) drops per unit distance decreases. This prohibits presenting loud sounds near the cages without also creating sounds that can be detected kilometers away. In situ measurements of the sound fields of two AHDs in the Bay of Fundy, Canada, were made. The distances at which a loud AHD with short duration pulses (1.8 msec, 195 dB re 1 μPa at 1 m) and a quieter AHD with longer pulse durations (>50 msec, 166 dB re 1 μPa at 1 m) would be 80 dB or more above the detection threshold of a harbor seal Phoca vitulina were 10 and 3.5 m, respectively. On a quiet day with no nearby vessel traffic, these AHDs would be clearly detectable by harbor seals at ranges up to 2.9 and 1.3 km, respectively. By increasing the pulse length of the louder AHD to >50 msec, the sound would be more than 80 dB above the seal's detection threshold at 100 m but the sound would also be clearly detectable up to 7.2 km. It is technically possible to produce AHDs that are louder. Using very high amplitude sounds to protect finfish aquaculture cages from seal predation without encountering prohibitively large capital and operating costs may not be feasible. 相似文献

Channel catfish Ictalurus punctatus were vaccinated at 12 d of age (post‐hatch) by a 2‐min bath immersion with attenuated Edwardsiella ictaluri RE‐33 at doses of 2.5 × 105, 2.5 × 106, and 2.4 × 107 colony‐forming units CFU/mL of water. Following vaccination, RE‐33 was recovered from a greater percentage of fry that were vaccinated at the high and intermediate doses compared to fry vaccinated at the lowest dose. Independent of dose, the greatest percentage of RE‐33 positive fry occurred between 1 and 6 d post‐vaccination with a significant decrease in positive fry observed on day 12. A significant increase in mortality occurred 6 to 12 d post‐vaccination in fry vaccinated at the highest dose. No differences in post‐vaccination mortalities occurred between the other treatments. Following virulent E. ictaluri challenge, mortalities of fish vaccinated at doses of 2.5 × 106 and 2.4 × 107 CFU/mL were significantly less than those of fish vaccinated at 2.5 × 105 CFU/mL and sham‐vaccinated control fish. These data show that vaccination with RE‐33 can offer protection against subsequent virulent E. ictaluri infection. 相似文献

This study was conducted to evaluate the effects of probiotic‐amended diets fed to juvenile Nile tilapia, Oreochromis niloticus, on growth and susceptibility to Streptococcus iniae infection. Fish (average weight 16.5 ± 0.2 g) were fed five diets formulated with Bacillus subtilis strains SB3086, SB3295, SB3615, or AP193 either individually or in combination of strains SB3086 and SB3615 at a targeted concentration of approximately 4 × 107 colony‐forming units (CFU)/g of feed or with a basal control diet with no additives for 21 d. After the 21‐d growth trial, no significant difference in growth performance was observed with any probiotic‐amended diet. Results from serum bactericidal activity showed a significant difference between treatments and the control (P = 0.0002), except for the SB3295‐amended diet (P = 0.9020). Lysozyme activity was also significantly different in fish fed probiotic diets from those fed control diet (P = 0.0001). After 21 d of feeding, fish were challenged with S. iniae by intraperitoneal injection at a dosage of 8 × 106 CFU per fish. Results from the challenge also showed a significant difference between treatments and control (P = 0.0001). Overall, fish fed with strain SB3615 showed the lowest percent mortality (44.0 ± 7.2%) and those fed the control diet showed the highest mortality (77.3 ± 7.0%). The combined feeding with strains SB3086 and SB3615 did not result in any significant difference in reducing mortality because of S. iniae infection in juvenile Nile tilapia when compared with the individual probiotic treatments. 相似文献
