Within different land‐use systems such as agriculture, forestry, and fallow, the different morphology and physiology of the plants, together with their specific management, lead to a system‐typical set of ecological conditions in the soil. The response of total, mobile, and easily available C and N fractions, microbial biomass, and enzyme activities involved in C and N cycling to different soil management was investigated in a sandy soil at a field study at Riesa, Northeastern Germany. The management systems included agricultural management (AM), succession fallow (SF), and forest management (FM). Samples of the mineral soil (0—5, 5—10, and 10—30 cm) were taken in spring 1999 and analyzed for their contents on organic C, total N, NH4+‐N and NO3‐N, KCl‐extractable organic C and N fractions (Corg(KCl) and Norg(KCl)), microbial biomass C and N, and activities of β‐glucosidase and L‐asparaginase. With the exception of Norg(KCl), all investigated C and N pools showed a clear relationship to the land‐use system that was most pronounced in the 0—5 cm profile increment. SF resulted in greater contents of readily available C (Corg(KCl)), NH4+‐N, microbial biomass C and N, and enzyme activities in the uppermost 5 cm of the soil compared to all other systems studied. These differences were significant at P ≤ 0.05 to P ≤ 0.001. Comparably high Cmic:Corg ratios of 2.4 to 3.9 % in the SF plot imply a faster C and N turnover than in AM and FM plots. Forest management led to 1.5‐ to 2‐fold larger organic C contents compared to SF and AM plots, respectively. High organic C contents were coupled with low microbial biomass C (78 μg g—1) and N contents (10.7 μg g—1), extremely low Cmic : Corg ratios (0.2—0.6 %) and low β‐glucosidase (81 μg PN g—1 h—1) and L‐asparaginase (7.3 μg NH4‐N g—1 2 h—1) activities. These results indicate a severe inhibition of mineralization processes in soils under locust stands. Under agricultural management, chemical and biological parameters expressed medium values with exception for NO3‐N contents which were significantly higher than in SF and FM plots (P ≤ 0.005) and increased with increasing soil depth. Nevertheless, the depth gradient found for all studied parameters was most pronounced in soils under SF. Microbial biomass C and N were correlated to β‐glucosidase and L‐asparaginase activity (r ≥ 0.63; P ≤ 0.001). Furthermore, microbial biomass and enzyme activities were related to the amounts of readily mineralizable organic C (i.e. Corg(KCl)) with r ≥ 0.41 (P ≤ 0.01), suggesting that (1) KCl‐extractable organic C compounds from field‐fresh prepared soils represent an important C source for soil microbial populations, and (2) that microbial biomass is an important source for enzymes in soil. A local strain of Nannochloropsis granulata (Ng) has been reported as the most productive microalgal strain in terms of both biomass yield and lipid content when cultivated in photobioreactors that simulate the light and temperature conditions during the summer on the west coast of Sweden. To further increase the biomass and the biotechnological potential of this strain in these conditions, mixotrophic growth (i.e., the simultaneous use of photosynthesis and respiration) with glycerol as an external carbon source was investigated in this study and compared with phototrophic growth that made use of air enriched with 1–2% CO2. The addition of either glycerol or CO2-enriched air stimulated the growth of Ng and theproduction of high-value long-chain polyunsaturated fatty acids (EPA) as well as the carotenoid canthaxanthin. Bioassays in human prostate cell lines indicated the highest antitumoral activity for Ng extracts and fractions from mixotrophic conditions. Metabolomics detected betaine lipids specifically in the bioactive fractions, suggesting their involvement in the observed antitumoral effect. Genes related to autophagy were found to be upregulated by the most bioactive fraction, suggesting a possible therapeutic target against prostate cancer progression. Taken together, our results suggest that the local Ng strain can be cultivated mixotrophically in summer conditions on the west coast of Sweden for the production of high-value biomass containing antiproliferative compounds, carotenoids, and EPA.  相似文献

It is predicted that dryland salinity will affect up to 17 Mha of the Australian landscape by 2050, and therefore, monitoring the health of tree plantings and remnant native vegetation in saline areas is increasingly important. Casuarina glauca Sieber ex Spreng. has considerable salinity tolerance and is commonly planted in areas with a shallow, saline water table. To evaluate the potential of using the nitrogenous composition of xylem sap to assess salinity stress in C. glauca, the responses of trees grown with various soil salinities in a greenhouse were compared with those of trees growing in field plots with different water table depths and groundwater salinities. In the greenhouse, increasing soil salinity led to increased allocation of nitrogen (N) to proline and arginine in both stem and root xylem sap, with coincident decreases in citrulline and asparagine. Although the field plots were ranked as increasingly saline-based on ground water salinity and depth-only the allocation of N to citrulline differed significantly between the field plots. Within each plot, temporal variation in the composition of the xylem sap was related to rainfall, rainfall infiltration and soil salinity. Periods of low rainfall and infiltration and higher soil salinity corresponded with increased allocation of N to proline and arginine in the xylem sap. The allocation of N to citrulline and asparagine increased following rainfall events where rain was calculated to have infiltrated sufficiently to decrease soil salinity. The relationship between nitrogenous composition of the xylem sap of C. glauca and soil salinity indicates that the analysis of xylem sap is an effective method for assessing changes in salinity stress in trees at a particular site over time. However, the composition of the xylem sap proved less useful as a comparative index of salinity stress in trees growing at different sites.  相似文献

Induction of discolored wood in Scots pine (Pinus sylvestris L.) trees by treatment with ethylene, carbon dioxide, nitrogen (hypoxia) or wounding from early April to late September was investigated. All treatments induced formation of discolored wood upward and downward from the drill hole. The amount of discolored wood formed above the drill hole depended on the treatment in the following order: ethylene > carbon dioxide = nitrogen > wounding; and below the drill hole in the order: ethylene > carbon dioxide = nitrogen = wounding. Based on chemical analyses (HPLC/UV, GS/MS, LC/MS and 1H-NMR), discolored wood induced by wounding or treatment with ethylene or carbon dioxide showed compositional similarities to natural heartwood, whereas discolored wood induced by nitrogen treatment showed fewer similarities to natural heartwood. The results suggest that ethylene is an important factor controlling heartwood formation, although wounding and internal concentrations of carbon dioxide may also play a role.  相似文献

Flowering time is the most critical developmental stage in wheat, as it determines environmental conditions during grain filling. Thirty-five spring durum genotypes carrying all known allele variants at Ppd-1 loci were evaluated in fully irrigated field experiments for three years at latitudes of 41°N (Spain), 27°N (northern Mexico) and 19°N (southern Mexico). Relationships between weight of central grains of main spikes (W) and thermal time from flowering to maturity were described by a logistic equation. Differences in flowering time between the allele combination causing the earliest (GS100/Ppd-B1a) and the latest (Ppd-A1b/Ppd-B1a) flowering were 7, 20 and 18 days in Spain, northern Mexico and southern Mexico, respectively. Flowering delay drastically reduced the mean grain filling rate (R) and W at all sites. At autumn-sowing sites, an increase of 1°C in mean temperature during the first half of the grain filling period decreased W by 5.2 mg per grain. At these sites, W was strongly dependent on R. At the spring-sowing site (southern Mexico), W depended on both R and grain filling duration. Our results suggest that incorporating the allele combinations GS100/Ppd-B1a and GS105/Ppd-B1a (alleles conferring photoperiod insensitivity) in newly released varieties can reduce the negative effects of climate change on grain filling at the studied latitudes.  相似文献

Genome-wide association studies (GWAS) are useful to facilitate crop improvement via enhanced knowledge of marker-trait associations (MTA). A GWAS for grain yield (GY), yield components, and agronomic traits was conducted using a diverse panel of 239 soft red winter wheat (Triticum aestivum) genotypes evaluated across two growing seasons and eight site-years. Analysis of variance showed significant environment, genotype, and genotype-by-environment effects for GY and yield components. Narrow sense heritability of GY (h 2  = 0.48) was moderate compared to other traits including plant height (h 2  = 0.81) and kernel weight (h 2  = 0.77). There were 112 significant MTA (p < 0.0005) detected for eight measured traits using compressed mixed linear models and 5715 single nucleotide polymorphism markers. MTA for GY and agronomic traits coincided with previously reported QTL for winter and spring wheat. Highly significant MTA for GY showed an overall negative allelic effect for the minor allele, indicating selection against these alleles by breeders. Markers associated with multiple traits observed on chromosomes 1A, 2D, 3B, and 4B with positive minor effects serve as potential targets for marker assisted breeding to select for improvement of GY and related traits. Following marker validation, these multi-trait loci have the potential to be utilized for MAS to improve GY and adaptation of soft red winter wheat.  相似文献

A number of different beverage products claim to have antioxidant potency due to their perceived high content of polyphenols. Basic and applied research indicates that pomegranate juice (PJ), produced from the Wonderful variety of Punica granatum fruits, has strong antioxidant activity and related health benefits. Although consumers are familiar with the concept of free radicals and antioxidants, they are often misled by claims of superior antioxidant activity of different beverages, which are usually based only on testing of a limited spectrum of antioxidant activities. There is no available direct comparison of PJ's antioxidant activity to those of other widely available polyphenol-rich beverage products using a comprehensive variety of antioxidant tests. The present study applied (1) four tests of antioxidant potency [Trolox equivalent antioxidant capacity (TEAC), total oxygen radical absorbance capacity (ORAC), free radical scavenging capacity by 2,2-diphenyl-1-picrylhydrazyl (DPPH), and ferric reducing antioxidant power (FRAP)]; (2) a test of antioxidant functionality, that is, inhibition of low-density lipoprotein (LDL) oxidation by peroxides and malondialdehyde methods; and (3) evaluation of the total polyphenol content [by gallic acid equivalents (GAEs)] of polyphenol-rich beverages in the marketplace. The beverages included several different brands as follows: apple juice (3), a?aí juice (3), black cherry juice (3), blueberry juice (3), cranberry juice (3), Concord grape juice (3), orange juice (3), red wines (3), iced tea beverages (10) [black tea (3), green tea (4), white tea (3)], and a major PJ available in the U.S. market. An overall antioxidant potency composite index was calculated by assigning each test equal weight. PJ had the greatest antioxidant potency composite index among the beverages tested and was at least 20% greater than any of the other beverages tested. Antioxidant potency, ability to inhibit LDL oxidation, and total polyphenol content were consistent in classifying the antioxidant capacity of the polyphenol-rich beverages in the following order: PJ>red wine>Concord grape juice>blueberry juice>black cherry juice, a?aí juice, cranberry juice>orange juice, iced tea beverages, apple juice. Although in vitro antioxidant potency does not prove in vivo biological activity, there is also consistent clinical evidence of antioxidant potency for the most potent beverages including both PJ and red wine.  相似文献
