Lactobacillus rhamnosus GG (LGG) is a well‐established probiotic strain. The beneficial properties of this strain are partially dependent on its prolonged residence in the gastrointestinal tract, and are likely influenced by its adhesion to the intestinal mucosa. The pilin SpaC subunit, located within the Spa pili structure, is the most well studied LGG adhesion factor. However, the binding epitopes of SpaC remain largely unknown. The aim of this study was to evaluate the binding properties of SpaC to the carbohydrate moieties of intestinal glycoconjugates using a recombinant SpaC protein. In a competitive enzyme‐linked immunosorbent assay, SpaC binding was markedly reduced by addition of purified mucin and the mucin oligosaccharide fraction. Histochemical staining revealed that the binding of SpaC was drastically reduced by periodic acid treatment. Moreover, in the surface plasmon resonance‐based Biacore assay, SpaC bound strongly to the carbohydrate moieties containing β‐galactoside at the non‐reducing terminus of glycolipids. We here provide the first demonstration that SpaC binds to the oligosaccharide chains of mucins, and that the carbohydrate moieties containing β‐galactoside at the non‐reducing termini of glycoconjugates play a crucial role in this binding. An 11-year-old neutered male Domestic Shorthair cat presented with a 3-month history of hypoglycemia, two episodes of seizure, and intermittent tick-like signs. Serum biochemistry revealed severe hypoglycemia associated with high insulin concentrations. Dynamic abdominal computed tomography (CT) indicated two pancreatic masses, which were enhanced most during the late arterial phase but had different degrees and variations of attenuation. Partial pancreatectomy was performed. Histopathology and immunohistochemistry confirmed that one mass was an insulinoma and the other was an ectopic splenic tissue, consistent with the differences in imaging findings. When an intrapancreatic lesion with hyper-attenuation on dynamic abdominal CT is detected, not only insulinoma or metastasis of malignancies but also intrapancreatic accessory spleen (IPAS) should be considered as differential diagnoses. 相似文献

Heat treatments change the chemical and physical properties of wood and dimensional stability and hygroscopicity are affected
as a result of modifications of wood cell components. This study evaluated the water absorption of wood specimens treated
with boron compounds followed by heat treatment. Sugi (Cryptomeria japonica D. Don) sapwood specimens treated with either boric acid (BA) or disodium octoborate tetrahydrate (DOT) solutions were heat-modified
at either 180° or 220°C for 2 or 4 h. Carbohydrate composition and water absorption of the specimens were then measured and
compared with those of untreated and unheated specimens. Wood carbohydrates were significantly degraded in the specimens after
heat treatment. The heat treatment evidently decreased the water absorption and the heat-modified specimens absorbed less
water than unheated specimens. The higher the treatment temperature and the longer the treatment time, the lower the amount
of absorbed water. The boron-treated and heat-modified specimens, however, showed increased water absorption due to the hygroscopic

In Japan, ammoniacal copper quaternary wood preservatives type-1 (ACQ-1), which contains copper and benzalkonium chloride
as its active ingredients, is among the most widely used wood preservatives in the pressure treatment of wood. Benzalkonium
chloride (BAC) in commercial ACQ-1 products mainly comprises C12 and C14 homologues. In the present study, the leaching characteristics
of these BAC homologues were investigated using the heartwood and sapwood portions of Japanese cedar, Japanese larch, and
Sakhalin fir treated with ACQ-1 and 1% monoethanolamine (MEA) solution containing equimolar amounts of homologues. Distilled
water (DW) and artificial seawater (SW) were used as leaching media. Consequently, it was observed that the leaching rate
of the C12 homologue tended to be higher than that of the C14 homologue in DW. The leaching of C12 homologues was accelerated
by using SW, resulting in a significantly higher leaching rate than the C14 homologue using SW. It was thought that the difference
in the hydrophobicities based on alkyl chain lengths resulted in these phenomena. However, when the heartwood portion of Japanese
larch was treated with the homologues in MEA, the leaching rate of the C14 homologue was significantly higher than that of

In order to confirm the in vivo effectiveness of anti- feline herpesvirus type 1 (FHV-1) mouse-cat chimeric antibody (FJH2), and anti-feline calicivirus (FCV) mouse-cat chimeric antibody (F1D7), cats that had been experimentally infected with FHV-1 or FCV were administered intravenously with the chimeric antibodies, and observed for clinical manifestations. The symptoms due to FHV-1 or FCV infection in the cats administered FJH2 or F1D7 were obviously decreased when compared with those of the non-administered control cats. From these results, it was confirmed that both FJH2 and F1D7 were effective at reducing the appearance of symptoms due to FHV-1 and FCV infection, respectively. 相似文献

Oral administration of O,O,S-trimethyl phosphorothioate (OOS), an impurity present in technical formulations of malathion, has been shown to be associated with a high incidence of pneumonia in rats and to be highly immunosuppressive in mice. Based on these findings, an in vitro model was established to study the effect of this and other organophosphorus compounds on murine cytotoxic T-lymphocyte (CTL) responses. The organophosphorus compounds were tested for their ability to block in vitro generation of CTL responses to alloantigen and/or the expression of these cytotoxic responses. Responses were generated in C57Bl/6 (H-2b) spleen cells to mitomycin C-blocked P815 (H-2d) tumor cells. The cytotoxicity of the cultured splenocytes to P815 target was measured using a 4-hr chromium release assay. These data demonstrated that malathion was able to block the ability of splenocytes to sensitize to P815 at concentrations as low as 25 μg/ml, but was not able to block the expression of cytotoxicity by mature killer T cells. The same was true for OOS which had been activated by preincubation with rat liver postmitochondrial supernatant (PMS). Activated OOS blocked the generation of CTL responses at concentrations as low as 75 μg/ml while having no effect on mature cytotoxic cells. In fact, both malathion and activated OOS were no longer able to suppress CTL responses if treatment was performed as early as 24 hr after exposure to antigen. Additionally, it was demonstrated that when malathion was preincubated with PMS it was no longer suppressive and that OOS without activation failed to suppress CTL responses. 相似文献

The relationship between the moisture content (MC) of wood blocks and the feeding activities of two Japanese subterranean termites, Coptotermes formosanus Shiraki and Reticulitermes speratus (Kolbe), was investigated with two choice tests. When three wood blocks with low MC (6%–12%), middle MC (79%–103%), and high MC (140%–182%) were exposed to workers of C. formosanus and R. speratus in a choice test, the feeding preferences were found in the middle MC blocks for C. formosanus, and the middle and high MC blocks for R. speratus, although wood blocks of low MC were also attacked. In a second choice test, wood blocks consisting of five pieces with water-impregnated (MC: 133%–191%) top, middle, or bottom piece were exposed to workers of both species. This study describes a laboratory evaluation of the efficacy of two alkylammonium compounds [didecyldimethylammonium tetrafluoroborate
(DBF) and didecyldimethylammonium chloride (DDAC)] when applied via vacuum impregnation or superficial treatment. Treated
wood specimens were tested for their termite and microbial resistance under controlled laboratory conditions. The higher chemical
retentions were needed to suppress the feeding by Coptotermes formosanus ≦3% mass loss in the multichoice test than in the no-choice test. The DBF and DDAC retention levels necessary to meet the
performance requirement ≦3% mass loss after 12-week fungal exposure varied with wood species. The retention level of 3 kg/m3 for DBF and DDAC was generally high to keep the nondurable wood species free of decay. Although there was no difference between
DBF and DDAC in the efficacy against decay and termite attack, the former slightly outperformed the latter as an antimold
