通过重叠PCR扩增得到烟草丛顶病毒(Tobacco bushy top virus,TBTV)中国分离物RdRp的编码序列,构建以pMALC2X为基础载体的原核表达载体pMAL-TB-RdRp。0.5 mM IPTG诱导可特异性表达分子量约为120 kDa的MBP-RdRp融合蛋白。温度梯度实验显示,18℃下诱导表达的MBP-RdRp融合蛋白的可溶性比例较高,约17%;经亲和层析纯化的MBPRdRp可特异性识别TBTV正链和负链的3'末端序列,催化体外复制;对正负链的3'末端的体外复制效率存在差别,识别负链3'末端的体外复制效率明显高于正链3'末端。本研究创建的TBTV RdRp介导的体外复制体系为进一步研究TBTV基因组复制调控奠定了基础。 相似文献

生食牡蛎的安全与质量是影响其销售价格的关键要素。本文首先回顾了各国关于生食牡蛎的安全质量管理现状,并从安全性、活力状态、风味质量三个方面综述。然后从流通链的角度,系统分析了养殖、捕捞、清洗、净化、运输、销售等关键节点对生食牡蛎品质的影响。牡蛎从养殖到销售的整个过程中,可能受到多重安全危害因子的影响,对其食用安全性造成潜在威胁,其中主要包括贝类毒素、重金属、有害微生物、病毒等,通过规范的养殖和流通管理可消除这些危害。此外,从离水到餐桌,牡蛎暴露于一系列可能引发应激反应的环境压力源(干露、温度、氧气、密度等),导致活力状态和风味品质下降严重,也造成了重大经济损失。由于胁迫方式、胁迫强度、持续时间等差异,不同节点上相同或不同的应激源的影响非常复杂。快速准确的活力评价是进行流通中质量管理的基础,本文综述了牡蛎流通过程中遭遇多重胁迫时,其代谢与活力的关联,以及代谢物对风味特性的影响。为生食牡蛎进行精准的品质控制提供理论依据。 相似文献

根据无乳链球菌的cfb基因和海豚链球菌16SrRNA基因序列,设计、合成2对引物,优化扩增条件,建立了快速鉴别无乳链球菌和海豚链球菌的双重PCR方法.用该方法扩增无乳链球菌和海豚链球菌,可分别获得474、296bp的特异性片段,扩增嗜水气单胞菌、铜绿假单胞菌等其他常见鱼病原菌无特异性片段.该方法可实现对无乳链球菌和海豚链球菌的快速鉴别,具较高的灵敏度,可检测到基因组含量分别为3.2×10-3ng/μL的无乳链球菌和3.0×10-2ng/μL的海豚链球菌.对采集自广东与海南两省养殖罗非鱼病鱼的11份病原样品进行检测,均可获得474bp片段,测序与BLAST分析结果表明,扩增到的序列均为无乳链球菌cfb基因序列,可从分子水平上确定这些样品为无乳链球菌,与生化鉴定结果一致. 相似文献

Essential oils of peppermint Mentha piperita L. (Lamiaceae), which are used in flavors, fragrances, and pharmaceuticals, were investigated for their antimicrobial properties against 21 human and plant pathogenic microorganisms. The bioactivity of the oils menthol and menthone was compared using the combination of in vitro techniques such as microdilution, agar diffusion, and bioautography. It was shown that all of the peppermint oils screened strongly inhibited plant pathogenic microorganisms, whereas human pathogens were only moderately inhibited. Chemical compositions of the oils were analyzed by GC and GC/MS. Using the bioautography assay, menthol was found to be responsible for the antimicrobial activity of these oils. 相似文献

The effects of months of the year and nitrogen (N) sources on salad rocket (Eruca sativa Mill.) yield, quality, and nitrate accumulation was investigated during the years 2002 and 2003. In both years, seeds were sown on the first day of April, May, June, July, August and September. Three different nitrogen sources were used: farmyard (cattle) manure (100 tonnes·ha?1), calcium nitrate [Ca(NO3)2]-15.5% N (150 kg N·ha?1) and ammonium sulfate [(NH4)2SO4)]-21% N (150 kg N·ha?1). Yield, leaf color, dry matter, vitamin C and total glucosinolate content and nitrate accumulation was assessed. Growing months affected all the assessed parameters significantly both years, with the exception of hue angle in 2003, whereas nitrogen source only influenced yield and nitrate accumulation in 2003. In both years the highest yield was obtained in April, but vitamin C and total glucosinolate contents were higher during summer months, and leaves were slightly darker colored. Chemical fertilizers increased the yield compared to farmyard manure; however, they also increased nitrate accumulation slightly, without any significant difference between them. Nitrate accumulation never exceeded 300 mg kg?1 fresh weight (FW), which is well below the acceptable daily intake of 3.7 mg nitrate per kg?1 bodyweight set by European Commission's Scientific Committee on Food. 相似文献

The effect of restricted feeding on growth, feed efficiency, and body composition was studied in juveniles of gilthead sea bream, Sparus aurata. Juveniles (6.4 g) were stocked into 12 tanks at a density of 16 fish per tank. Four different feeding schedules were tested on triplicate groups of juvenile fish: (1) control fed for 48 d without deprivation, (2) starvation for 1 d and then refed for 2 d (S1), (3) 50% satiation for 2 d and then refed to apparent satiation for 2 d (R2), and (4) 50% satiation for 6 d and then refed to apparent satiation for another 6 d (R6). Results indicated that all fish subjected to cycled restricted feeding regimes were unable to achieve catching up with control group. The specific growth rate of fish in the control was significantly higher than those in S1, R2, and R6, which were not significantly different from each other. Protein efficiency and protein productive value were significantly higher in R2 compared to control, S1, and R6. Fish in R2 had lowest feed conversion ratio (1.12) compared to the control (1.17). Body protein composition in R6 was less than that of the control, S1, and R2, while moisture, lipid, and ash content were not significantly different compared to the control. 相似文献

在青海省乌兰县舍外温度-6.30℃条件下,选用40头荣昌猪分作2组,每组20头,在自行设计的半开放-封闭式多用塑料暖棚猪舍与常用简易塑料暖棚猪舍内开展对比试验。结果表明,前者可显著提高舍内温度,降低舍内湿度,且育猪增重效果明显。 相似文献
