Exercise-related changes in body protein 'turnover' and in the absolute amounts of body protein were studied in racing pigeons. Whole body radioactivity was followed in racing and control (limited exercise) birds after protein labelling by the injection of 75Seselenomethionine. Because of re-utilisation of the label this does not give a true picture of body protein turnover but the comparative data suggested an increased turnover in racing compared to control birds. Carcase analysis on a group of pigeons demonstrated a water content for lean body mass of 72.7 per cent +/- 3.54. Lean body mass and exchangeable body potassium were used as indices of total body protein in a group of pigeons participating in an endurance race (15 + hours of flying). The results indicated that no body protein had been used as an energy source. These findings are compatible with the presence in pigeons of a small labile pool or pools of protein. The presence and characteristics of such pools remains to be determined.  相似文献

Chickens from a randomly bred genetic line were segregated into high and low growth rates and high and low water-holding capacities (WHCs). The objective of this study was to identify protein markers associated with slow and fast growth rates and low and high WHCs from water-soluble protein (WSP) and crude myofibrillar protein (CMP) extracts of chicken breast muscle. Proteins were fractionated using two-dimensional electrophoresis, and a total of 22 protein spots were selected, excised, and analyzed by in-gel tryptic digestion and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Proteins expressed in extracts from slow and fast growth rates and low and high WHCs included metabolic enzymes, such as creatine kinase, pyruvate kinase, triosephosphate isomerase, and ubiqitin; housekeeping proteins, such as heat shock protein; contractile proteins, such as myosin heavy chain; actin; and also MHC isoforms and actin isoforms. The mass spectra of 20 protein spots significantly matched (protein score >83; P < 0.05) an online database. In CMP, there were unique proteins that were present only in the fast-growth population: gi|118099530 , gi|20664362 , gi|71895043 , gi|114794125 , gi|297343122 , and gi|71895043 . This information identified protein markers associated with growth rate and water holding capacity. Some of those protein markers could be added to the chicken database.  相似文献

The present study aims to evaluate carbon recovery and analytical precision of two methods based on dry combustion for organic carbon (OC) quantification in base metal mine tailings: (1) IC subtraction method (inorganic C(IC) measured and subtracted from total C) and (2) direct OC quantification after acid pretreatment. Results showed IC subtraction method effectively hydrolyzed a range of IC minerals [calcium carbonate (CaCO3), magnesium carbonate (MgCO3), dolomite, magnesite, calcite] with satisfactory IC (as CO2 released) recovery (87–103 percent). In the direct OC quantification method, 5 M hydrochloric acid (HCl) pretreatment resulted in a satisfactory recovery (76–92 percent) of most organic compounds [ethylenediaminetetraacetic acid (EDTA), cellulose, plant litter, and charcoal], except for water-soluble OC (sucrose, 40 percent recovery). The precision of both methods declined when C levels were <5 g kg?1 with RSD (Relative Standard Deviation) > 10 percent. The OC values in test samples of base metal mine tailings were comparable between the two methods. However, IC subtraction method is not applicable for tailings with low OC levels (<5 g kg?1) until the precision is substantially improved. Moreover, compared to the IC subtraction method, OC values are significantly lower in direct OC quantification method for tailings with high OC levels.  相似文献

Production diseases of the dairy cow are caused by a level of production inconsistent with nutrient intake, provision of an inadequate diet, an unsuitable environment, an inappropriate breeding policy or various combinations of these factors. Although the transition period of 3 weeks pre-calving until 3 weeks post-calving is associated with a peak incidence of production disease, the effects of these diseases on dairy cow health and productivity extend far into the following lactation. Recent advances in understanding of production diseases include the emergence of propylene glycol and rumen protected choline as the supplements of choice for preventing fatty liver and the absence of any preventative effect of increased energy density in the close-up dry period diet on this condition; the linear negative influence of dietary cation anion difference (DCAD) on the incidence of milk fever regardless of urinary pH or the target level of dietary DCAD achieved; the inflammatory response associated with subacute rumen acidosis and its effect on feed intake; an increased awareness of the potential for antioxidant status to improve immunity and health in the transition period; the development of more standardised diagnostic criteria and treatment protocols for uterine infection. A significant body of knowledge already exists which should allow for the optimal management and prevention of bovine production diseases. One of the important challenges facing the dairy industry is the development, implementation and economic assessment of practical, integrated, blueprints of best practice for prevention of the production diseases and other diseases of the dairy cow.  相似文献

Sixty cows (40 multiparous and 20 primiparous) averaging 140 days in milk (DIM) were assigned to five treatments to evaluate the effect of pasture allowance and supplements of a) maize silage b) high crude protein concentrate and, c) low crude protein concentrate on milk production and nitrogen (N) excretion. Two of the treatments (HG and LG) were offered herbage only (allowances of 20 and 15 kg dry matter (DM) per cow/day, respectively) while the remaining three treatments were offered a herbage allowance of 15 kg DM per cow/day plus 4 kg DM per cow/day of maize silage (M), a high CP concentrate (CP concentration of 194 g/kg DM) (HC), or citrus pulp (CP concentration of 69 g/kg DM) (LC). Total DM intake (DMI) was similar for HG, M, HC, and LC but was lower for LG (15.2 kg DM per cow/day) than HC (17.4 kg DM per cow/day). The reduction in pasture DMI per kg of supplement DM ingested was 0.44, 0.45, and 0.54 kg for cows offered maize silage, high CP concentrate, and low CP concentrate, respectively. Milk yield was greater for the supplemented treatments (23.7–24.7 kg/day) than for LG (20.7 kg/day) but not for HG (23.2 kg/day). Milk fat concentration was greater for HC (35.3 g/kg) than for HG, M, and LC but not greater than LG, while milk protein concentration was greater for HG (34.8 g/kg) than for LG and HC but not greater than M and LC. The greatest levels of N and PDIN intake were recorded for HG (662 and 2502 g/day) and HC (654 and 2506 g/day) which were greater than LC but not greater than LG and M. Treatment HC recorded the highest PDIE intake (1743 g/day) which was greater than LG, M and LC but not greater than HG. Output of N in milk was greater on HC (134 g/day) than on LG but was not greater than on HG, M, and LC. Faecal N excretion was greater on HG (171 g/day) than on all other treatments while estimated urinary N excretion was greater on HG and HC than LC (320 g/day). Treatment LC had a higher proportion of N output in milk (0.23) than treatment LG but not higher than HG, M and HC treatments. Urinary N expressed as a proportion of total N excretion was lower for HG (0.68) than all other treatments. The results of this study show that there is a large response in milk production to supplementing cows on a restricted grass allowance and that cows offered low CP supplements had similar levels of production to those offered high CP supplements. Nitrogen utilisation was improved by offering supplements of lower CP content.  相似文献

A nutritional evaluation of an Irish dairy herd indicated gross overfeeding of late lactation cows, over-conditioning of cows at parturition and a high rate of body condition loss in early lactation. Metabolisable-energy based nutritional modelling software was used to guide recommended dietary changes to prevent excessive condition gain in late lactation. Immediately following the implementation of the changes there was an unexpected reduction in performance affecting both milk yield and protein concentration. An investigation into the poor performance revealed underestimation of peak milk yield; over-estimation of maize silage quality; a large difference in the concentrate being fed compared to the concentrate recommended, and failure of the blend of concentrate ingredients to maintain the intended proportions in the in-parlour feeding system. The estimated maximum cumulative effect of these errors was to cause undersupply of energy and protein in the recommended diet of 16% and 3% respectively to cows in early lactation. Use of a net-energy nutritional model would have indicated a requirement for a higher energy supply in this case. This report highlights the challenges in obtaining accurate on-farm data for use in dairy cow nutritional models.  相似文献

Botanical pyrethrins and synthetic pyrethroids are highly potent and environmentally safe insecticides that are used to control a wide range of disease vector and pest arthropods. Unfortunately, resistance to these insecticides has been demonstrated in numerous medically important mosquito species. In this study, adult Culex pipiens sensu lato were captured in agricultural and urban locations in Fresno County, California, and subsequently exposed to a commercial formulation of pyrethrin insecticide by ultra-low-volume spraying. Following insecticide exposure, two pyrethroid-like, fluorescent substrates (4-methyl-2-oxo-2H-chromen-6-yl, cis-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropanecarboxylate (cis-DCVC) and 4-methyl-2-oxo-2H-chromen-6-yl, cis-3-((Z)-2-chloro-3,3,3-trifluoroprop-1-enyl)-2,2-dimethylcyclopropanecarboxylate (cis-TFMCVC)) and 1-chloro-2,4-dinitrobenzene (CDNB) were used to measure esterase and glutathione S-transferase (GST) activities in surviving mosquitoes. Elevated esterase activity (2.5-fold) was found in surviving urban mosquitoes at 12-h post-pyrethrin exposure (in comparison to non-insecticide-exposed control mosquitoes) when cis-TFMCVC was used as a substrate. Additionally, when CDNB was used as a substrate, 2.8-fold higher GST activity was found. A simple assay was established using our pyrethroid-like, fluorescent substrates that was able to detect low-level esterase activities in homogenates made from individual mosquitoes. The cis-TFMCVC-based assay suggested that esterase activity plays a role in pyrethrin resistance in urban mosquitoes in California.  相似文献
