Association of transfer RNA acceptor identity with a helical irregularity   总被引:8,自引:0,他引:8  
The aminoacylation specificity ("acceptor identity") of transfer RNAs (tRNAs) has previously been associated with the position of particular nucleotides, as opposed to distinctive elements of three-dimensional structure. The contribution of a G.U wobble pair in the acceptor helix of tRNA(Ala) to acceptor identity was examined with synthetic amber suppressor tRNAs in Escherichia coli. The acceptor identity was not affected by replacing the G.U wobble pair in tRNA(Ala) with a G.A, C.A, or U.U wobble pair. Furthermore, a tRNA(Ala) acceptor identity was conferred on tRNA(Lys) when the same site in the acceptor helix was replaced with any of several wobble pairs. Additional data with tRNA(Ala) show that a substantial acceptor identity was retained when the G.U wobble pair was translocated to another site in the acceptor helix. These results suggest that the G.U wobble pair induces an irregularity in the acceptor helix of tRNA(Ala) to match a complementary structure in the aminoacylating enzyme.  相似文献

Visual and infrared images produced by the Seasat visible and infrared radiometer (VIRR) are adequate for the identification of cloud, land, and water features. A statistical comparison of VIRR-derived sea-surface temperatures in a cloud-free region with a National Oceanic and Atmospheric Administration analysis based on various surface measurements taken in the same region showed agreement to +/- 1.7 degrees K root-mean-square.  相似文献

A method was developed specifically to detect naturally occurring Listeria monocytogenes in meat because the traditional cold enrichment procedure was extremely slow and other procedures were ineffective. This method could identify beta-hemolytic Listeria colonies in 3-4 days. The use of a 2-stage enrichment, highly selective LPM agar, and a thin-layer horse blood agar plate for the detection of beta-hemolytic Listeria isolates are the important steps of this method. L. monocytogenes was recovered from 20 of 41 samples of frozen ground beef, 12 of 23 samples of pork sausage, and 7 of 22 samples of poultry. These results indicate that L. monocytogenes is common in raw meat and that this method is effective for its recovery.  相似文献

Wire-mesh enclosures were used in production ponds to conduct growth trials in which population density and feeding rate were evaluated in a 2 × 2 factorial arrangement for their effects on crawfish growth. Juvenile Procambarus clarkii of uniform size (0.5 g) were stocked at 2 or 20 animals/m2 in experiment 1 and weighed biweekly for 12 wk. Rice forage served as the detrital base, and supplemental feed (25% crude protein) was provided at either a low (26–52 kg/ha per week) or high (104–418 kg/ha per week) rate. In experiment 2, mixed sizes of crawfish were stocked at 10/m2 or 20/m2 and fed the formulated feed at either 52 or 312 kg/ha per week. Feed consumption was estimated for each treatment combination. Only population density significantly affected crawfish growth. Mean weight gain and final weights were inversely proportional to density but only when total crawfish biomass exceeded 1,235 kg/ha. Growth was not significantly influenced by feeding rate, but the high feeding rate resulted in a significantly greater ratio of hepatopancreas weight to body weight and tended to decrease hepatopancreas moisture levels, indicating improved condition. Population density, feeding rate, and their interaction had significant effects on estimated feed consumption. From the simple linear regression of mean feed consumption on crawfish standing crop, it was estimated that crawfish consume about 0.1 kg of dry feed per ha per week per kg of crawfish biomass on a 5 d per week feeding schedule at mean water temperatures, ranging from 14 to 30 C.  相似文献

Aeroz™ (9,10-anthraquinone (AQ)), a chemical rice seed treatment that effectively deters birds from depredating newly planted fields, could be an important management tool that enables rice producers to plant earlier in the season and increase yields. This bird repellent, while non-lethal to birds, has unknown toxicity to crayfish that often are closely integrated with rice production in southern regions of the USA. This study was conducted to assess if AQ, the active ingredient in Aeroz™, could be acutely toxic to the red swamp crayfish Procambarus clarkii (Girard), the crayfish species most often associated with rice cultivation, via seed ingestion and/or through their contact with chemical in pond water. Mortality data from a 96 h feeding study indicate that AQ-treated (1.76% by wt) rice seed was not acutely toxic to juvenile (∼3 mo.) crayfish through the ingestion exposure route. In addition, a 96 h aquatic acute toxicity test showed that the LC50 of AQ in juvenile crayfish is >85 μg L−1 and above the water solubility limit of AQ. These results indicate that very minimal, if any, acute toxicity would be observed in juvenile crayfish if exposed to AQ via ingestion or dissolved or suspended in the water column.  相似文献

High-value phytochemicals could be extracted from biomass prior to the current cellulosic pretreatment technologies (i.e., lime, ammonia, dilute acid, or pressurized hot water treatments) provided that the extraction is performed with a solvent that is compatible with the pretreatment. This work reports on the extraction of flavonoids from Albizia julibrissin biomass. While extracting A. julibrissin foliage with 50 degrees C water, 2.227 mg/g of hyperoside and 8.134 mg/g quercitrin were obtained, which is in the realm of what was obtained with 60% methanol. A. julibrissin foliage, flower, and whole plant extracts were tested in terms of their potential to inhibit low-density lipoprotein (LDL) oxidization. The highest inhibition was obtained with foliage water extracts, which were standardized at 2.5 microM of flavonoids. Also, the 2.5 microM foliage water extract resulted in a reduction from 43% to only 1% of the observed monocyte adherence. To have commercial application, A. julibrissin water extracts should be devoid of toxicity. The A. julibrissin foliage, flower, and whole plant water extracts were not toxic to Vero 76 cells. In summary, A. julibrissin biomass can be extracted with 50 degrees C water to yield an antioxidant stream, showing that it may be possible to couple extraction of valuable phytochemicals to the cellulosic pretreatment step.  相似文献

Conventional heterogeneous dispersion polymerizations of unsaturated monomers are performed in either aqueous or organic dispersing media with the addition of interfacially active agents to stabilize the colloidal dispersion that forms. Successful stabilization of the polymer colloid during polymerization results in the formation of high molar mass polymers with high rates of polymerization. An environmentally responsible alternative to aqueous and organic dispersing media for heterogeneous dispersion polymerizations is described in which supercritical carbon dioxide (CO(2)) is used in conjunction with molecularly engineered free radical initiators and amphipathic molecules that are specifically designed to be interfacially active in CO(2). Conventional lipophilic monomers, exemplified by methyl methacrylate, can be quantitatively (>90 percent) polymerized heterogeneously to very high degrees of polymerization (>3000) in supercritical CO(2) in the presence of an added stabilizer to form kinetically stable dispersions that result in micrometer-sized particles with a narrow size distribution.  相似文献
