Renal effects of the selective alpha(2)-adrenoceptor agonist, medetomidine, were investigated in anesthetized dogs. Animals were administered medetomidine 20 and 40 microg/kg intravenously (IV) and 80 mug/kg intramuscularly (IM) or 1 ml of saline IV. Urine and blood samples were collected before and at 30, 60, 90 and 120 min following medetomidine injection. Mean arterial blood pressure (MABP), renal blood flow (RBF), glomerular filtration rate (GFR), urine volume (U(v)), urine osmolality (U(osm)), free water clearance (C(H2O)), fractional clearance of sodium (F(Na)), plasma osmolality (P(osm)), plasma glucose levels and plasma antidiuretic hormone (ADH) concentrations were measured. The results showed that IV administration of medetomidine initially increased MABP 5-15 min followed by long-lasting decrease. The initial hypertension was not observed after IM administration, which was accompanied by a more profound hypotensive effects. RBF, GFR, U(v), C(H2O) increased after IV injection and decreased after IM. Medetomidine increased FNa and Posm and decreased U(osm). Plasma glucose levels initially increased and subsequently decreased. Plasma ADH concentration was decreased by IV injection but increased by IM administration. Our data imply that: 1) IV administration of medetomidine at dose rates of 20 and 40 microg/kg results in profound diuresis up to 2 hr; 2) Suppression of ADH release from the CNS is one of the mechanisms of medetomidine-induced diuresis although it may not be the principal one.  相似文献

To study the effect of estrogenic chemicals on fish, the gonadosomatic index (GSI = [testis weight/body weight] x 100) and testis histology of mature common carp (Cyprinus carpio) from 2 contaminated sites (Ishizu and Wada rivers, Osaka) and a control site were examined between June 1998 and March 2001. The concentration of nonylphenol, bisphenol A and 17beta-estradiol in the Ishizu river was 3-4 times higher than in the Wada river. In the pre-breeding and breeding seasons, there were no significant differences in body weight among carp from the 3 sites, the body weight of Ishizu river carp being significantly lower (p<0.05) than that of Wada river fish only in the post-breeding season. The GSI and testis weight in fish from the Ishizu river were significantly lower (p<0.05) than in control fish during all phases of gonadal cycle and lower than in Wada river fish in the pre-breeding and post-breeding season. No histological abnormalities were found in the testes of the males examined. Histological observation of the testes revealed a delay in the onset of spermatogenesis in fish from the Ishizu river compared with those from the other sites. These results clearly imply that the estrogenic chemicals in the Ishizu river adversely affect the testis development of the fish.  相似文献

Bisphenol-A (BPA) has been reported to have some xenoestrogenic effects on the reproductive system of male animals. In this study, we examined the growth of combs and testes of the male chickens exposed to BPA. White leghorn male chicks were administered 200 mg BPA orally every week from 2 wk of age. The combs and testes were examined at 16 wk of age. The body growth showed no significant difference between BPA-administered and control birds. However, the weight of the combs and testes were lower in the BPA-treated birds. Histologically, testes of the control birds were well matured; the seminiferous tubuli were filled with sperm. In contrast, the testes of most of the BPA-treated birds showed an immature appearance with smaller seminiferous tubuli and limited spermatogenesis. These findings suggest that the xenoestrogenic property of BPA might disturb the growth of the comb and testes of male chickens by a possible endocrine disrupting mechanism.  相似文献

Fababean (Vicia faba L.) is one of the oldest known important grain legume food crops grown within the temperate and subtropical regions of the world. This species is adapted for both forage and food grain production as a cool season annual crop on a wide range of soil and climatic conditions with effective tripartite symbiosis. Both Rhizobium and endophyte mycorrhiza are essential for high levels of production and symbiotic N2 fixation. The objective of these greenhouse and field studies was to determine effects of Glomus fasciculatum colonization with soil fertility treatments to a Psammentic Paleustalf (Eufaula series) on growth, nodulation, nitrogenase activity and nodule composition for V. faba, var. Major, fuh Rumi (Nile) inoculated with R. leguminosarum Frank.

Top growth and nodule mass were increased approximately 10 fold and nitrogenase activity about 7 fold with the highly significant effect of mycorrhiza and response to low soluble Ca3(PO4)2 fertilization in greenhouse studies. With both effective mycorrhiza and Rhizobium inoculation in the field experiments, seed yields were correlated with top growth (r = 0.841). Phosphorus and Ca fertilization resulted in highly significant increases in seed yields. Nitrogenase activity was correlated with nodule wt. (r = 0.958) and highly significant increases resulted with P and Ca soil amendment. Plant nutrient element composition of nodules increased with the fertilization treatments for P, Ca and increased K levels. Sodium content decreased significantly with increased K fertilization (r = ‐0.846). Potassium composition increased significantly with P content (r = 0.523). Enhanced N2‐fixation along with increased high protein forage and food grain production with Fababeans have much potential. However, soil fertility and management techniques for improved production include effective mycor‐rhizal colonization.  相似文献

This study assessed the immune response of Nile tilapia (Oreochromis niloticus L.) after feeding on different levels (0.1%, 0.2% and 0.4%) of dietary Indian lotus (Nelumbo nucifera) leaf powder for 45 days. We evaluated both the nonspecific immune response at the end of the feeding period and the resistance to Aeromonas hydrophila. Exposure to Indian lotus resulted in a significant elevation in serum total globulins, serum lysozyme activity, serum killing percentage and the phagocytic activity (p < 0.05). Total serum protein and albumin showed no remarkable variation between tilapia fed on 0.1% Indian lotus and the control group (p > 0.05). In addition, the relative expressions of immune‐related genes, namely interleukin–1β and tumour necrosis factor–α were significantly up‐regulated in tilapia fed on 0.4% Indian lotus as compared to the control group; their expressions were down‐regulated in the other tested groups (p < 0.05). The survival rate of Nile tilapia postchallenge to A. hydrophila reported a significant and dose‐dependent increase in the Indian lotus‐supplemented groups (p < 0.05). Therefore, dietary incorporation of Indian lotus leaves (0.4%, 0.2% and 0.1%) could strengthen the immunity of Nile tilapia and improve its resistance to A. hydrophila infection. Therefore, Indian lotus leaves could serve as potential feed supplements for Nile tilapia.  相似文献

Indian lotus, Nelumbo nucifera (Gaertn.) is a valued medicinal plant that exhibits several pharmacological properties. The present work aimed to investigate the effect of Indian lotus as a feed supplement on the growth performance, haematological and biochemical indices, and intestinal histo‐morphology of Nile tilapia (Oreochromis niloticus L.). The fish were randomly distributed into four groups and fed on a basal diet containing Indian lotus leaf powder at different concentrations (0, 0.1, 0.2 and 0.4%) for up to 60 days. The results elucidated that Indian lotus supplemented diets (0.2% and 0.4% followed by 0.1%) significantly improved weight gain, specific growth rate (%) and feed conversion ratio (p < .05). The feed intake was significantly increased in the fish fed on the Indian lotus supplemented diets in a dose‐dependent manner (p < .05). However, Indian lotus had no significant effect (p > .05) on survival rate, total erythrocytes (RBCs) count, haemoglobin (Hb) and hematocrit value (PCV, %) except a significant reduction on Hb content of the fish fed on 0.2% Indian lotus and a significant increase in PCV (%) in the fish fed on 0.1% Indian lotus (p < .05). The total leucocytes (WBCs), neutrophils and lymphocytes counts displayed significant elevations in the fish fed on the Indian lotus diets especially at 0.2% and 0.4%, whereas significant reduction in neutrophils count in the fish fed on 0.1% Indian lotus (p < .05) was observed. The serum glucose was significantly decreased in the fish fed on Indian lotus (0.1% and 0.2%), whereas cholesterol and triglycerides were markedly increased in the fish fed on the Indian lotus supplemented diets (p < .05). Moreover, the intestinal villous heights and the numbers of goblet cells and intraepithelial lymphocytes (IEL) were significantly boosted in all parts of the intestine in all Indian lotus diet groups as compared with the control group. However, as an exception, villous heights in the middle part of the intestine in the fish fed on 0.1% and 0.4% diets (p < .05) were not affected. In the proximal part, the villous heights and the numbers of goblet cells were markedly increased in fish fed on 0.2% and 0.4% diets followed by 0.1% diet. The fish fed on 0.2% lotus supplemented diet followed by 0.1% and 0.4% supplemented diets exhibited significant elevations in villous heights in the distal part and the numbers of goblet cells in the middle part of the intestine. There were no significant differences between the fish fed on the supplemented diets in the numbers of goblet cells in the distal part and the numbers of IEL in the proximal and middle parts (p > .05). In the distal part, the numbers of IEL revealed a significant elevation in 0.4% lotus supplemented diet followed by 0.1% and 0.2% lotus supplemented diet groups (p < .05). Therefore, this study indicates the advantageous effect of the Indian lotus leaves as a natural feed additive for improving growth, intestinal structure and hence, health status of Nile tilapia.  相似文献

Florfenicol (FFC) as a chloramphenicol’s derivative is a special broad-spectrum antibiotic that was used in veterinary clinics. In the present study, we investigated the effect of different doses of FFC on the humoral immune response of broiler chickens to Newcastle disease virus (NDV) vaccine under the impact of E. coli infection. In addition, the expression of the interferon-inducible genes (IRF7, 2′-5′OAS and Mx1) were analyzed in the spleen tissue of these chickens using quantitative real-time PCR (qRT-PCR). The non-treated group with FFC and non-infected with E. coli had the highest immune responses against NDV compared with the FFC treated groups. In the case of E. coli infection, the group treated with FFC (30 mg/Kg BWt) showed lower NDV HI and IgG ELISA Ab levels compared to the group treated with FFC (60 mg/Kg BWt). A dose dependent up-regulation was observed in the level of the interferon-alpha pathway related genes (IRF7 and 2′-5′OAS) in the FFC treated groups compared to the non-treated group. At the slaughter time, the numbers of adipocyte in the bone marrow were significantly higher with moderate atrophy of the hematopoietic lineages in the FFC treated birds compared to the non-treated birds. These results indicated that this FFC dosage dependent increase in the humoral immune responses against NDV vaccine could be attributed to its efficient therapeutic effect on the E. coli infection. However, the increase in the FFC dosage can negatively but temporarily affect the chicken body weights. Additionally, it can exert up regulation effect on the chicken innate immune response with moderate hypoplasia of the bone marrow cells.  相似文献

To understand the detoxification and bioactivation mechanisms for organic contaminants, it is essential to identify the cytochrome P450 (CYP) complement. Therefore, this study aimed to clone a partial cDNA sequence of the novel CYP1D1 gene from the fish Oreochromis niloticus and examine whether intraperitoneal injection of benzo-a-pyrene (BaP), a potent AHR agonist, is capable of inducing CYP1D1 mRNA expression in different tilapia fish tissues. The cloned nucleotide sequence consisted of 713 bp representing a portion of the tilapia CYP1D1 cDNA ORF, encoding 237 amino acids. Amino acid sequence comparison of O. niloticus CYP1D1 with the sequences of CYP1D1 from other species showed that this gene shared the highest identity of 81% with Fundulus heteroclitus CYP1D1. Furthermore, analysis of the percent identities shared by the deduced amino acid sequence of O. niloticus CYP1D1 with the sequences of CYP1 from other species revealed that the highest identities were shared with fish CYP1As. Real-time PCR results revealed that the highest expression level of CYP1D1 mRNA was found in muscles, followed by gills, liver, and intestine, while there was no detectable expression recorded in bile acid. These results indicate that tilapia CYP1D1 plays an important role in the metabolism of xenobiotics, expanding our knowledge regarding the diversity of CYP1 genes in this important model fish species.

Ten populations of Austropotamobius italicus from the Marches region (Central Italy), two populations from the Appennines of Calabria and Basilicata (Southern Italy) and four populations from Friuli Venezia Giulia region were sampled to elucidate the geographical pattern of genetic variability in relation to recent and historical factors.
  • 2. A total of nine COI haplotypes belonging to three separate lineages have been characterized. Both lineage A (Northern Marches region) and C (Friuli Venezia Giulia region) were represented by one haplotype and were related to the Istria 1 peninsula and SouthAlps/Western Balkans lineages respectively. The lineage B included seven haplotypes belonging to the Appenines lineage.

Two hypotheses on the origin of individuals belonging to the Istrian lineage in Marches are discussed: (i) a natural colonization from the Istria peninsula refugia by migrating westwards across the bridge between the Istria peninsula and Italy, (ii) an anthropogenic origin. Based on 16S rRNA sequences, the taxonomic status of lineage B and C, defined from COI mtDNA markers, belongs to A. i. meridionalis and that of lineage A belongs to A. i. carsicus. The existence of a complex geographic structure between Marches populations (Φst=0.66) of A. i. meridionalis should be further considered in conservation management.
