This study aimed to study the characteristics and subpopulations of spermatozoa from bulls with low and high reproductive performance based on pregnancy rates. Based on historical records of pregnancy rate from four farms, 24 bulls were selected. Two groups were established, with low pregnancy rates (n = 12; LOW), including bulls that presented pregnancy rates <52.27% (33.33% to 51.81%); and a group with high pregnancy rates (n = 12; HIGH), with pregnancy rates >52.27% (52.27% to 69.64%), after fixed-time artificial insemination (FTAI). The thawed sperm straws were analysed to sperm kinetics, morphology, plasma membrane integrity and sperm subpopulations. The LOW group exhibited a higher proportion of static cells (p < .05). In contrast, the HIGH group showed greater percentages for membrane integrity and total and progressive motility, and cells with fast and medium velocity (p < .05). In the cluster procedures, four sperm subpopulations were established. The low-fertility bulls presented the highest percentage of subpopulation 2 (41.46%), characterized by slow and progressive spermatozoa. The high-fertility bulls exhibited the highest percentage of subpopulation 3 (37.17%), characterized by fast and nonlinear spermatozoa. Results from this study indicated that bulls with greater percentages of fast and nonlinear spermatozoa seem to have greater fertilization capacity and the subpopulations analysis can be considered a tool to identify ejaculates with high fertility.  相似文献

The Brucella mdh gene was successfully cloned and expressed in E. coli. The purified recombinant malate dehydrogenase protein (rMDH) was reactive to Brucella-positive bovine serum in the early stage, but not reactive in the middle or late stage, and was reactive to Brucella-positive mouse serum in the late stage, but not in the early or middle stage of infection. In addition, rMDH did not react with Brucella-negative bovine or mouse sera. These results suggest that rMDH has the potential for use as a specific antigen in serological diagnosis for early detection of bovine brucellosis.  相似文献

Context

Landscape and local habitat traits moderate wild bee communities. However, whether landscape effects differ between local habitat types is largely unknown.

Objectives

We explored the way that wild bee communities in three distinct habitats are shaped by landscape composition and the availability of flowering plants by evaluating divergences in response patterns between habitats.

Methods

In a large-scale monitoring project across 20 research areas, wild bee data were collected on three habitats: near-natural grassland, established flower plantings and residual habitats (e.g. field margins). Additionally, landscape composition was mapped around the research areas.

Results

Our monitoring produced a dataset of 27,650 bees belonging to 324 species. Bee communities on all three habitats reacted similarly to local flower availability. Intensively managed grassland in the surrounding landscape had an overall negative effect on the studied habitats. Other landscape variables produced diverging response patterns that were particularly pronounced during early and late season. Bee communities in near-natural grassland showed a strong positive response to ruderal areas. Flower plantings and residual habitats such as field margins showed a pronounced positive response to extensively managed grassland and woodland edges. Response patterns regarding bee abundance were consistent with those found for species richness.

Conclusion

We advise the consideration of local habitat type and seasonality when assessing the effect of landscape context on bee communities. A reduction in the intensity of grassland management enhances bee diversity in a broad range of habitats. Moreover, wild bee communities are promoted by habitat types such as ruderal areas or woodland edges.

Carbendazim-resistant (RS) and control (CS) strains ofFusarium sporotrichioides Sherb., previously developed in our laboratory, were exposed to graded concentrations of azoxystrob in in broth media under shake-culture conditions for 2, 3, 4 and 8 days. Azoxystrobin concentrations were 0, 1, 10 and 100 mg 1-1 broth and cultures were incubated at a constant 25°C. Mycelial growth was significantly affected by strain (P<0.01), azoxystrobin concentration (P<0.001) and incubation time (P<0.001). Combined results for the four incubation times showed that CS yielded higher mycelial mass than RS (P<0.01) only in the absence of azoxystrobin. At fungicide additions of 1, 10 and 100 mg P-1 mycelial growth was reduced (P<0.001) with minimal strain differences (P>0.05) at all three doses of azoxystrobin. Significant (P<0.05 or better) strain-fungicide interactions were recorded in trichothecene production following exposure to azoxystrobin. At 4 and 8 days of incubation, the 10 mg 1-1 addition of azoxystrobin stimulated T-2 toxin synthesis (P<0.05) only in RS cultures. In contrast, T-2 toxin enhancement in CS cultures occurred only on day 8 but at a lower level of azoxystrobin (1 mg1-1). Thus, the stimulation of T-2 toxin synthesis depended upon strain and azoxystrobin level. Production of diocetoxyscirpenol (DAS) was affected by a more complex set of interactions. Overall means showed that, in comparison with initial values (on day 2 or 3), DAS output maximized significantly(P<0.05) on day 4 in RS cultures and on day 8 in CS. Marked strain effects were observed on exposure to the 10 mg 1-1 level of azoxystrobin. At this level, DAS production was enhanced in RS only after 4 (P<0.01 ) and 8 (P<0.05) days of incubation, while in contrast, CS reduced DAS production. As with T-2 toxin, DAS production in CS was stimulated (P<0.05 or better) only at low exposure levels of azoxystrobin. In the case of neosolaniol (NEO), however, the main effect of strain was significant (P<0.05), with CS producing consistently more of the mycotoxin than RS on day 4 of the experiment. At this point, the NEO:T-2 toxin ratio was also higher in CS (0.63) than in RS (0.12), a feature reported by us previously. In conclusion, the present investigation has shown for the first time that the development of resistance to one fungicide can affect trichothecene production inF. sporotrichioides on exposure to a second fungicide.
