The hypothesis that an altered expression of CD11/CD18 on bovine circulating monocytes, polymorphonuclear leukocytes (PMN), or both, contributes to an increased mastitis susceptibility in periparturient cows was tested. Expression of CD18 and CD11a, -b, -c on bovine monocytes and PMN were assessed in 8 Friesian-Holstein cows by flow cytometry from 2 wk before calving to 5 wk after calving. Minor changes in adhesion molecule expression levels were detected throughout the experimental period. Compared with PMN, monocytes exhibited an expression level that was similar for CD18, higher for CD11a and CD11c, but lower for CD11b. Differences in density may reflect the relative importance of these adhesion molecules on both leukocyte types. In this study, the decreased number of milk resident macrophages and PMN observed during the periparturient period could not be attributed to changes of CD11/CD18 levels on circulating leukocytes.  相似文献

To determine the plasma pharmacokinetics of suppository acetaminophen (APAP) in healthy dogs and clinically ill dogs. This prospective study used six healthy client‐owned and 20 clinically ill hospitalized dogs. The healthy dogs were randomized by coin flip to receive APAP orally or as a suppository in crossover study design. Blood samples were collected up to 10 hr after APAP dosing. The hospitalized dogs were administered APAP as a suppository, and blood collected at 2 and 6 hr after dosing. Plasma samples were analyzed by ultra‐performance liquid chromatography with triple quadrupole mass spectrometry. In healthy dogs, oral APAP maximal concentration (CMAX=2.69 μg/ml) was reached quickly (TMAX=1.04 hr) and eliminated rapidly (T1/2 = 1.81 hr). Suppository APAP was rapidly, but variably absorbed (CMAX=0.52 μg/ml TMAX=0.67 hr) and eliminated (T1/2 = 3.21 hr). The relative (to oral) fraction of the suppository dose absorbed was 30% (range <1%–67%). In hospitalized ill dogs, the suppository APAP mean plasma concentration at 2 hr and 6 hr was 1.317 μg/ml and 0.283 μg/ml. Nonlinear mixed‐effects modeling did not identify significant covariates affecting variability and was similar to noncompartmental results. Results supported that oral and suppository acetaminophen in healthy and clinical dogs did not reach or sustain concentrations associated with efficacy. Further studies performed on different doses are needed.  相似文献

Although there has been increasing research on the adoption of agroforestry technologies over the last decade, few such studies have assessed uptake over a long period and many are based on a single snapshot in time. Furthermore, most of these studies have mainly looked at non-adopters and adopters: only recently have social scientists considered testers. A further category of users neglected in adoption studies has been re-adopters of technologies. Studying this group provides an interesting and more nuanced understanding of adoption and re-adoption. Methodologically, most adoption studies use quantitative methods and fail to link their findings to wider socio-economic, political and institutional settings. This paper presents a study of the dynamics of improved tree fallow use by farmers in Siaya and Vihiga districts of western Kenya over a period of eight years. It uses both qualitative and quantitative data to critically discuss the motivations of adopters, testers/rejecters and re-adopters. The results show that the process of adoption is highly dynamic and variable with farmers planting improved fallows and discontinuing or re-adopting them due to a whole range of factors, of which soil fertility improvement is just one. These factors included incentives from projects, the tying of adoption to credit programmes, prestige, participation in seminars/tours and the availability of a seed market from projects promoting improved fallows. Farmers planting improved fallows for such reasons may be termed ‘pseudo-adopters’. There were significant differences in adoption between the two districts, with more farmers in Siaya planting improved fallows than in Vihiga. A majority of farmers in Vihiga (53%) who were given seed never planted improved fallows, even though they had been exposed to the technology. Some 40% of farmers in Siaya and 38% in Vihiga planted improved fallows but later rejected them. This has some important implications for research and development. For improved fallow technologies to be attractive to farmers, they must provide other tangible economic benefits besides soil fertility improvement. This presents a challenge to researchers who must better attune themselves to the needs and demands of farmers if they wish to see their research findings widely adopted.  相似文献

Many biotic and abiotic factors influence recovery of soil communities following prolonged disturbance. We investigated the role of soil texture in the recovery of soil microbial community structure and changes in microbial stress, as indexed by phospholipid fatty acid (PLFA) profiles, using two chronosequences of grasslands restored from 0 to 19 years on silty clay loam and loamy fine sand soils in Nebraska, USA. All restorations were formerly cultivated fields seeded to native warm-season grasses through the USDA’s Conservation Reserve Program. Increases in many PLFA concentrations occurred across the silty clay loam chronosequence including total PLFA biomass, richness, fungi, arbuscular mycorrhizal fungi, Gram-positive bacteria, Gram-negative bacteria, and actinomycetes. Ratios of saturated:monounsaturated and iso:anteiso PLFAs decreased across the silty clay loam chronosequence indicating reduction in nutrient stress of the microbial community as grassland established. Multivariate analysis of entire PLFA profiles across the silty clay loam chronosequence showed recovery of microbial community structure on the trajectory toward native prairie. Conversely, no microbial groups exhibited a directional change across the loamy fine sand chronosequence. Changes in soil structure were also only observed across the silty clay loam chronosequence. Aggregate mean weighted diameter (MWD) exhibited an exponential rise to maximum resulting from an exponential rise to maximum in the proportion of large macroaggregates (>2000 μm) and exponential decay in microaggregates (<250 μm and >53 μm) and the silt and clay fraction (<53 μm). Across both chronosequences, MWD was highly correlated with total PLFA biomass and the biomass of many microbial groups. Strong correlations between many PLFA groups and the MWD of aggregates underscore the interdependence between the recovery of soil microbial communities and soil structure that may explain more variation than time for some soils (i.e., loamy fine sand). This study demonstrates that soil microbial responses to grassland restoration are modulated by soil texture with implications for estimating the true capacity of restoration efforts to rehabilitate ecosystem functions.  相似文献

During the Galileo probe's descent through Jupiter's atmosphere, under the ionosphere, the lightning and radio emission detector measured radio frequency signals at levels significantly above the probe's electromagnetic noise. The signal strengths at 3 and 15 kilohertz were relatively large at the beginning of the descent, decreased with depth to a pressure level of about 5 bars, and then increased slowly until the end of the mission. The 15-kilohertz signals show arrival direction anisotropies. Measurements of radio frequency wave forms show that the probe passed through an atmospheric region that did not support lightning within at least 100 kilometers and more likely a few thousand kilometers of the descent trajectory. The apparent opacity of the jovian atmosphere increases sharply at pressures greater than about 4 bars.  相似文献

In gold mining regions, the risk of soil pollution by mercury is a major environmental hazard, especially in tropical areas where soil microflora plays a major part in soil functioning, major bio-geochemical cycles and carbon turn-over. The impact of mercury pollution on soil microflora should thus be carefully assessed in such environments while taking into consideration the specificities of tropical soils. The aim of this study was to compare the effects of mercury (0, 1 and 20 μg of inorganic mercury per gram of soil) on the functional diversity and genetic structure of microbial communities in a tropical soil. We investigated the effects of mercury on tropical soil microflora using soil microcosms spiked with mercury and incubated at 28 °C for 1 month. Microcosm flora, its biomass and its activity, as well as its functional and genetic structure, were followed by cultural methods, measures of respiration, ECOLOG plates, and DGGE (denaturing gel gradient electrophoresis), respectively. Fate of total and bioavailable mercury was estimated by CVAFS (cold vapor atomic fluorescence spectrometry). Results obtained for the microcosms enriched with only 1 μg g?1 mercury were indistinguishable from controls. Conversely, in the presence of high mercury contents (20 μg g(1), an immediate effect was measured on soil respiration, functional diversity (ECOLOG plates) and genetic structure (DGGE), although no significant effect was observed on plate counts or microbial biomass. In addition, whereas microbial activities (respiration and functional diversity) rapidly regained control values, a lasting effect of the high mercury concentration was observed on the genetic structure of the soil microbial community. These modifications took place during the first week of incubation when total mercury concentration was declining and bioavailable mercury was at its highest.This multiple approach study is one of the first attempts at investigating the effects of mercury on soil microbial communities in tropical soils. Our results demonstrate that in the tropical soil under study, mercury affects the soil microbial communities in a different manner than was previously reported in temperate soils. Furthermore, mercury toxicity on soil microbes may be modulated by typical tropical soil characteristics.  相似文献

The cell-mediated immune response of blood lymphocytes from 110 untreated dogs with different tumours was evaluated. The influence of different tumour types on the cellular immune system was examined by assessing the percentage of isolated large granular lymphocytes (LGL), in vitro natural cytotoxicity and mitogen-induced lymphocyte proliferation. Although the overall natural cytotoxicity of dogs with different tumours was decreased, the overall difference from control values was not statistically significant. However, mitogen-induced lymphocyte proliferation was significantly depressed in dogs with tumours in comparison with the controls. Dogs with mammary carcinomas showed significantly lower natural cytotoxicity than controls and dogs with myeloid neoplasms showed significantly lower mitogen-induced lymphocyte proliferation. Abnormalities exist not only in natural cytotoxicity but also in mitogen-induced lymphocyte proliferation. For the dog, this is the first study to assess the influence of different tumours using a combined evaluation of natural cytotoxicity and mitogen-induced lymphocyte proliferation in such a large number of animals.  相似文献

Field strains of Botrytis cinerea Pers ex Fr, the causal agent of grey mould diseases, were collected from French vineyards between 1993 and 2000. Several phenotypes have been characterized according to the inhibitory effects of fungicides towards germ-tube elongation and mycelial growth. Two types of benzimidazole-resistant strains (Ben R1 and Ben R2) could be detected; negative cross-resistance to phenylcarbamates (e.g. diethofencarb) was only found in Ben R1. Benzimidazole resistance was related to point mutations at codon 198 (Ben R1) or 200 (Ben R2) of the beta-tubulin gene. Most dicarboximide-resistant strains were also weakly resistant to aromatic hydrocarbon fungicides (e.g. dicloran) but remained sensitive to phenylpyrroles (e.g. fludioxonil). These resistant field strains (Imi R1) contained a single base pair mutation at position 365 in a two-component histidine kinase gene, probably involved in the fungal osmoregulation. Three anilinopyrimidine-resistant phenotypes have been identified. In the most resistant one (Ani R1), resistance was restricted to anilinopyrimidines, but no differences were observed in the amino-acid sequences of cystathionine beta-lyase (the potential target site of these fungicides) from Ani R1 or wild-type strains. In the two other phenotypes (Ani R2 and Ani R3), resistance extended to various other groups of fungicide, including dicarboximides, phenylpyrroles and sterol biosynthesis inhibitors. This multi-drug resistance was probably determined by over-production of ATP-binding cassette transporters. The hydroxyanilide fenhexamid is a novel botryticide whose primary target site is the 3-keto reductase involved in sterol C-4 demethylations. Apart from the multi-drug-resistant strain Ani R3, three other fenhexamid-resistant phenotypes have been recognized. For two of them (Hyd R1 and Hyd R2) fenhexamid-resistance seemed to result from P450-mediated detoxification. Reduced sensitivity of the target site could be the putative resistance mechanism operating in the third resistant phenotype (Hyd R3). Increased sensitivity to inhibitors of sterol 14 alpha-demethylase recorded in Hyd R1 strains was related to two amino-acid changes at positions 15 and 105 of this enzyme.  相似文献
