The effects of energy level on milk yield, milk composition and growth of the piglets were studied in 20 gilts (10 replicates of two littermates) and their litters during a 21-d lactation. The metabolizable energy (ME) levels were 14.2 and 10.4 Mcal ME X d-1 X sow-1 in the high energy (HE) and low energy (LE) groups, respectively. The daily supply of other nutrients in the diets was identical in both treatments. Milk yield measured every 4 d was lower in the LE group. But fat (8.0 vs 6.9%), energy (1.27 vs 1.14 Mcal/g) and nitrogen (.78 vs .74%) contents of milk were significantly higher in the LE gilts compared with the HE gilts. The daily output of energy and nitrogen was therefore equivalent in both treatments (respectively, 8.28 Mcal and 51.8 g/d), but the fat output was higher in the energy-restricted gilts (532 vs 490 g/d). Growth rate of the litter was similar in both treatments, but piglets suckling LE dams had higher dry matter, fat and energy contents in their body at weaning. Piglets retained 89, 54 and 55% of nitrogen, fat and energy, respectively, of milk between birth and weaning. The results suggest that the ability of sows to mobilize body lipids in order to maintain the output of energy in milk is reduced as body fat reserves are depleted.  相似文献

Bovine herpesvirus 1 glycoprotein D (gD) gene expression by recombinant replication defective human adenovirus type 5 (HAdV-5) was investigated in calves using indirect immunofluorescence microscopy (IIFM), confocal laser scanning microscopy (CLSM) and RT-PCR. One fold intranasal instillation of HAdV-5-expressing gD in the cattle upper respiratory tract showed a short term expression of at least 5 days, but not 10 days, limited only to epithelial cells localised in the epithelium of the nasal mucosa in one out of six calves. Observed limited gene transfer into well differentiated cattle airway epithelial cells must be taken into consideration in order to enhance transfection efficiency, and consequently the vaccine potential of this vector.  相似文献

In order to assess the respective impacts of combinatorial rearrangement, junctional diversification, somatic hypermutation and gene conversion in the generation of immunoglobulin heavy chain variable regions diversity, the sequences of 42 variable regions from late fetal, newborn and young sheep were determined and compared to those of adult animals. At earlier stages of development, the use of germline diversity segments appears restricted, junctional variability is already established, and somatic hypermutations are scarce. The sequence diversity in adults is much higher, which we suggest results from a higher hymermutation activity and possibly from the use of a variety of diversity segments. Altogether, this pattern is very reminiscent of the situation observed in cattle, except for the length of the third complementarity determining regions (CDR3) which are shorter in sheep than in bovine. Unlike the chicken and rabbit systems, it seems that new rearrangements continue to occur in sheep for at least several months after birth.  相似文献

Background

Early recognition of left‐sided congestive heart failure (CHF) in dogs with degenerative mitral valve disease (DMVD) is important because it influences medical therapy, timing of follow‐up, and outcome.

Hypothesis

Pulmonary vein diameter‐to‐pulmonary artery diameter ratio (PV/PA) measured by echocardiography can predict CHF.

Animals

Ninety‐eight client‐owned dogs, 37 controls, and 61 dogs with DMVD.

Methods

Prospective clinical cohort study. History, physical examination and Doppler‐echocardiography were performed. Dogs were classified as International Small Animal Cardiac Health Council class I, II or III. Congestive heart failure was identified in a subset of 56 dogs based on radiographic findings. The PV/PA was measured in bidimensional (2D) and M‐mode by 2 investigators blinded to the radiologists’ conclusions.

Results

Interobserver coefficients of variation for PV/PA acquisition and measurement were <10%. The PV/PA in control dogs was approximately 1 and increased with class of heart failure. The presence of CHF could be best predicted by measuring PV/PA in 2D echocardiography (cut‐off, 1.7; area under the curve, 0.98; CI, 0.97–0.98; P < .001) with a sensitivity of 96% and a specificity of 91%.

Conclusion and clinical importance

The PV/PA is a simple and reproducible echocardiographic variable that increases with class of heart failure and may help discriminate dogs in CHF from asymptomatic dogs with DMVD. Additional studies are required to determine whether PV/PA might provide additional information in the integrated interpretation of Doppler‐echocardiographic indices of left ventricular filling pressures and could be used for rapid assessment of CHF in dogs in a critical care setting.  相似文献

This experiment was undertaken to determine whether pregnancy affects glucose metabolism and insulin sensitivity in sows fed at different levels. Four replicates of six multiparous Large White sows were involved. In each replicate, four sows were inseminated on the first postweaning estrus (pregnant group) and the two remaining were kept nonpregnant. Half of the sows of each group were fed 2.5 kg/d (low level) and the others 4 kg/d (high level) of the same standard pregnancy diet. Jugular catheters were implanted 2 to 3 d after estrus. Plasma concentrations of glucose, insulin, and FFA were determined before and during the 4 h following the morning meal at 10, 30, 59, 87, 93, 101, and 110 d of gestation and at equivalent periods for the nonpregnant sows. Intravenous glucose tolerance tests were performed at 33, 71, 85, 96, and 108 d by i.v. injection of 0.5 g glucose/kg BW. Compared with the glycemia before the meal, all the sows showed hyperglycemia 30 min after the initiation of the meal and hypoglycemia thereafter, with a minimum reached at approximately 75 min. Insulinemia increased from 20 min after food access, reached a maximum at 40 min, and returned to the basal level after 180 min. The higher feeding level increased plasma insulin and lowered plasma glucose levels. Glycemia and insulinemia profiles changed from 87 d onward in the pregnant sows. The peak of glucose induced by the meal was higher, and the subsequent period of hypoglycemia almost disappeared. The area under the insulin curve was unchanged, but insulin secretion was delayed. The glucose tolerance tests showed that between d 85 and 108 the half-life of injected glucose increased and insulin secretion was delayed in the pregnant sows. Compared to the following stages, plasma FFA were high before and after the meal at 10 d, which most likely was a residual effect from the previous gestation/lactation cycle. They were lower from 30 to 101 d in the pregnant and nonpregnant sows. At 110 d, fasting FFA were high again in the pregnant sows only, very likely in relation to the preparation for lactation. This experiment showed that insulin sensitivity decreases after 85 d of pregnancy in multiparous sows.  相似文献

This study on primiparous sows was designed to 1) determine the impact of nursing a large litter on LH secretion and follicular development, and 2) investigate the metabolic adaptations by which milk yield increases with litter size. At farrowing, crossbred, primiparous sows were assigned to 1 of 3 experimental groups differing in litter size and feed allowance. Sows with 13 or 14 piglets (13AL, n = 7) were fed ad libitum. Sows with 7 piglets were fed ad libitum (7AL, n = 6) or were feed-restricted (7R, n = 8). The restriction was based on the estimated energy deficiency for the 13AL sows. On d 9 +/- 1 of lactation, a jugular catheter was surgically implanted. Serial blood samplings and glucose tolerance tests were performed in mid- and late lactation. Sows were slaughtered 3 d after weaning, and ovarian characteristics were recorded. During lactation, the 7AL sows lost no or little body reserves, and their estimated energy balance was near zero. The 13AL and 7R sows exhibited similar negative energy balances and similar losses of backfat and estimated lipid content. Litter growth rate was greater (P < 0.05) in the 13AL than in the 7AL and 7R groups. After weaning, the volume of the largest 14 follicles was smaller (P < 0.05) in sows nursing 13 or 14 piglets than in sows with 7 piglets. Plasma concentrations of LH and LH pulse frequency did not differ between groups (P > 0.1). The longer glucose half-life on d 16 than on d 27 of lactation (22.5 vs. 18.8 min; P < 0.05) indicated a lower glucose tolerance in mid- than in late lactation. The area under the insulin curve was greater in the 7AL than in the 13AL sows (P = 0.08) and intermediate in the 7R group, with no differences in glucose profiles. This led to the suggestion that the 7AL sows were more resistant to insulin than the 13AL sows. In all groups of sows, follicular development after weaning was correlated with LH secretion in midlactation. Active follicular development was associated with prolonged secretion of insulin in response to glucose challenge. Our results show that besides litter size, a sow's metabolic status in lactation influences follicular maturation after weaning and also indicate that the metabolic adaptations by which primiparous sows nursing large litters increase litter growth rate and body reserve mobilization do not involve an accentuated peripheral insulin resistance.  相似文献

ABSTRACT: In the rapidly changing context of research on animal health, INRA launched a collective discussion on the challenges facing the field, its distinguishing features, and synergies with biomedical research. As has been declared forcibly by the heads of WHO, FAO and OIE, the challenges facing animal health, beyond diseases transmissible to humans, are critically important and involve food security, agriculture economics, and the ensemble of economic activities associated with agriculture. There are in addition issues related to public health (zoonoses, xenobiotics, antimicrobial resistance), the environment, and animal welfare.Animal health research is distinguished by particular methodologies and scientific questions that stem from the specific biological features of domestic species and from animal husbandry practices. It generally does not explore the same scientific questions as research on human biology, even when the same pathogens are being studied, and the discipline is rooted in a very specific agricultural and economic context.Generic and methodological synergies nevertheless exist with biomedical research, particularly with regard to tools and biological models. Certain domestic species furthermore present more functional similarities with humans than laboratory rodents.The singularity of animal health research in relation to biomedical research should be taken into account in the organization, evaluation, and funding of the field through a policy that clearly recognizes the specific issues at stake. At the same time, the One Health approach should facilitate closer collaboration between biomedical and animal health research at the level of research teams and programmes.  相似文献
