This study sought to isolate and characterize lactic acid bacteria (LAB) from stomach of adult snakehead fish, Channa striatus, to be used as probiotics for freshwater fish. A total of 13 strains were isolated from the stomach of 10 fish, and 4 of these belonged to LAB. Strain LAB‐3 showing highest in vitro growth inhibition of Aeromonas hydrophila in a disk diffusion test was identified as Lactobacillus fermentum by conventional and molecular techniques and evaluated in vitro through various tests. The bacterium could grow at pH 3–8; but the optimum growth was observed at pH 6. Moreover, LAB‐3 grew at 0.15 and 0.3% bile salt concentrations, from 15 to 45 C, and at 4% NaCl. L. fermentum showed in vitro inhibitory activity against three fish pathogens, A. hydrophila, Pseudomonas aeruginosa, and Shewanella putrefaciens, tested by disk diffusion and well diffusion methods. Antibiotic sensitivity tests indicated that L. fermentum was resistant to streptomycin, gentamycin, and kanamycin, intermediate to tetracycline, but sensitive to chloramphenicol, amoxicillin, and ampicillin. Challenge test by using A. hydrophila showed that survival of snakehead was significantly (P < 0.05) improved when 2 × 106 LAB‐3/g was supplemented to the diet. Therefore, this study suggests that L. fermentum might be a promising probiotic in snakehead aquaculture .  相似文献

In China, the risk of cotton production on arable land contaminated with heavy metals has increased in recent years. Compared with other major crops, cotton shows stronger and widespread resistance to abiotic stresses such as heavy metals. As the main product of cotton is non-edible fibers, and cotton possesses a set of sophisticated stress-resistance strategies, cotton is an ideal crop for use in phytoremediation of soil contaminated by heavy metals. We present a global review of recent literature on the toxic effects of five heavy metals (cadmium, chromium, lead, arsenic, and mercury) at present in China and their effects on cotton, as well as summarize recent studies on the mechanisms of avoidance and avoidance both. We aim to provide insights into understanding the mechanisms of resistance to heavy metal stress in cottonunderstanding the relationship between cotton and heavy metal stress.  相似文献

On 26 December 2004, a tsunami caused extensive loss of life, damaged property and degraded agricultural land in the province of Aceh, Indonesia. While some of the associated soil chemical changes have been documented, information on soil physical properties is sparse. The objective of this study was to quantify physical properties of some tsunami-affected upland agricultural soils in Aceh, Indonesia. Soil was sampled approximately 21/2 years after the tsunami, from the 0–0.1 m, 0.1–0.3 m and 0.3–0.5 m depths in four sites in the villages of Kling Cot Aroun in Aceh Besar sub-district, Kuta Kruen in Aceh Utara sub-district, Udjong Blang Mesjid in Bireuen sub-district and Meue in Pidie Jaya sub-district on the east coast of Aceh. These sites were located within 1 km from the sea at elevations ranging from 0 to 5 m ASL. The soils were Ultisols except for Meue, which was an Entisol. Soil properties measured were bulk density, structural stability and particle size distribution. Soil water retention, pore-size distribution and saturated hydraulic conductivity were estimated by inserting the values of bulk density, clay, sand and silt contents into pedotransfer functions from the literature. The analyses conducted during this study did not permit us to ascertain what proportion of the soil particles were of tsunami-origin. Nonetheless, deposition of finer-textured material may have occurred in two of the sites. In comparison with the greyish-white, coarse textured soil in the rest of the profile, a finer-textured yellow horizon was present in the lower slopes of the Udjong Blang Mesjid site. At Meue, clay and silt contents were higher in the surface 0.3 m than in the 0.3–0.5 m depth, although a distinct horizon was absent. Particle size distribution in all sites was dominated by the sand fraction, although clay and silt contents were relatively high (20–30 g 100 g− 1) at Kuta Kruen. Among the sand fractions, fine sand (0.02–0.25 mm) was highest at Kling Cot Aroun, Kuta Kruen and in the “yellow horizon” at Udjong Blang Mesjid, making them more prone to hardsetting and compaction after intensive tillage. Soil compaction was present in all sites with that in the “yellow horizon” at Udjong Blang Mesjid being highest. The relatively low porosity in this layer may be beneficial, as it is likely to reduce the high rates of water drainage and nutrient leaching in this sandy soil. The more compacted soils were characterised by higher numbers of micropores (r, pore radius < 4.3 μm), lower water retention at saturation, smaller numbers of macropores (r > 14.3 μm), lower hydraulic conductivity and intensive gleying, indicating frequent waterlogging. The soils in all depths from Kling Cot Aroun and the “yellow horizon” at Udjong Blang Mesjid were very dispersive, that at Meue moderately dispersive in the 0.3–0.5 m depth but stable in the 0–0.1 m depth, and at Kuta Kruen very stable in all depths. Soil physical degradation was a feature of the soils examined, and its amelioration will be the key to improving and sustaining crop yields in these soils. Possible management interventions include organic amendments such as compost or manure, and minimum tillage options such permanent beds or zero tillage with retention of crop residues as in situ mulch together with suitable cover crops.  相似文献

Nowadays, many researches were conducted in minimizing tissue culture technology due to the overhead of cost needed. The purpose of this study was to investigate the effects of using five kinds of organic additives at four level concentrations responsive to the number of shoots produced for eight weeks in culture. Stem segment explants of Celosia sp. were cultured on MS medium that have been supplemented with different kinds of extract juice that serve as organic additives which are mature coconut, young coconut, papaya, banana and tomato at 20, 30, 50 and 70 ml L-1. The numbers of shoot on each explant were recorded and the mean of ten replicates explants were calculated. Among the media used, young coconut water at 70 ml L1- induced the highest shoot regeneration (14.21+/-8.26), followed by mature coconut water at 50 ml L-1 (13.14+/-10.33). Banana and tomato juice promote highest shoot regeneration of stem segments at 50 ml L-1 that produced 9.57+/-4.68 and 9.28+/-5.82 shoots per explants, respectively. While the lowest concentration which at 20 ml L-1 of papaya juice showed highest shoot regeneration (10.50+/-3.45) produced among the three other concentration tested. Statistical results showed that there were significant differences interactions effects (p<0.05) in terms of number of shoot regenerated between the types of extracts juices determined by ANOVA test. Comparing number of shoots regenerated that were cultured in control media, it showed higher than all of experimental medium composition. There were no big different in cost required in preparation of control media and the experimental media. Applications of five kinds of local fruit in tissue culture media should be considered since it responsive in shoot regeneration.  相似文献

Efficient plant regeneration of Saintpaulia ionantha (African violet) has been obtained in the present study. MS medium supplemented with 1.0 mg L(-1) IAA and 2.0 mg L(-1) Zeatin resulted in 100% shoot regeneration and induced the highest number of shoots (average 15.0 +/- 0.8 shoots per explant) after being cultured for 8 weeks. The above hormone combination was optimum for shoot regeneration. Most of Saintpaulia ionantha plantlets derived from tissue culture system could be hardened and transferred to the greenhouse conditions with 84.0 +/- 1.6% success rate. However, regenerated plantlets of Saintpaulia ionantha (even after 12-months-old) failed to flower. Morphological characters of regenerated plantlets of Saintpaulia ionantha were observed and compared with in vivo (intact) plants. Regenerated plantlets showed some differences in morphological characters, such as height and leaf size, texture and colour, but the plantlets showed no variation in leaf arrangement and leaf margin. However, the morphological characters of the regenerated plantlets were found to be unstable.  相似文献

The study assessed the effect of hybridization on reproductive performance of Oreochromis karongae, Oreochromis shiranus and Oreochromis mossambicus for a period of 120 days. Number of eggs per batch, number of females spawned, relative fecundity, egg size, hatching period, hatchability and hatchling survival were assessed. The results revealed higher number of eggs per batch and relative fecundity in the crosses where O. shiranus and O. mossambicus were females than female O. karongae. The results also revealed that egg size, hatching period, hatchability and hatchling survival were species specific. Significant differences were noted for number of eggs and larvae produced by the three species (p < .05). Interspecific and pure crosses for the same female species did not differ significantly for the number of eggs per batch, relative fecundity, egg size, hatching period, hatchability and hatchling survival. However, number of female spawned was found to be higher in the interspecific crosses of female O. karongae than in pure cross of O. karongae. Findings from this study suggest that hybridization can be used to improve reproductive performance of O. karongae as the females spawned increased, and it is a value addition in aquaculture management of indigenous species.  相似文献
