Butyrolactone I (BTL-I) is a butanolide isolated from the deep-sea-derived fungus, Aspergillus sp. It provides a potential new target for the prevention and treatment of food allergies. This study aimed to investigate the metabolic and pharmacokinetic profile of BTL-I in rats. The metabolic profiles were obtained by UHPLC–Q-TOF-MS. As a result, eleven metabolites were structurally identified, and the proposed metabolic pathways of BTL-I were characterized. The main metabolites were the oxidative and glucuronidative metabolites. In addition, a sensitive UHPLC–MS/MS method was established for the quantitation of BTL-I in rat plasma (LOQ = 2 ng/mL). The method was fully validated and successfully applied to the pharmacokinetic study of BTL-I in rats after oral administration or intravenous administration. The oral bioavailability was calculated as 6.29%, and the maximum plasma concentrations were 9.85 ± 1.54 ng/mL and 17.97 ± 1.36 ng/mL for intravenous and intragastric dosing groups, respectively.  相似文献

稻田系统生产力及其稳定性对施肥制度的响应   总被引:1,自引:0,他引:1  
为探讨长期不同施肥制度对稻田系统生产力及其稳定性的影响,以国家生态系统研究网络中国科学院桃源农业生态试验站稻田施肥试验(2004—2010年)监测数据为基础,分析不同施肥制度下红壤性稻田系统生产力及其稳定性。结果表明,施肥是维持和提升稻田系统生产力的物质基础,稻田系统生产力稳定性对不同施肥制度响应不同:(1)各施肥处理总生物量生产力显著高于不施肥处理,是不施肥处理的1.33~2.23倍,从高至低依次为:优化施肥处理>常规施肥处理>化肥处理>不施肥处理,且优化施肥能显著提高总生物量生产力的稳定性(P<0.05);(2)各施肥处理的水稻产量均显著高于不施肥处理,是不施肥处理的1.35~2.02倍,年际变异系数分别为:不施肥处理22.4%、化肥处理15.8%、优化施肥处理10.3%、常规施肥处理15.7%,优化施肥处理产量稳定性最高;(3)地下部分生产力施肥处理显著高于不施肥处理,施肥年限较长的处理地下部分生物量生产力的稳定性显著高于年限较短的施肥处理(P<0.05)。总的来说,优化施肥处理水稻生产力及其稳定性较好,不施肥处理水稻生产力及其稳定性最差;地上部分生产力及其稳定性和地下部分生产力受施肥制度的影响较大,而地下部分生产力的稳定性则受施肥年限影响较大。  相似文献

A experiment was carried to evaluate the effects of Al on growth, accumulations of free proline and amino acid in 2 wheat cultivars (Triticum aestivum L.), Yangmai No.5 and Jian 864, differing in Al sensitivity. Plants grew initially in a nutrient solution without Al for 13 days before the addition of Al and finally in a nutrient solution containing 0.5mmol Al (L-1) for 19 days. The results showed that there were marked decreases in dry weight, relative growth rate (RGR) and net assimilation rate (NAR) of Al-treated seedlings compared with control plants. The Al effects were more evident in Yangmai No.5 than Jian 864. Leaf area ratio(LAR) was little affected by Al. RGR was highly correlated with NAR rather than LAR. Aluminum increased the concentrations of free proline and total free amino acid in shoots of both the cultivars. The increases were greater in Yangmai No.5 than in Jian 864. The percentage of free proline in total amino acid in shoots was not affected by Al treatment. It was possible that accumulation of proline was merely a symptom of Al injury. The concentrations of total nitrogen in Al-treated plants did not significantly differ from those of control plants. Nitrate reductase activity (NRA) in leaves was severely decreased by Al, and a greater decrease was noted in Yangmai No.5 than in Jian 864, but NRA in roots of both the cultivars was not affected. The decreases in NRA might be an indirect (accumulation of amino acid) rather than a direct result of Al toxicity.  相似文献

华北平原小麦-玉米农田生态系统服务评价   总被引:8,自引:1,他引:7  
本研究于2006 年和2007 年在中国科学院栾城农业生态系统试验站田间试验基础上, 评价了华北平原小麦-玉米农田的初级产品生产、气体调节、土壤有机质累积、水调节和氮素转化等5 项生态系统服务。研究表明, 华北平原小麦-玉米农田初级产品量包括籽粒产量5.04~5.71 t·hm-2·a-1(小麦)和6.69~8.24t·hm-2·a-1(玉米), 秸秆量8.58~9.72 t·hm-2·a-1(小麦)和6.97~8.58 t·hm-2·a-1 (玉米); 农田气体调节包括释放O2 24.99~28.64 t·hm-2·a-1, 固定CO2 34.23~39.22 t·hm-2·a-1, 排放N2O 0.72~1.13 kg·hm-2·a-1, 吸收CH4 3.39~5.70 kg·hm-2·a-1; 农田耕层土壤有机质累积量为1.13~2.39 t·hm-2·a-1; 水资源消耗量为2 890~3 830 m3·hm-2·a-1; 农田土壤氮素几乎都处于亏缺状态, 变化范围为-107.73~5.33 kg(N)·hm-2·a-1, 不施氮肥农田亏缺较多。综合评价发现, 小麦-玉米农田提供生态服务的经济价值为5.48~6.25 万元·hm-2·a-1, 是粮食生产价值的3 倍左右。氮肥施用对农田生态系统服务及其产生福利的影响较为复杂, 这主要是由于施加氮肥明显增加了氮素转化功能导致的经济损失, 而同时可能会增加初级产品生产、气体调节中作物固定CO2 和释放O2 功能的经济价值。尽管目前有关生态系统服务评价研究主要关注生态系统产生的正效应, 但仍有必要对农田产生的负效应做出评价, 以便客观看待农田生态系统价值, 正确认识农田生态系统对人类福利的影响。  相似文献

Ten new (1–10) and 26 known (11–36) compounds were isolated from Penicillium griseofulvum MCCC 3A00225, a deep sea-derived fungus. The structures of the new compounds were determined by detailed analysis of the NMR and HRESIMS spectroscopic data. The absolute configurations were established by X-ray crystallography, Marfey’s method, and the ICD method. All isolates were tested for in vitro anti-food allergic bioactivities in immunoglobulin (Ig) E-mediated rat basophilic leukemia (RBL)-2H3 cells. Compound 13 significantly decreased the degranulation release with an IC50 value of 60.3 μM, compared to that of 91.6 μM of the positive control, loratadine.  相似文献

Early acute kidney injury (AKI) is a devastating complication in critical burn patients, and it is associated with severe morbidity and mortality. The mechanism of AKI is multifactorial. Astaxanthin (ATX) is a natural compound that is widely distributed in marine organisms; it is a strong antioxidant and exhibits other biological effects that have been well studied in various traumatic injuries and diseases. Hence, we attempted to explore the potential protection of ATX against early post burn AKI and its possible mechanisms of action. The classic severe burn rat model was utilized for the histological and biochemical assessments of the therapeutic value and mechanisms of action of ATX. Upon ATX treatment, renal tubular injury and the levels of serum creatinine and neutrophil gelatinase-associated lipocalin were improved. Furthermore, relief of oxidative stress and tubular apoptosis in rat kidneys post burn was also observed. Additionally, ATX administration increased Akt and Bad phosphorylation and further down-regulated the expression of other downstream pro-apoptotic proteins (cytochrome c and caspase-3/9); these effects were reversed by the PI3K inhibitor LY294002. Moreover, the protective effect of ATX presents a dose-dependent enhancement. The data above suggested that ATX protects against early AKI following severe burns in rats, which was attributed to its ability to ameliorate oxidative stress and inhibit apoptosis by modulating the mitochondrial-apoptotic pathway, regarded as the Akt/Bad/Caspases signalling cascade.  相似文献
