Prunus scoparia is a wild deciduous shrub, usually living on dry calcareous soils of the rocky mountains and has been used as a grafting rootstock for domesticated almonds to provide drought resistance. In the current study, micropropagation ability of P. scoparia was investigated using cytokinin and auxin. Uniform nodal shoot pieces (3–5 cm in length) of seedlings were used as explants. The explants were disinfected with 10% sodium hypochlorite solution. For adventitious shoot induction and proliferation, Murashige and Skoog (MS) media containing 7.00 g/l agar and 30.00 g/l sucrose containing five concentrations of benzyl adenine (BA) (0.00, 0.50, 1.00, 2.00, and 4.00 mg/1) and also containing six concentrations of Thidiazuron (TDZ) (0.00, 0.50, 1.00, 2.00, 5.00, and 7.00 mg/1) were compared. For rooting, in vitro shoots (2–3 cm) were transferred into ½ MS medium supplemented with 30 g/l sucrose, 7.50 g/l agar, and different concentrations of IBA (0.00, 0.25, 0.50, and 1.00 mg/l) and NAA (0.00, 0.25, 0.50, and 1.00 mg/l). Based on the results obtained for shoot proliferation, only 2.00 and 4.00 mg/l BA and 2.00 mg/l TDZ concentrations generated shoots, while other treatments did not show shoot proliferation. Among the three treatments that generated shoots, the best results for shoot number, leaf number, and leaf color quality were observed in media containing 2.00 mg/l TDZ. Based on the results obtained for rooting, the effect of IBA concentrations on the rooting percentage, root number, and root length was significant. Among IBA concentrations, only 0.50 mg/l IBA induced rooting, while there was no rooting in the media containing other IBA concentrations. None of the NAA concentrations showed rooting. In conclusion, MS culture medium supplemented with 2.00 mg/l TDZ and ½ MS culture medium supplemented with 0.50 mg/l IBA are suggested for in vitro shoot proliferation and rooting of P. scoparia, respectively. A comparison was made of the response to storage temperatures (0–17°C) of grapefruit ‘Shamouti’ and ‘Valencia’ oranges, and lemon fruit. There was no chilling injury (CI) and the incidence of rot was relatively low at 12°C or higher for all cultivars. At low storage temperatures, however, pitting and subsequent rot development in grapefruit and in ‘Shamouti’ and ‘Valencia’ oranges were highest at 6°C, while in lemon the incidence of these disorders rose with the decrease in storage temperature, and was highest at 2°C. Grapefruit and ‘Shamouti’ oranges exhibited the highest susceptibility to CI, lemon showed an intermediate susceptibility, and ‘Valencia’ orange a very low one. In grapefruit, severe peel pittings that developed in cold storage were covered by mold rots during cold storage and shelf-life, but in ‘Valencia’ and particularly in ‘Shamouti’ oranges the incidence of decay during cold storage was low. During shelf-life, a pronounced CI-induced increase in the incidence of mold rots was evident. This breakdown during shelf-life may be associated with microscopic peel injuries which are formed during cold storage and covered by mold rots during shelf-life. Increased susceptibility of the cultivar to CI could be correlated with an increase in ethanol content at low temperatures. 相似文献

ABSTRACTBamboo scrimber is one of the most emerging structural materials for future building applications and it possesses properties comparable to other natural wood-based engineered materials such as glulam, laminated veneer lumber and cross-laminated timber. The goal of this work was to study the decay resistance of bamboo scrimber against white-rot (Trametes versicolor) and brown-rot fungi (Serpula lacrymans). Bamboo scrimber samples were incubated in petri dishes with the wood-decaying fungi and the weight loss after 12 weeks was measured. The surface morphology of fungal-degraded bamboo scrimber was evaluated using optical microscopy. Based on the percentage weight loss, bamboo scrimber could be classified as highly resistant against bio-deterioration by white and brown-rot fungi. 相似文献

This study was aimed at construction of reference (physiological) value intervals determined by lower 2.5% and upper 97.5% quantiles for total plasma protein (TPP) in farmed rainbow trout, Oncorhychus mykiss (Kamloops strain), aged 10–12 months (n=755). Total plasma protein in male trout from raceway culture was found to be significantly greater (38–58.3 g L?1) relative to females (31–51.5 g L?1). Total plasma protein in trout from cage culture was significantly greater (32–54 g L?1) compared with the raceway culture (31–49 g L?1). Moderate product–moment correlations between TPP and cholesterol (r=0.8730), TPP and total calcium (r=0.8188), TPP and alanine aminotransferase (r=0.6200) and between TPP and aspartate aminotransferase (r=0.6430) were estimated. Technologies to assess the molecular targets of biomolecules in living cells are lacking. We have developed a technology called magnetism-based interaction capture (MAGIC) that identifies molecular targets on the basis of induced movement of superparamagnetic nanoparticles inside living cells. Efficient intracellular uptake of superparamagnetic nanoparticles (coated with a small molecule of interest) was mediated by a transducible fusogenic peptide. These nanoprobes captured the small molecule's labeled target protein and were translocated in a direction specified by the magnetic field. Use of MAGIC in genome-wide expression screening identified multiple protein targets of a drug. MAGIC was also used to monitor signal-dependent modification and multiple interactions of proteins. 相似文献

For prolonged storage of lemons, the best temperature to avoid rots and physiological disorders such as internal browning and peel pitting, was 14°C; for short periods only, storage at lower temperatures, 2–8°C, did not cause damage. Physiological disorders appeared in storage earlier, the lower the storage temperatures, and in fruit picked at a less ripe stage. The general appearance of the fruit is better when stored at the lower temperature. Three strains of mice, BALB/c, IL-12 knock-out (KO) and INF-gamma knock-out, were chosen as an experimental model for the study of intestinal immunity induction against Encephalitozoon cuniculi Levaditi, Nicolau et Schoen, 1923 infection. Mice were infected perorally with 10(7) spores and re-infected with the same dose 70 days after the first infection. The anti-E. cuniculi IgA, IgG and IgM responses in sera and extracts of stool samples were determined by ELISA. Results have shown specific antibody production in the sera and intestinal secretions of all three strains of mice induced orally by E. ciniculi spores. BALB/c mice developed a stronger humoral immune response than IL-12 KO mice. The lowest antibody response developed in INF-gamma KO mice that succumbed to the infection within 28 days post infection. 相似文献

The Labyrinthulomycetes or Labyrinthulea are a class of protists that produce a network of filaments that enable the cells to glide along and absorb nutrients. One of the main two Labyrinthulea groups is the thraustochytrids, which are becoming an increasingly recognised and commercially used alternate source of long-chain (LC, ≥C20) omega-3 containing oils. This study demonstrates, to our knowledge for the first time, the regiospecificity of the triacylglycerol (TAG) fraction derived from Australian thraustochytrid Aurantiochytrium sp. strain TC 20 obtained using 13C nuclear magnetic resonance spectroscopy (13C NMR) analysis. The DHA present in the TC 20 TAG fraction was determined to be concentrated in the sn-2 position, with TAG (16:0/22:6/16:0) identified as the main species present. The sn-2 preference is similar to that found in salmon and tuna oil, and differs to seal oil containing largely sn-1,3 LC-PUFA. A higher concentration of sn-2 DHA occurred in the thraustochytrid TC 20 oil compared to that of tuna oil. 相似文献
