Rotaviruses (RV) have a high prevalence in piggeries worldwide and are one of the major pathogens causing severe diarrhoea in young pigs. RV species A, B, and C have been linked to piglet diarrhoea in Australian pig herds, but their genetic diversity has not been studied in detail. Based on sequencing of the structural viral protein 7 (VP7) RVA G genotypes G3, G4 and G5, and RVC types G1, G3, G5, and G6 have been identified in Australian piggeries in previous studies. Although occurrence of RVB was reported in Australia in 1988, no further genetic analysis has been conducted. To improve health management decisions in Australian pig herds, more information on RV prevalence and genetic diversity is needed. Here, 243 enteric samples collected from 20 pig farms within Eastern Australia were analysed for the presence of RV in different age groups using a novel PCR-based multiplex assay (Pork MultiPath™ enteric panel). RVA, RVB, and RVC were detected in 10, 14, and 14 farms, respectively. Further sequencing of VP7 in selected RV-positive samples revealed G genotypes G2, G5, G9 (RVA), G6, G8, G14, G16, G20 (RVB), and G1, G3, G5, G6 (RVC) present. RVA was only detected in young (<10 weeks old) pigs whereas RVB and RVC were also detected in older animals (>11 weeks old). Interestingly, RVB and RVC G-type occurrence differed between age groups. In conclusion, this study provides new insights on the prevalence and diversity of different RV species in pig herds of Eastern Australia whilst demonstrating the ability of the Pork MultiPath™ technology to accurately differentiate between these RV species.  相似文献

Stripe rust is a continuous threat to wheat crop all over the world. It causes considerable yield losses in wheat crop every year. Continuous deployment of adult plant resistance(APR) genes in newly developing wheat cultivars is the most judicious strategy to combat this disease. Herein, we dissected the genetics underpinning stripe rust resistance in Pakistani wheat germplasm. An association panel of 94 spring wheat genotypes was phenotyped for two years to score the infestation of stripe rust on each accession and was scanned with 203 polymorphic SSRs. Based on D' measure, linkage disequilibrium(LD) exhibited between loci distant up to 45 c M. Marker-trait associations(MTAs) were determined using mixed linear model(MLM). Total 31 quantitative trait loci(QTLs) were observed on all 21 wheat chromosomes. Twelve QTLs were newly discovered as well as 19 QTLs and 35 previously reported Yr genes were validated in Pakistani wheat germplasm. The major QTLs were QYr.uaf.2 AL and QYr.uaf.3 BS(PVE, 11.9%). Dissection of genes from the newly observed QTLs can provide new APR genes to improve genetic resources for APR resistance in wheat crop.  相似文献

The deleterious effects of ethanol on the developing human brain are poorly understood. Here it is reported that ethanol, acting by a dual mechanism [blockade of N-methyl-D-aspartate (NMDA) glutamate receptors and excessive activation of GABA(A) receptors], triggers widespread apoptotic neurodegeneration in the developing rat forebrain. Vulnerability coincides with the period of synaptogenesis, which in humans extends from the sixth month of gestation to several years after birth. During this period, transient ethanol exposure can delete millions of neurons from the developing brain. This can explain the reduced brain mass and neurobehavioral disturbances associated with human fetal alcohol syndrome.  相似文献

Plant growth regulators are biologically active signaling molecules that regulate a number of plant physiological processes. Auxin (indole-3-acetic acid) is an important plant growth regulator and is synthesized within plant tissues through L-tryptophan (L-TRP)-dependent and -independent pathways. It has been found that plants respond to exogenously applied L-TRP due to insufficient endogenous auxin biosynthesis. The exogenous application of L-TRP is highly significant for normal plant growth and development. L-tryptophan is applied through foliar spray, seed priming, and soil application. Soil-applied L-TRP is either directly taken up by plants or metabolized to auxin by soil microbiota and then absorbed by plant roots. Similarly, foliar spray and seed priming with L-TRP stimulates auxin synthesis within plants and improves the growth and productivity of agricultural crops. Furthermore, L-TRP contains approximately 14% nitrogen (N) in its composition, which is released upon its metabolism within a plant or in the rhizosphere and plays a role in enhancing crop productivity. This review deals with assessing crop responses under the exogenous application of L-TRP in normal and stressed environments, mode of action of L-TRP, advantages of using L-TRP over other auxin precursors, and role of the simultaneous use of L-TRP and auxin-producing microbes in improving the productivity of agricultural crops. To the best of our knowledge, this is the first review reporting the importance of the use of L-TRP in agriculture.  相似文献

A study was conducted to isolate P-solubilizing bacteria from the rhizosphere of three wild rice species and to test their ability to mobilize P from rock phosphate (RP). Inoculated seeds or seedlings of eight different strains were grown in soils supplemented with a P fertilizer mixture (PFM) consisting triple super phosphate (TSP) and RP, each providing equal amounts of P2O5. Crop growth, NaHCO3-extractable P, crop P uptake and yield were compared with two uninoculated controls, with either TSP or PFM added. In the pot experiment, P availability varied from 20 to 48 mg P kg?1 soil. Yields ranged between 4.8 and 6.6 g per pot and were not significantly different between treatments. In the field experiment, shoot P accumulation in inoculated and TSP-control treatments at the heading stage ranged between 79–129 mg and 219 mg per pot, respectively. Dual inoculants comprising Staphylococcus scirui, Bacillus pumilus, Bacillus subtilus and Bacillus cereus increased yield by about 29% over PFM-controls (324 g m?2) but those yields were 21% lower than TSP-controls (510 g m?2). Therefore, application of inoculants combined with PFM is not a viable alternative for TSP under the tested conditions because yield was limited by the P availability.  相似文献

  1. As a result of sea-level rise (SLR), coastal anchialine pool habitats will be lost in some locations and will expand inland into low-lying areas in others. New and existing habitats may risk additional SLR-related degradation from non-native species transmission, groundwater pollution, and increased contact with human infrastructure. Despite a worldwide distribution, anchialine ecosystems and biota have been largely omitted from SLR risk assessments for coastal ecosystems.
  2. Anchialine pools and caves are composed of brackish groundwater that connects to the marine environment through porous rocky substrate but have no overland connection to the ocean. They support unique endemic biota.
  3. The goal of this study was to develop methods to assess potential impacts to anchialine pools from future coastal flooding, using the island of Hawai'i as a case study. Flood predictions incorporated pool surveys, groundwater-level measurements, and statistical analysis of flood frequencies observed in tide gauges combined with regional scenario-based projections of future sea levels. High-resolution geospatial models were then generated to predict anchialine pool location, density, and risk factors for the next 60 years.

Along 40 km of coastline, up to 80% of current anchialine pools will be lost by 2080 as pools merge with ocean habitats; however, as groundwater flooding occurs more frequently and new habitats are created inland, the total pool counts will rise from 509 in 2018 to 1,000 by 2080. As a result of extreme water-level events, non-native fishes are predicted to disperse into 42% of pools by 2030. Based on current conditions, development and cesspool risks are quite low for anchialine pools in the study area.
  5. Outcomes from this study are guiding conservation actions, including habitat restoration, non-native fish removal, and development planning decisions in coastal areas. This study illustrates methods that can be used to assess the effects of SLR on anchialine pool habitats worldwide.

The Hass cultivar of avocado is the most widely grown commercial cultivar in Mexico. Unfortunately, this cultivar is poorly adapted to the Mexican low-lands with hot dry climates characteristic of northwestern Mexico. Other well-adapted avocado accessions are available for these regions, but their nutritional traits and genetic diversity have yet to be explored. In this study, we analyze oil content, (α-tocopherol) and genetic variation among five local varieties from northwest Mexico that grow in high temperatures regimes in unfertilized soils and without any agronomic management. We report significant phenotypic variability in oil and α-tocopherol components in different accessions of avocado as determined by HPLC. Interestingly, we find higher α-tocopherol content (45.02–50.66 μg/g of fresh pulp) in some local avocados compared to Hass (32.28 μg/g of fresh pulp). The analyzed accessions represent a moderately polymorphic set of genotypes as measured by microsatellite (10 alleles by locus) and SNP (1 SNP every 164.4 bp) analysis of the VTE3 and VTE4 genes, implied in the biosynthesis of tocopherols. SNP data allowed also identifying differences between the local varieties and controls (Hass and the Mexican race accession). The variation observed at the genetic, morphologic and nutritional levels provide significant new information that may be valuable in selecting and developing avocado genotypes adapted to high-temperature environments.  相似文献
