3-Methyl- and 5-methyl-2-(1-pyrrolidinyl)-2-cyclopenten-1-one were recently identified as intense cooling compounds in roasted dark malt. To gain more insights into the molecular requirements of these compounds for imparting a cooling sensation, 26 cyclic alpha-keto enamine derivatives were synthesized, and their physiological cooling activities were evaluated. Any modification of the amino moiety, the carbocyclic ring size, or incorporation of additional methyl groups led to a significant increase of the cooling threshold. Insertion of an oxygen atom into the 2-cyclopenten-1-one ring, however, increased the cooling activity, e.g., the cooling threshold of the 5-methyl-4-(1-pyrrolidinyl)-3(2H)-furanone was found to be 16-fold below the threshold concentration determined for the 3-methyl-2-(1-pyrrolidinyl)-2-cyclopenten-1-one. Shifting the oxygen atom from the 4- into the 5-position of the cyclopentenone ring resulted in a even more drastic increase in cooling activity, e.g., the 4-methyl-3-(1-pyrrolidinyl)-2(5H)-furanone exhibited the strongest cooling effect at the low oral threshold concentration of 0.02-0.06 mmol/L, which is 35-fold below the value determined for (-)-menthol. In contrast to the minty smelling (-)-menthol, most of the alpha-keto enamines were found to be virtually odorless but impart a sensation of "cooling" to the oral cavity as well as to the skin, thus illustrating that there is no physiological link between cooling activity and mint-like odors.  相似文献

Long-term monitoring, 1973 to 1987, of reactions to liming and reacidification of a forest lake ecosystem near the Swedish west coast is reported in this study. Treatment of Lake Lysevatten with a slag product of limestone in 1974 resulted in neutralization and a positive alkalinity. Prolonged dissolution proceeded for about 7 yr whereby 86% dissolved. During 1984–86 Lake Lysevatten approached maximum reacidification with high Al concentrations and an affected biota. Asellus aquaticus L. decreased and dominance within chironomid groups approached preliming conditions. However, the most obvious biological change was the development of the filamentous algal genus Mougeotia and increased growth of Sphagnum. Populations of both plants increased notably when pH declined to about 5. Our study suggests that extensive reacidification (pH < 6.0) of limed lakes should be avoided by successive treatments to prevent development of destabilized lake ecosystems.  相似文献

During November 2006, two live and one dead mouflon (Ovis ammon musimon) were presented with a history of weakness, tremors, and paralysis. After a detailed gross and histologic examination and a bacteriologic, parasitologic, and rabies evaluation, a preliminary diagnosis of tick paralysis was established. A thorough field search revealed 13 affected mouflons found in the open hunting ground "Sveti Juraj" near the town of Senj (Croatia), along with an additional 35 mouflon carcasses. All 13 mouflons were placed in a quiet, semidark stable. All detectable ticks were removed manually, and the animals were topically treated with 250 ppm of Amitraz water emulsion (Taktic 12.5% EC, Intervet International, 5830 Boxmeer, Netherlands). The collected ticks were identified as Ixodes ricinus, Dermacentor marginatus, and Haemaphysalis punctata. In the following 24 hr, all treated animals recovered fully. This report describes a naturally occurring outbreak of tick paralysis in free-ranging mouflons from a karst habitat.  相似文献

Application of a novel screening procedure, the comparative taste dilution analysis (cTDA), on the non-solvent-extractable reaction products formed in a thermally processed aqueous solution of glucose and l-alanine led to the discovery of the presence of a sweetness-enhancing Maillard reaction product. Isolation, followed by LC-MS and 1D- and 2D-NMR measurements, and synthesis led to its unequivocal identification as N-(1-carboxyethyl)-6-(hydroxymethyl)pyridinium-3-ol inner salt. This so-called alapyridaine, although being tasteless itself, is the first nonvolatile, sweetness-enhancing Maillard reaction product reported in the literature. Depending on the pH value, the detection thresholds of sweet sugars, amino acids, and aspartame, respectively, were found to be significantly decreased when alapyridaine was present; for example, the threshold of glucose decreased by a factor of 16 in an equimolar mixture of glucose and alapyridaine. Studies on the influence of the stereochemistry on taste-enhancing activity revealed that the (+)-(S)-alapyridaine is the physiologically active enantiomer, whereas the (-)-(R)-enantiomer did not affect sweetness perception at all. Thermal processing of aqueous solutions of alapyridaine at 80 degrees C demonstrated a high thermal and hydrolytic stability of that sweetness enhancer; for example, more than 90 or 80% of alapyridaine was recovered when heated for 5 h at pH 7.0, 5.0, or 3.0, respectively.  相似文献

  • 1. Fishing spans all oceans and the impact on ocean predators such as sharks and rays is largely unknown. A lack of data and complicated jurisdictional issues present particular challenges for assessing and conserving high seas biodiversity. It is clear, however, that pelagic sharks and rays of the open ocean are subject to high and often unrestricted levels of mortality from bycatch and targeted fisheries for their meat and valuable fins.
  • 2. These species exhibit a wide range of life‐history characteristics, but many have relatively low productivity and consequently relatively high intrinsic vulnerability to over‐exploitation. The IUCN — World Conservation Union Red List criteria were used to assess the global status of 21 oceanic pelagic shark and ray species.
  • 3. Three‐quarters (16) of these species are classified as Threatened or Near Threatened. Eleven species are globally threatened with higher risk of extinction: the giant devilray is Endangered, ten sharks are Vulnerable and a further five species are Near Threatened. Threat status depends on the interaction between the demographic resilience of the species and intensity of fisheries exploitation.
  • 4. 4. Most threatened species, like the shortfin mako shark, have low population increase rates and suffer high fishing mortality throughout their range. Species with a lower risk of extinction have either fast, resilient life histories (e.g. pelagic stingray) or are species with slow, less resilient life histories but subject to fisheries management (e.g. salmon shark).
  • 5. 5. Recommendations, including implementing and enforcing finning bans and catch limits, are made to guide effective conservation and management of these sharks and rays.

Haemophilus parasuis infection in pigs is characterized by fibrinous polyserositis, arthritis and meningitis. Despite the fact that traditional diagnosis is based on herd history, clinical signs, bacterial isolation and serotyping, molecular-based methods are alternatives for species-specific tests and epidemiological studies. The aim of this study was to characterize H. parasuis field strains from different states of Brazil, employing serotyping and genotyping methods. Serotyping revealed that serovar 4 was the most prevalent (26.1%), followed by serovars 5 (17.4%), 14 (8.7%), 13 (4.4%) and 2 (4.4%), whereas 39% of the strains were considered as untypeable. AFLP with a single enzyme and PFGE were able to type all isolates tested, generating 34 and 20 different profiles, respectively, including untypeable strains. Besides the slightly higher discrimination index presented by AFLP, PFGE with Not I restriction enzyme showed a better correlation with epidemiological data, grouping strains of the same serovar, animal or farm origin. The results indicated AFLP and PFGE as valuable tools for typing H. parasuis isolates collected in Brazil.  相似文献

Throughfall has been sampled in SW Sweden from 60 to 80 yr old Norway spruce (Picea abies) of two stands with different crown thinning classes, representing healthy reference trees and injured trees. the condition of the canopy had a significant influence on quantity and quality of deposition to the forest floor. Annual throughfall deposition from injured spruce showed an increase in glucose 270%, total sugar 80%, organic carbon (DOC) 40%, organic N 50%, Mg 100%, Ca 30%, and Fe 40% while Mn and K deposition decreased 30% and 20%, respectively, compared to annual throughfall deposition from healthy spruce.  相似文献

Activity-directed fractionation of heated carbohydrate/alanine solutions recently led to the discovery of (+)-(S)-1-(1-carboxyethyl)-5-hydroxy-2-(hydroxymethyl)pyridinium inner salt (1, alapyridaine), and it has been shown that this compound lowers the detection thresholds of sugars, glutamate, and NaCl solutions, whereas no influence on bitter perception was observed. As this class of Maillard-derived pyridinium betaines seemed to be promising targets for further research on their taste modulatory activity, the objective of the present investigation was to screen for bitter taste-suppressing target molecules in combinatorial libraries of pyridinium betaines prepared from 5-(hydroxymethyl)furan-2-aldehyde and amino acid mixtures by use of Maillard-type reaction chemistry instead of synthesizing and purifying each derivative individually. By application of hydrophilic interaction liquid chromatography in combination with the recently developed comparative taste dilution analysis, followed by structure determination, synthesis, and sensory studies, we have now succeeded in identifying 1-carboxymethyl-5-hydroxy-2-hydroxymethylpyridinium inner salt (2) as a potential bitter-suppressing candidate. While tasteless on its own, 2 was found to reduce the bitterness of various bitter tastants such as the amino acid L-phenylalanine, the peptide Gly-Leu, the alkaloid caffeine, and the bitter glycosides salicin and naringin.  相似文献
