For the use of gene expression for nutritional diagnosis, a rapid and easy method of detecting levels of gene expression was proposed. Agrobacterium tumefaciens carrying the GUS reporter gene was injected into intact tobacco leaves with a plastic syringe, and transient GUS expression was determined. This article suggests a linear functional relationship model for comparing two sets of circular data subject to unobservable errors. Unlike the corresponding and relatively well-studied model for linear data, maximum likelihood estimation for this model is very complicated and no explicit solutions are possible. Using a numerical approximation, we are able to solve the likelihood equations approximately, and to obtain good approximations to the likelihood estimates of the parameters. The quality of our estimates and the feasibility of the estimation method are illustrated via simulation. By establishing a parallel with the model for linear data, we are able to explain the various problems occurring in the process of estimation and to substantiate our numerical results. The interest in the model arose in connection with the study of ocean wave data; an application to such data is also given. 相似文献

The mechanisms by which hydrophobic molecules, such as long-chain fatty acids, enter cells are poorly understood. In Gram-negative bacteria, the lipopolysaccharide layer in the outer membrane is an efficient barrier for fatty acids and aromatic hydrocarbons destined for biodegradation. We report crystal structures of the long-chain fatty acid transporter FadL from Escherichia coli at 2.6 and 2.8 angstrom resolution. FadL forms a 14-stranded beta barrel that is occluded by a central hatch domain. The structures suggest that hydrophobic compounds bind to multiple sites in FadL and use a transport mechanism that involves spontaneous conformational changes in the hatch. 相似文献

【目的】介绍ANUSPLIN在黄土高原多沙粗沙区时间序列气象要素的空间插值过程,为相关人员在ANUSPLIN的参数设置、误差分析和协变量要素选择等方面提供参考。【方法】以薄盘样条函数为插值理论,以专用气象数据插值软件ANUSPLIN为实现工具,并引入一个或多个协变量线性子模型,来实现多个气象要素的空间插值。【结果】完成了黄土高原多沙粗沙区时间序列(1980~2000年)多个气象要素月平均数据的栅格化,计算了气象要素随其影响因子变化的关系。【结论】ANUSPLIN以薄盘光滑样条函数为理论基础,引入协变量线性子模型,能较好地提高气象要素空间插值精度,且能反映气象要素随其影响因子变化的比率关系。在大多数情况下,该区模型选择以样条次数为3次的局部薄盘光滑样条函数模型为最佳。温度的空间插值相对比较容易,且误差较小,1995-07平均相对误差为1%;风速、水汽压的误差中等;日照时数和降雨量的误差较大,个别情况相对误差可超过50%。 相似文献

As the aquaculture industry intensively develops, its environmental impact increases. Discharges from aquaculture deteriorate the receiving environment and the need for fishmeal and fish oil for fish feed production increases. Rotating biological contactors, trickling filters, bead filters and fluidized sand biofilters are conventionally used in intensive aquaculture systems to remove nitrogen from culture water. Besides these conventional water treatment systems, there are other possible modi operandi to recycle aquaculture water and simultaneously produce fish feed. These double-purpose techniques are the periphyton treatment technique, which is applicable to extensive systems, and the proteinaceous bio-flocs technology, which can be used in extensive as well as in intensive systems. In addition to maintenance of good water quality, both techniques provide an inexpensive feed source and a higher efficiency of nutrient conversion of feed. The bio-flocs technology has the advantage over the other techniques that it is relatively inexpensive; this makes it an economically viable approach for sustainable aquaculture. 相似文献

Tree removal in Latin American coffee agroforestry systems has been widespread due to complex and interacting factors that include fluctuating international markets, government-supported agricultural policies, and climate change. Despite shade tree removal and land conversion risks, there is currently no widespread policy incentive encouraging the maintenance of shade trees for the benefit of carbon sequestration. In facilitation of such incentives, an understanding of the capacity of coffee agroforests to store carbon relative to tropical forests must be developed. Drawing on ecological inventories conducted in 2007 and 2010 in the Lake Atitlán region of Guatemala, this research examines the carbon pools of smallholder coffee agroforests (CAFs) as they compare to a mixed dry forest (MDF) system. Data from 61 plots, covering a total area of 2.24 ha, was used to assess the aboveground, coarse root, and soil carbon reservoirs of the two land-use systems. Results of this research demonstrate the total carbon stocks of CAFs to range from 74.0 to 259.0 Megagrams (Mg)?C ha?1 with a mean of 127.6?±?6.6 (SE)?Mg?C ha?1. The average carbon stocks of CAFs was significantly lower than estimated for the MDF (198.7?±?32.1?Mg?C?ha?1); however, individual tree and soil pools were not significantly different suggesting that agroforest shade trees play an important role in facilitating carbon sequestration and soil conservation. This research demonstrates the need for conservation-based initiatives which recognize the carbon sequestration benefits of coffee agroforests alongside natural forest systems. 相似文献

This paper reviews the currently available instruments for measuring the distribution of biomass within forest canopies. We compare the most well-established approaches and present the different measurable parameters. A special focus lies on the resolution of the obtained data.? Results
It was found that only 3D laser scanners offer data with the resolution required by ecologists, private landholders, the forest industry and the public to detect trends in tree growth patterns and canopy interactions in all three spatial dimensions. But data validation, data analysis and parameter extraction are still under development, and the price of the instrument is quite high.? Conclusion
Research should focus on the parameter extraction from terrestrial laser scanner data as this could allow the calculation of functional attributes for different sections of a canopy on a high spatial resolution. We used a multiplexed approach based on flow-stretched DNA to monitor the enzymatic digestion of lambda-phage DNA by individual bacteriophage lambda exonuclease molecules. Statistical analyses of multiple single-molecule trajectories observed simultaneously reveal that the catalytic rate is dependent on the local base content of the substrate DNA. By relating single-molecule kinetics to the free energies of hydrogen bonding and base stacking, we establish that the melting of a base from the DNA is the rate-limiting step in the catalytic cycle. The catalytic rate also exhibits large fluctuations independent of the sequence, which we attribute to conformational changes of the enzyme-DNA complex. 相似文献
