Rainbow trout (average weight of 2 g) in fresh water experienced high mortality and were infected with a diplomonad intestinal parasite. Tanks of fish experienced an immediate reduction in mortality after an in-feed treatment with 3% Epsom salts for 2 d. Treatments had to be applied several times, but in each case there was a similar reduction in mortality.  相似文献

A highly selective enzyme-linked immunosorbent assay (ELISA) has been developed for the quantitative detection of the Cry1Ac protein expressed in transgenic cotton. Two Cry1Ac-specific monoclonal antibodies (MAb), Kbt and 158E6, were developed and selected to form a sandwich format ELISA. The MAb Kbt was used as a capture antibody, and 158E6 was conjugated with horseradish peroxidase and served as a detection antibody. The assay was optimized and validated with different cotton matrices. Tissues were extracted with phosphate-buffered saline containing 0.05% Tween 20 and 1% polyvinylpyrrolidone. The extract was then treated with trypsin to truncate full-length Cry1Ac into the core toxin for quantitation. The resulting assay has good accuracy and precision with a validated limit of quantitation ranging from 0.1 to 0.375 mug/g dry weight of cotton tissues. This assay is highly specific for Cry1Ac protein and has no cross-reactivity with the nontarget proteins tested such as Cry1Ab and Cry1F.  相似文献

The active site of voltage-activated potassium channels is a transmembrane aqueous pore that permits ions to permeate the cell membrane in a rapid yet highly selective manner. A useful probe for the pore of potassium-selective channels is the organic ion tetraethylammonium (TEA), which binds with millimolar affinity to the intracellular opening of the pore and blocks potassium current. In the potassium channel encoded by the Drosophila Shaker gene, an amino acid residue that specifically affects the affinity for intracellular TEA has now been identified by site-directed mutagenesis. This residue is in the middle of a conserved stretch of 18 amino acids that separates two locations that are both near the external opening of the pore. These findings suggest that this conserved region is intimately involved in the formation of the ion conduction pore of voltage-activated potassium channels. Further, a stretch of only eight amino acid residues must traverse 80 percent of the transmembrane electric potential difference.  相似文献

Quantifying fish movements in river networks helps identify critical habitat needs and how they change with environmental conditions. Some of the challenges in tracking fish movements can be overcome with the use of passive integrated transponder (PIT) tagging and antennas. We used PIT technology to test predictions of movement behaviour for four fish species at a mainstem–tributary confluence zone in an arid‐land river system. Specifically, we focused on the McElmo Creek tributary confluence with the San Juan River in south‐western Utah, USA. We quantified variation in species occurrences at this confluence zone from May 2012 to December 2015 relative to temporal and environmental conditions. We considered occurrences among species relative to tagging origins (tributary versus mainstem), season and time of day. Generally, fishes tagged in the focal tributary were more likely to be detected compared to fish tagged in the mainstem river or other tributaries. Additionally, adults were most likely to be detected across multiple years compared to subadults. Based on a Random Forests model, the best performing environmental variables for predicting seasonal detections included mainstem discharge during run‐off season (razorback sucker Xyrauchen texanus), tributary discharge during monsoon season (Colorado pikeminnow Ptychocheilus lucius) and mainstem water temperature (flannelmouth sucker Catostomus latipinnis and channel catfish Ictalurus punctatus). The variable responses by endemic and introduced fishes indicate tributary habitats provide several key functions within a fish community including spawning, rearing, foraging and refuge.  相似文献

Methylibium petroleiphilum PM1, which is capable of degrading of methyl tert-butyl ether (MTBE), was immobilized in calcium alginate gel beads. Various applications were explored to increase the mechanical strength of these gel beads. The introduction of 0.3 mol/L calcium chloride into the crosslinking solution, 0.002 mol/L calcium chloride into the growth medium, and 0.2% polyethyleneimine (PEI) as chemical crosslinking agent increased the stability of the Ca-alginate gel beads under the operation conditions of the bioreactor. The degradation rates of MTBE by the immobilized cells in the bioreactor system operated in batch and continuous mode , respectively, were compared. A MTBE biodegradation rate of 5.79 mg/L·h was reached for over 400 h (50 batches), and the immobilized cells in the bioreactor removed >96% MTBE during 50 days of operation. Molecular analysis of the PM1 cells revealed that microbial growth occurred predominantly as microcolonies in the outer area of the beads during the first 20 days of operation. The results of this study show that a continuous-mode, fixed-bed bioreactor reactor coupled with PM1-immobilized cells is a promising technology for remediating MTBE-contaminated groundwater.  相似文献

A commercially available one-row harvester was modified to meet a number of needs that are often encountered when conducting field trials with potatoes. The harvester can be utilized for both small plot collection (e.g., 8 m row lengths) as well as for bulk harvesting of larger plots (e.g., 65 m row lengths). The equipment alteration time required for conversion from one system to the other is minimal (less than 30 min) and the harvester has been altered to accommodate typical row widths and harvesting conditions in many N. American potato growing areas. It has been structurally modified to improve stability during operation to overcome a tendency to tip-over under certain circumstances. In addition, bulk harvesting operations with this machine involves a minimum of three persons as compared to a minimum of six generally required for a traditionally utilized digger-picker harvester.  相似文献

Context

This study synthesizes evidence from three separate surveys of American Black Duck and Mallard breeding habitat usage to quantify the effects of agriculture at the landscape scale.

Objectives

To assess duck breeding activity in agricultural landscapes within the Canadian maritimes in order to measure the overall impact of agricultural land use, the response to particular agricultural activities, and the influence of landscape configuration.

Methods

Models, constructed using a long-term census (SURVEY1), were used to predict habitat selection for two other independent surveys (SURVEY2, SURVEY3). Predictions incorporated information about wetland area and diversity, as well as anthropogenic factors, allowing subsequent analyses to focus on the remaining residual error attributable to agricultural effects.

Results

SURVEY2 results demonstrated that the proportion of active agriculture is an important indicator of the severity of human disturbance, yielding threshold estimates of 39% for Mallards and 60% for Black Ducks, with an overall average of 50%. Agricultural conversion beyond these thresholds deterred breeding ducks independently of other factors. SURVEY3 land cover information demonstrated that the presence of cropland intensified this deterrence effect, even at levels as low as 10%. Woodland cover (in excess of 30%) was important for both species, but its configuration was not.

Conclusions

In addition to quantifying threshold effects, this study reaffirms that woodland is an important part of the maritime landscape matrix, and contributes positively to habitat diversity in mixed use, moderate intensity agricultural regions. Wetland restoration in agricultural landscapes should monitor and promote less crop-intensive, mixed-use practices.

TRAAK channels, members of the two-pore domain K(+) (potassium ion) channel family K2P, are expressed almost exclusively in the nervous system and control the resting membrane potential. Their gating is sensitive to polyunsaturated fatty acids, mechanical deformation of the membrane, and temperature changes. Physiologically, these channels appear to control the noxious input threshold for temperature and pressure sensitivity in dorsal root ganglia neurons. We present the crystal structure of human TRAAK at a resolution of 3.8 angstroms. The channel comprises two protomers, each containing two distinct pore domains, which create a two-fold symmetric K(+) channel. The extracellular surface features a helical cap, 35 angstroms tall, that creates a bifurcated pore entryway and accounts for the insensitivity of two-pore domain K(+) channels to inhibitory toxins. Two diagonally opposed gate-forming inner helices form membrane-interacting structures that may underlie this channel's sensitivity to chemical and mechanical properties of the cell membrane.  相似文献
