Morphologic polarity is necessary for chemotaxis of mammalian cells. As a probe of intracellular signals responsible for this asymmetry, the pleckstrin homology domain of the AKT protein kinase (or protein kinase B), tagged with the green fluorescent protein (PHAKT-GFP), was expressed in neutrophils. Upon exposure of cells to chemoattractant, PHAKT-GFP is recruited selectively to membrane at the cell's leading edge, indicating an internal signaling gradient that is much steeper than that of the chemoattractant. Translocation of PHAKT-GFP is inhibited by toxin-B from Clostridium difficile, indicating that it requires activity of one or more Rho guanosine triphosphatases. 相似文献

Résumé Les auteurs ont contrôlé, pendant deux cycles annuels, les analyses d'échantillons mensuels d'huile de palme produite industriellement dans une quinzaine de points-origines. L'enquête couvrait les territoires de l'Afrique Noire Française, portugaise, espagnole et la Malaisie britannique. Elle portait sur l'étude de certaines caractéristiques et sur leurs variations: indice d'iode, titre des acides gras, point de fusion et spécialement carotène de ces huiles de palme, selon origine variétale, territoire et saison de production.En ce qui concerne, en particulier, l'indice d'iode et la teneur en carotène des huiles de palme, ces résultats mettent en évidence la prépondérance du facteur variétal: les huiles provenant des palmeraies naturelles de Côte d'Ivoire, ont un indice d'iode élevé (57 à 60). Celles des palmeraies naturelles du Togo, du Dahomey, des Guinée portugaise et française, ont les plus fortes teneurs en carotène (maxima à 0,16 et même 0,19%) avec un indice d'iode moyen (54–56).Les huiles de plantations industrielles sélectionnées en Dura-Deli, qu'elles soient d'Extrême-Orient ou de Côte d'Ivoire, ont simultanément des indices d'iode faibles (52–53) et des teneurs très basses en carotène (0,05%).Au Sud de l'Equateur en Afrique, toutes les huiles de palme observées au cours de cette enquête, avaient conjointement un indice d'iode faible ou moyen (53 à 55) et une teneur généralement faible en carotène (0,05).Au cours du cycle annuel, la teneur en carotène varie sensiblement du tiers de son maximum: ce dernier, très marqué en Côte d'Ivoire, au Togo et au Dahomey, est atteint à la période de forte production d'huile (de janvier à mai) sur les palmeraies naturelles, c'est-à-dire en saison sèche et chaude avec bonne insolation.De fortes précipitations font baisser rapidement la teneur en carotène avec un décalage de l'ordre de six semaines, durée connue de la formation de l'huile dans le fruit.Conjointement, on pousse les études sur les huiles résiduelles de fibres extraites au solvant; elles sont enrichies de 2 à 3 fois en caroténoïdes par rapport à l'huile de palme, mais la proportion de bétacarotène semble légèrement inférieure.Les auteurs tendent vers une spécification de certaines huiles pour des emplois nettement définis.
Summary During two years the authors have assembled monthly analytical data of oilpalms, from 15 different stations. The determinations spread as far as the French, Portuguese and Spanish territory in Africa and British Malaya also. The following analyses were carried out: iodine number, titer point, melting point and the carotenoids of these oils, according to origine, race and time of gathering.As to iodine number and amount of carotenoid, the race is decisive for these data; the oils from the natural stock of the Ivory Coast have a higher iodine number (57–60). The oils from the natural stock of Togo, Dahomey, Portuguese and French Guinea are richer in carotene (up to 0.16, even 0.19%) with medium iodine number (54–56).The oils from the industrial plantations, with selected trees of the race Dura-Deli, from the Far East as well as from the Ivory Coast, have lower iodine numbers (52–53) and are poor in carotene (0.05).South of the equator in Africa, all analysed races of oil palms had a lower iodine number (53–55) and were poor in carotene (0.05).During one year the amount of carotene fluctuates about one third of its maximum; this maximum is rather striking in Togo and Dahomey; it is to be found from January to May; period of high production of the oil in the Palm groves, e.g. in the dry season with warm climate and good insolation. Heavy rain-showers effect a rapid decrease of the contents of carotene after six weeks (duration of the formation of the fruit).Furthermore, the residual oils (extracted by solvents) were analysed; they are 2 to 3 times richer in carotenoids than the common palm oil; but the contents of -carotene seems somewhat lower.The authors think it possible to find exactly defined uses for the different oils.
Zusammenfassung Zwei Jahre lang haben die Autoren monatliche Analysendaten von Oelpalmen aus 15 verschiedenen Stationen zusammengestellt. Die Untersuchungen erstreckten sich auf ein Gebiet, das die französischen, portugiesischen, und spanischen Territorien aus Afrika umfasst, und dazu Britisch Malaya. Es wurden bestimmt: Iodzahl, Säuretiter, Schmelzpunkt und besonders der Karotingehalt dieser Oele, je nach Herkunft, Sorte, und Zeitpunkt der Ernte.Was Iodzahl und Karotinmenge betrifft, ist die Sorte massgebend für diese Daten; die Oele aus den natürlichen Beständen der Elfenbeinküste haben eine höhere Iodzahl (57–60). Weiterhin, wurden die extrahierten Oele der Abfälle untersucht; sie sind 2 bis 3 mal reicher an Karotin als gewöhnliches Palmoel, aber der -Karotin-Gehalt scheint etwas geringer zu sein. It has been argued that large ungulates play a key role in natural forest dynamics, but in Britain, the largest native ungulates
(aurochs and elk) are extinct. Cattle could have some similar effects, and are widely used, but rarely tested, for nature
conservation management. Here, we test conservation management with cattle at a native Scots pine Pinus sylvestris forest in Scotland. Our hypotheses were that cattle impacts would (a) increase the abundance of an understorey shrub of conservation
importance, bilberry Vaccinium myrtillus; and (b) increase Scots pine seedling establishment. Two cattle trials were established, one for each objective, based on
a 14-ha plot within the forest, and a 6.1-ha plot at the forest edge. Cattle were introduced for 2 months to both plots, giving
an approximately 100- to 150-fold increase in ungulate biomass per hectare when compared to background deer abundance. The
ground layer vegetation in both treatment and matched control plots was monitored before and after treatment, and subsequently
over a 4-year period. At the within-forest trial, bilberry percent-cover, 4 years after treatment, was 1.9 times higher in
the cattle area than in the control (95% confidence limits 1.6–2.3). Bilberry percent-cover increases were directly related
to the degree of trampling impact on heather Calluna vulgaris, as recorded immediately after the trial. At the forest-edge trial, there were vegetation changes in the cattle plot that
were considered favourable to pine seedling establishment: reduced moss/litter depth and vegetation percent-cover, and increased
ground-level light incidence. However, too few new pine seedlings were found for formal analysis, partly due to unexpectedly
low seed-fall. Nevertheless, the potential for cattle to create conditions that would subsequently promote pine regeneration
was illustrated by the observation of large numbers of new, young pine seedlings at the within-forest cattle plot. We recommend
that managers and researchers collaborate to develop further trials like the ones reported here, to give an improved understanding
of the conditions under which cattle can yield nature conservation benefits in woodlands. 相似文献
