The aim of this study was to determine the synergistic effects of centrifuged egg yolk (EY) and soybean lecithin on post-thaw Caspian horse sperm motility, morphological abnormalities, and assessment of membrane integrity. The centrifuged EY (CEY) was added at concentrations of 2% and 4% to a defined INRA plus 1.25% soybean lecithin extender used to freeze Caspian horse semen. In this experiment, ejaculates collected from each Caspian horse (n = 4) were divided into three equal aliquots and diluted in CEY 2% (INRA2), 4% (INRA4) supplemented, and without any CEY (INRA0) in INRA plus 1.25% soybean lecithin extender, respectively. Thereafter, samples were frozen and thawed following a standard protocol. Sperm cryosurvival was evaluated in vitro by microscopy assessments of post-thaw sperm motility (by means of computer-assisted semen motility analysis [CASA]), acrosomal and other abnormalities (head, mid-pieces, and tail) and plasma membrane integrity (evaluated by HOST). In Caspian stallion, semen extended with INRA2 had significantly higher CASA motility and CASA progressive motility than those extended with the rest of extenders after freezing and thawing (P < .001). There was no significant difference in path velocity (VAP), VCL, and ALH among three groups (P > .05). For straight line velocity (P < .01) and LIN (P < .001), the highest values were obtained from the INRA4 group. The highest percentages of acrosomal and other abnormalities were found in semen diluted in INRA4 (P < .001). In the group frozen INRA2, the percentage of membrane integrity was significantly higher than that of the other groups (P < .001). The use of CEY 2% in combination with soybean lecithin significantly improved Caspian horse semen freezability. 相似文献

Mesenchymal stem cells are undifferentiated cells that have the ability to divide continuously and tissue regeneration potential during the transplantation. Aging and loss of cell survival, is one of the main problems in cell therapy. Since the production of free radicals in the aging process is effective, the use of antioxidant compounds can help in scavenging free radicals and prevent the aging of cells. The aim of this study is evaluate the effects of L-carnitine (LC) on proliferation and aging of rat adipose tissue-derived mesenchymal stem cells (rADSC). rADSCs were isolated from inguinal region of 5 male Rattus rats. Oil red-O, alizarin red-S and toluidine blue staining were performed to evaluate the adipogenic, osteogenic and chondrogenic differentiation of rADSCs, respectively. Flow cytometric analysis was done for investigating the cell surface markers. The methyl thiazol tetrazolium (MTT) method was used to determine the cell proliferation of rADSCs following exposure to different concentrations of LC. rADSCs aging was evaluated by beta-galactosidase staining. The results showed significant proliferation of rADSCs 48 h after treatment with concentrations of 0.2 mM LC. In addition, in the presence of 0.2 mM LC, rADSCs appeared to be growing faster than control group and 0.2 mM LC supplementation could significantly decrease the population doubling time and aging of rADSCs. It seems that LC would be a good antioxidant to improve lifespan of rADSCs due to the decrease in aging. 相似文献

The aim of this study was to investigate the effects of transplanted Wharton’s jelly mesenchymal stem cells (WJMSCs) of caprine
umbilical cord on cutaneous wound healing process in goat. After collection of caprine pregnant uterus of mixed breed goats
from abattoir, the Wharton’s jelly (WJ) of umbilical cord was harvested. The tissues were minced in ventilated flasks and
explant culture method was used for separating mesenchymal stem cells (MSCs). The isolated cells were immunostained for Actin
protein, histochemically assayed for the presence of alkaline phosphatase activity, and analyzed for detection of matrix receptors
(CD44) and hematopoetic lineage markers (CD34), using flow cytometery. After The isolated cells, 3 × 106 MSCs were stained with BrdU and prepared for transplantation to each wound. Four 3-cm linear full thickness skin incisions
were made on both sides of thoracic vertebrate of four Raeini goats (two wounds on each side). The left wounds were implanted
with MSCs in 0.6 ml of Phosphate buffer saline (PBS), and the right wounds considered as control group that received 0.6 ml
of PBS. The samples were taken from the wounds 7 and 12 days after the wounding, and healing process was compared histologically
between the two groups. Anti-BrdU staining showed that the transplanted cells were still alive in the wound bed during the
study. The histopathological study revealed that re-epithelialization was complete at days 7 in treated wounds with WJMSCs,
whereas in control wound the wounds still showed incomplete epithelialization 12 days after wounding. Also, microscopic evaluation
showed less inflammation, thinner granulation tissue formation with minimum scar in the treated wounds in comparison with
control wounds. In conclusion, this study demonstrates the beneficial effect of caprine WJMSCs in cutaneous wound healing

We have employed both common and novel methods to measure the amount of silver released from nanoparticle composite packaging. Forty-eight packages of green tiger prawns (Penaeus semisulcatus) were prepared: 24 packages with nanoparticle packaging and 24 packages without nanoparticle packaging. The normal shelf life of P. semisulcatus in the refrigerator is 1–2 days, and we therefore we tested the packages after 0, 1, 2, 3, 4, 5 and 6 days. We measured the diameter of the inhibition zone of the targeted microorganisms (Escherichia coli and Staphylococcus aureus) and examined scanning electron microscopy (SEM) images, using both titration and migration methods, to measure the amounts of silver nanoparticles released. Compared to the standard amount allowed by standard 13737-3 of the National Information Standards Organization, our results indicated the superior sensitivity of the titration method over the migration method in terms of determining the remaining amount of nanoparticles (P < 0.05). The evaluation of SEM images indicated the homogeneity of the surface of the silver nanoparticle packaging and the even exchange capacity of titanium dioxide. The average size of the nanoparticles in the packaging films was determined to be 30–48 nm. The diameter of the inhibition zone in the silver nanoparticle, measured at the six time points, was significantly larger than that seen with other packaging, except for the phosphomycin and penicillin antibiogram disks (P < 0.01). The sensitivity of the titration method using the titration equipment was significantly higher than that of the migration method (P < 0.05). The findings indicate that nanoparticle packaging decreased the microbial count and enhanced the shelf life of P. semisulcatus, leading us to recommend the utilization of silver nanoparticle packaging. 相似文献
