Dietary ellagic acid and related polyphenols are metabolized in humans to dibenzopyran-6-one derivatives, and the microbial origin of these metabolites has been suggested. However, this has not been demonstrated so far. Fecal samples donated by six volunteers were incubated under anaerobic conditions, and aliquots were used to evaluate the fecal metabolism of ellagic acid, the ellagitannin punicalagin, and an ellagitannin rich extract from walnuts. The isoflavone daidzein was also incubated with the same fecal samples to follow the production of the microbial metabolites previously reported (dihydrogenistein, O-demethylangolensin, and equol) as a positive control of the system and to evaluate similarities between isoflavone and ellagic acid fecal flora metabolism. After fermentation the metabolite "urolithin A" (3,8-dihydroxy-6H-dibenzo[b,d]pyran-6-one) was produced from ellagic acid, punicalagin, and the ellagitannin extract in all the fecal cultures from different volunteers, but with very different production rates and concentrations. This large variability in the concentration of metabolite and kinetics of metabolite production is consistent with the large variability found in the excretion of these metabolites in urine in vivo after human consumption of ellagitannins, and with differences in the composition of the fecal microflora. No correlation between isoflavone and ellagic acid metabolism by fecal microflora was observed. The present study confirms the microbial origin of the recently reported in vivo generated hydroxy-6H-dibenzo[b,d]pyran-6-one derivatives in humans and is a further step in the study of the bioavailability and metabolism of ellagic acid and ellagitannins.  相似文献

The analytical process of lycopene extraction and photometrical determination was critically examined for raw tomato and processed tomato products by means of a 2 IV (15-10) Plackett-Burman experimental design in order to identify the key factors (KFs) involved. Fifteen apparent key factors (AKFs) reported in the literature were selected: sample weight (X1); volume of extraction solution (X2); antioxidant concentration (BHT, X3); neutralizing agent concentration (MgCO 3, X4); light presence during lycopene extraction (X5), homogenization velocity (X6) and time (X7), agitation time (X8), and temperature (X9) during the extraction process; water volume for separation of polar/nonpolar phases (X11); presence of inert atmosphere throughout the process (X12); time (X13), temperature (X14), and light presence (X10) during separation of phases and time delay for reading (X15). In general, higher lycopene concentrations in samples led to a higher number of key factors (KF). Thus, for raw tomato (lycopene range 1.22-2.29 mg/100 g) no KF were found, whereas for tomato sauce (lycopene range from 5.80 to 8.60 mg/100 g) one KF (X4) and for tomato paste (lycopene range from 35.80 to 51.27 mg/100 g) five KFs (X1, X2, X4, X11, and X12) were detected. For lycopene paste, X1 and X2 were identified as the KFs with the greatest impact on results, although in fact the X1/X2 ratio was the real cause. The results suggest that, with increased processing, the physical and chemical structure of lycopene becomes less important since the identified KFs explain almost 90% of variability in tomato paste but only 32% in raw tomato.  相似文献

During an experimental infection of sheep with Fasciola hepatica or F. gigantica, MM3-SERO and MM3-COPRO ELISA tests were applied to compare the kinetics of antibody production and coproantigen release between the 2nd and 32nd week post-infection (wpi). The Kato-Katz technique was used to measure the kinetics of egg shedding by both Fasciola species (eggs per gram of feces, epg). The kinetics of IgG antibodies for all sheep infected with F. hepatica and F. gigantica followed a similar pattern. Optical density (OD) increased rapidly between the 4th until the 12th wpi, when the highest values were reached and then decreased slowly until the 32nd wpi. Coproantigen levels increased above the cut-off value between 6 and 9 wpi in the F. hepatica group, and between 9 and 11 wpi in the F. gigantica group. The comparison between coproantigen levels and epg indicated that F. hepatica-infected sheep had detectable amounts of coproantigens 4–7 weeks before patency (egg shedding), while F. gigantica-infected sheep had detectable amounts of coproantigens 3–6 weeks before patency. When comparing the kinetics of coproantigen release vs the kinetics of epg, a similar pattern emerged, but with a two-week time-lag in epg, for both F. hepatica and F. gigantica infections. The amount of coproantigen release by each adult was not burden dependent for F. hepatica infection (burden of 33–66 adults), while it was for F. gigantica infection (burden of 17–69 adults). The results demonstrate the usefulness of the MM3-SERO and MM3-COPRO ELISAs as tools for the diagnosis of early as well as long-term fascioliasis infections, and suggest that they can potentially be applied to human fascioliasis even in countries where F. hepatica and F. gigantica co-exist. These tests can be employed not only in the diagnosis, but also in studies on epidemiology as well as pathogenesis and treatment in animals and humans since they allow post-treatment infection monitoring.  相似文献

We studied short‐scale variation in the total concentration of copper and its fractions in a soil vineyard. Soil samples were collected at a depth of 0–20 cm between plant rows (1), between individual plants (2) and at their base (3) in a vine‐grown plot in NW Spain. The mean total content in Cu (Cut) in the soil was found to be 97 mg kg−1 and that of potentially available Cu (CuEDTA) 34 mg kg−1. Copper bound to organic matter (CuOM) and to non‐crystalline inorganic soil components (CuIA) were the dominant fractions and accounted for 34% of total copper each. The contents in exchangeable (Cue), pyrophosphate‐extractable (Cup), oxalic/oxalate‐extractable (Cuo) and total copper (Cut) exhibited statistically significant correlations with pH, sum of base cations (S), cation‐exchange capacity (CEC) and exchangeable calcium (Cae). Both total and fractional copper contents were higher in plant rows than between them, particularly in the centre of the plot. Also, CuOM and CuIA were higher in planting rows than between rows. These copper results may have been influenced by the vine‐growing practices of the area and also by the distribution of plants and their pruning. This variability pattern for Cu distribution is crucial with a view to minimising potential adverse effects of fungicides and optimising any reclamation treatments needed. Production of triploid turbot is a relevant goal for the aquaculture industry due to the benefits associated with their sterility and growth. In order for a better understanding of the final quality of the species, muscle cellularity and quality parameters of the flesh were studied in seven diploid and seven triploid turbot after sexual maturation. White muscle cellularity was evaluated throughout white muscle cross‐sectional area from the epaxial left muscle section of each fish. To ascertain the flesh quality, several physico‐chemical parameters (moisture, ash, protein, total fat, fatty acids and minerals) were analysed, and textural mechanical properties (springiness, hardness, chewiness, cohesiveness, gumminess and adhesiveness) were determined objectively with a texturometer. Muscle cellularity was different between both groups, such that muscle fibre density was higher for diploid specimens (< 0.001) and muscle fibres diameters were higher for triploids (< 0.001). Both ploidy groups showed no significant differences in moisture, ash, protein and total fat content (> 0.05). All textural properties analysed showed no significant differences between both ploidy groups (> 0.05). In both ploidy groups, fibre density and fibre diameter showed no significant correlations (> 0.05) with textural parameters such as springiness, cohesiveness, chewiness and adhesiveness. Ploidy did not affect fatty acid composition, with the exception of eicosapentaenoic acid (C20:5ω3) which was higher in the triploid group. No significant differences (> 0.05) existed between diploid and triploid turbot in content of all minerals analysed. According to our results, the ploidy status determines significant variations on muscle cellularity but does not influence the flesh quality of the turbot.  相似文献

Soil inputs produced by rainfall waters washed off through a Ribeiro vineyard (Galicia, northwestern Spain) confirmed that levels recovered are <5% of the added fungicides. Laboratory studies showed that some fungicides were degraded in the ODS cartridge used for the collection of fungicides washed off by rainfall. Procymidone wash-off data obtained in the vineyard can be explained by a first-order rate mass transfer model. A procedure for the collection of wash-off waters based on octadecylsilane solid phase extraction, followed by an analytical method of reverse-phase liquid chromatography with diode array detection is presented for the evaluation of fungicide inputs into soils after being washed off vineyards by rainfalls. Quality parameters of the analytical method yielded good precision (RSD < 10%) and low detection limits (ranging between 1 and 21 microg/L).  相似文献

The aim of this work is to assess the effect of planting patterns on the spatial distribution of total copper and other Cu fractions in vineyard soils. Both classical and geostatistical tools were used for the study. The soil of the plot had a loam texture and was strongly acid. The mean total Cu concentration (CuT) was 368 mg kg−1. The mean value of potential available fractions was 188 mg kg−1 for CuEDTA and 122 mg kg−1 for CuDPTA, whereas the mean exchangeable Cu (CuEX) was 5·2 mg kg−1. All Cu measurements exhibited a wide variation. These values are very high compared with those found in non‐polluted soils, and they can affect the soil, plants and microorganisms. The best correlation for CuEX was with soil pH, whereas for CuEDTA, CuDPTA, and CuT, the best correlation was with soil organic carbon. Directional semivariograms were fitted with a spherical model (parallel to plant rows) and a periodic model (perpendicular) showing a dependence on orientation and distance. All Cu measurements were higher along plant rows than among them, finding a periodic pattern in the variance for the normal direction from plant rows. However, in site‐specific management, it is crucial not only to describe the pattern of variation but also to estimate the Cu content in the soil. Copper concentration maps were estimated by kriging interpolation. These maps show a higher Cu accumulation along the cultivated rows than the uncultivated rows.
