In order to characterise and classify an unknown maize-infecting potyvirus isolated from fields in northeast Spain, the entire coat protein gene and the C-terminal twothirds of the large nuclear inclusion protein (NIb) gene were cloned and sequenced. Protein sequencing enabled the cleavage site between the two proteins to be deduced and also revealed that on storage the viral coat protein undergoes a specific degradation in which the N-terminal 39 amino acids are removed. Comparison of the nucleotide sequence of the 3 non-coding region of the viral RNA and the predicted amino acid sequence of the coat protein with the equivalent regions of other members of the potyvirus group revealed that the Spanish virus is closely related to maize dwarf mosaic virus strain A. 相似文献

We evaluated the effects of the addition of two types of spent mushroom substrate (SMS), SMS from an Agaricus bisporus crop (SMS1) and a mixture of SMSs from an A. bisporus crop and a Pleurotus crop (50% v/v each) (SMS2), on the characteristics of a calcareous clayey‐loam soil and the yield and nutritional status of lettuce (Lactuca sativa L.), relative to crops receiving mineral fertilizer (M) and a control (C) without amendment. The application of SMS, especially SMS1, improved soil fertility compared with C and M soils. Moreover, the use of these organic substrates as soil amendments did not harm the plants and gave yields similar to that obtained with mineral fertilizer. The nutritional contents of the lettuce plants were greater than or similar to those of the plants from treatment C or M, except for the plant tissue concentrations of K, Fe and Zn, which were significantly reduced by SMS application. However, this latter fact did not reduce the lettuce yield in the amended soils. Therefore, the use of SMSs as organic amendments contributes to residue utilization, in an environmentally friendly way, and to improved soil fertility and crop yield. 相似文献

The objective of the present work was to study the effect of annealing and concentration of Ca(OH)2 (lime) and calcium salts on the thermal and rheological properties of maize starch during an ecological nixtamalization process. Thermal and rheological properties of maize starch changed during the ecological nixtamalization process because of three main causes: the annealing phenomenon, type of calcium salt, and calcium salt concentration. In all treatments thermal properties (To, Tp, and Tf) of nixtamal starch increased owing to the annealing process, whereas the type of salt or lime increased thermal properties and decreased pasting properties in this order: CaCl2 > CaSO4 > Ca(OH)2 ≈ CaCO3. This behavior was because of the dissociation of each salt or lime in water. Anions (OH−) can penetrate much more easily into the starch granule and start the gelatinization process by rupturing hydrogen bonds. Additionally, amylose‐lipid complexes were formed during the nixtamalization processes, as indicated by an increasing peak at 4.5 Å in X‐ray diffraction patterns. 相似文献

This study assesses the effect of salinity in bioavailability and toxicity of Zn by means of laboratory bioassays by observing contamination in both sediment and water, accumulation of Zn in biological tissues, and histopathological damage in the gills and guts tissues of Ruditapes philippinarum clams, which were exposed to different types of sediments from the Gulf of Cádiz (SW Spain) as well as two dilutions of toxic mud coming from an accidental mining spill. With this objective, the coefficients of distribution (K D) for Zn between overlying water and sediments were calculated, the histopathological frequencies in the tissues of the gills and guts of clams were determined, and the biota-sediment bioaccumulation factors as well as the bioaccumulation factors were quantified in the different stations. Results showed that the greatest histopathological damages appeared when the salinity values decreased. Statistical results showed that salinity was inversely correlated with histopathological damage (p?<?0.01) for the lesion index for gills. The most outstanding results were observed in the two dilutions of toxic mud (0.3% and 7.9%) at a salinity value of 10. Salinity was inversely correlated with the concentration of Zn in biological tissues (p?<?0.05) and inversely correlated with the concentration of Zn in water and sediment. The muscle composition of a pelagic fish species, Atlantic mackerel (Scomber scombrus), has been studied to determine the relationship with its susceptibility to develop lipid oxidation during chilled storage. For such an aim, the initial concentrations of the major components (water, total lipids, protein, and PUFAs) and minor pro-oxidant and antioxidant components (ascorbic acid, α-tocopherol, hemoglobin, total iron, LMW-iron, copper, and zinc) of different batches of mackerel were characterized. For the study, several batches of mackerel were caught during the spring and summer periods. The different batches were subjected to chilled storage, and the onset of lipid oxidation was statistically related with the initial muscle composition. Results showed significant compositional differences among the mackerel lots, especially for the muscle lipid content (2.83-9.50%). In a first step, a Pearson correlation test was used to check the influence of each component on the progress of lipid oxidation. Results showed a significant relationship between shelf life and water and total lipid contents. Multiple regression was performed to reveal the contribution of each component to the susceptibility to lipid oxidation. The model obtained combines the content of PUFAs, total iron, hemoglobin, and ascorbic acid. An accurate prediction of shelf life in terms of rancidity was achieved by the model created (R(2) = 0.9975). These results establish that the levels of endogenous pro-oxidants and antioxidants present in fish muscle together with the polyunsaturated lipids are relevant factors affecting the shelf life of mackerel muscle. 相似文献

The aim of the work was to study the influence of particle size in the composition, physicochemical, techno-functional and physio-functional properties of two flours obtained from persimmon (Diospyros kaki Trumb. cvs. ‘Rojo Brillante’ (RBF) and ‘Triump’ (THF) coproducts. The cultivar (RBF and THF) and particle size significantly affected all parameters under study, although depending on the evaluated property, only one of these effects predominated. Carbohydrates (38.07–46.98 g/100 g) and total dietary fiber (32.07–43.57 g/100 g) were the main components in both flours (RBF and THF). Furthermore, insoluble dietary fiber represented more than 68% of total dietary fiber content. All color properties studied were influenced by cultivar and particle size. For both cultivars, the lower particle size, the higher lightness and hue values. RBF flours showed high values for emulsifying activity (69.33–74.00 mL/mL), while THF presented high values for water holding capacity (WHC: 9.47–12.19 g water/g sample). The bile holding capacity (BHC) and fat/oil binding values were, in general, higher in RBF (19.61–12.19 g bile/g sample and 11.98–9.07, respectively) than THF (16.12–12.40 g bile/g sample and 9.78–7.96, respectively). The effect of particle size was really evident in both WHC and BHC. Due to their dietary fiber content, techno-functional and physio-functional properties, persimmon flours seem to have a good profile to be used as potential functional ingredient. 相似文献

Summary The severity of late blight tuber rot in five potato cultivars (Green Mountain, Sebago, Dorita, AC Brador and Island Sunshine)
resulting from inoculation with US-1/US-8 combinations of Canadian isolates ofPhytophthora infestans was evaluated. In addition, the potential for mixed (A1∶A2) inoculations to result in oospore formation was examined. In
studies conducted in 1996 and 1997, cv. Dorita (and Island Sunshine in 1997) developed the least amount of surface or internal
necrosis relative to the other cultivars following co-inoculation with US-1 and US-8 isolates ofP. infestans. Differences also were revealed among isolate combinations used for inoculation. The combination of a US-1 isolate from Prince
Edward Island with a US-8 isolate from New Brunswick consistently produced the least amount of necrosis in tubers of the various
cultivars. There was no evidence of oospore production in any of the tubers examined.
