The aim of the study was to examine the effect of boron (B) fertilization on the vegetative and the reproductive responses of apple (Malus domestica Borkh.) trees grown at low soil B availability. The experiment was carried out in 2005 under a greenhouse on 5-year-old ‘Jonagold’ apple trees/M.9 EMLA planted singly in 50-L containers filled with a sandy loam soil with hot water-soluble B concentration of 0.32 mg kg−1. The trees were fertilized with B as foliar or soil application. Foliar B sprays were applied at the stage of pink bud, beginning of flowering, petal fall, and 10 days after flowering, at a solution concentration of 0.03%. Soil B fertilization was done at the bud break stage at a rate of 2 g per tree (27 mg B kg−1 soil). The trees untreated with B served as the control. The results showed that soil B fertilization improved root development and tree vigor. Leaves of trees supplied with B to the soil had higher B concentration and chlorophyll, net photosynthetic rate, stomatal conductance, and activity of catalase and glutathione reductase than those of the control plants. Boron fertilization, regardless of application mode, increased fruit yield; the efficiency of foliar B sprays was higher than soil B application. Apple fruits of trees fertilized with B to the soil were bigger, more colored, richer in B, and had higher soluble solids concentration, and titratable acidity compared to those of the control trees.  相似文献

In this paper, coke wastewater that had passed through biological and integrated membrane processes (filtration on sand bed—reverse osmosis) was chosen to assess the phytotoxicity of selected industrial wastewater with regard to the test plant—Vicia faba. An innovative research technique in vitro test was conducted in a large scale phytothrone chamber on two matrices: cotton and Murashige and Skoog Basal Medium (MSBM). The toxicity of wastewater was evaluated for samples: (1) treated in the treatment plant by biological processes, (2) filtrated through a sand bed and filtrated (3) reverse osmosis (RO) membrane. The results showed that there is a noticeable correlation between increasing concentrations of wastewater and seed germination of the test plant. Although the wastewater collected from the coke plant was treated biologically, it showed very high levels of germination inhibition (90–98% for cotton matrix and 92–100% for MSBM matrix) and strong toxic effects. The wastewater collected from the coke plant showed a significantly greater phytotoxic effect compared with those obtained from the effluent treated on a sand bed and in RO. However, wastewater, even after treatment on a sand bed (reduction of COD—39%, TN—46%, TOC—42%, TC—47%, SS—50%, 16PAHs—53%), was still toxic and germination inhibition was in the range of 24–48% for the cotton matrix and 14–54% for the MSBM matrix. The toxicity of wastewater treated in the membrane process was the lowest (reduction of COD—85%, TN—95%, TOC—85%, TC—86%, SS—98%, 16PAHs—67%). The germination inhibition was in the range of 4–10% for the cotton matrix and 2–12% for the MSBM matrix. These samples are classified as non-toxic or slightly toxic to the model plant. The present study highlights the necessity of monitoring not only the basic physical and chemical indicators (including the level of toxic substances as PAHs), but also their effect on the test organisms in wastewater samples.  相似文献

Defensins are antimicrobial peptides that contribute broadly to innate immunity, including protection of mucosal tissues. Human α-defensin (HD) 6 is highly expressed by secretory Paneth cells of the small intestine. However, in contrast to the other defensins, it lacks appreciable bactericidal activity. Nevertheless, we report here that HD6 affords protection against invasion by enteric bacterial pathogens in vitro and in vivo. After stochastic binding to bacterial surface proteins, HD6 undergoes ordered self-assembly to form fibrils and nanonets that surround and entangle bacteria. This self-assembly mechanism occurs in vivo, requires histidine-27, and is consistent with x-ray crystallography data. These findings support a key role for HD6 in protecting the small intestine against invasion by diverse enteric pathogens and may explain the conservation of HD6 throughout Hominidae evolution.  相似文献

The contents of ascorbate, thiols, and phenolic compounds and antioxidative enzyme activity were measured in the apple peel of 56 genotypes after harvest in two vegetation seasons, 2003 and 2004. The main reason of great interest in these bioactive compounds is their well-established physiological role in all living systems. The biggest differences between tested genotypes were noted for ascorbate peroxidase and glutathione reductase (GR) activity, followed by total ascorbate, phenolics, and glutathione concentration; the least difference was observed in the case of catalase. A large cultivar variation was noted in the anthocyanins and flavonols contents. Distinguishing the cultivars with the lowest, highest, relatively stable or those in which antioxidant content greatly differed depending on growing seasons was attempted. The GR activity is proposed as an environmental stress marker of apple fruit.  相似文献

Two liquid culture systems, rotary shaker (RS) and temporary immersion system (TIS) for the propagation of Rosa rubiginosa and Rosa tomentosa were investigated and compared with solid medium culture. Shoot tips were grown on basic Murashige and Skoog medium with 20 mg dm?3 Fe EDDHA, 1 µM BA, 1.5 µM GA3 and 3% sucrose for six weeks. Of the different culture systems, liquid cultures resulted in better growth and development of roses. Both RS and TIS cultures improved biomass growth, multiplication rate, leaf blade area and plant height; however, RS reduced the shoot dry mass content. There were no changes in the content of chlorophyll a, b, a + b in plants developed in the tested systems. Use of a solid medium favoured the accumulation of phenolic compounds, soluble sugars, and carotenoids, but it was accompanied by browning and necrosis of shoots. Plants propagated in TIS were characterised by the high content of phenolic compounds and soluble sugars, but they had the highest multiplication rate. Use of TIS resulted in eight times the number of shoots for R. tomentosa and twice as many for R. rubiginosa compared to solid medium.  相似文献

Translesion synthesis (TLS) is the major pathway by which mammalian cells replicate across DNA lesions. Upon DNA damage, ubiquitination of proliferating cell nuclear antigen (PCNA) induces bypass of the lesion by directing the replication machinery into the TLS pathway. Yet, how this modification is recognized and interpreted in the cell remains unclear. Here we describe the identification of two ubiquitin (Ub)-binding domains (UBM and UBZ), which are evolutionarily conserved in all Y-family TLS polymerases (pols). These domains are required for binding of poleta and poliota to ubiquitin, their accumulation in replication factories, and their interaction with monoubiquitinated PCNA. Moreover, the UBZ domain of poleta is essential to efficiently restore a normal response to ultraviolet irradiation in xeroderma pigmentosum variant (XP-V) fibroblasts. Our results indicate that Ub-binding domains of Y-family polymerases play crucial regulatory roles in TLS.  相似文献
