Summary Capsaicin contents were measured in the progenies from the test crosses between the strain, G5S17 derived from fruit with the graft-induced change in fruit shape and the original two cultivars, Yatsubusa (Capsicum annuum L. var fasciculatum Ilish) and Spanish Paprika (C. annuum L. var. grossum Sendt) used for grafting.The strain, G5S17 contained a lower amount of capsaicin than the original pungent cultivar Yatsubusa used as scion. The characteristic of the lowered capsaicin content in the strain has been stably inherited through seed propagation, and has also been transmitted to the progenies from the crosses with the original two cultivars used for scion or stock. These features in the capsaicin content agreed with those in fruit shape in the graft-induced change.  相似文献

A labeled avidin-biotin enzyme-linked immunosorbent assay (LAB-ELISA) for detecting antibody to infectious laryngotracheitis (ILT) virus in chicken sera was developed and compared with ordinary ELISA. Purified ILT virus, biotin-labeled anti-chicken IgG rabbit IgG conjugate, and horseradish-peroxidase-labeled avidin were used in the LAB-ELISA. When sera from farm chickens were tested by serum neutralization (SN) and two kinds of ELISA, the correlation rate between SN and LAB-ELISA was 50/50 (100%), and that between SN and ordinary ELISA was 39/50 (78%). In LAB-ELISA, all of the sera that were antibody-negative by SN had low absorbance (A) values (below 0.05), and the A values were closely correlated with the SN indexes. In ordinary ELISA, however, the sera antibody-negative by SN had various A values ranging from 0.06 to 0.32. LAB-ELISA had much lower nonspecific reactions than ordinary ELISA against sera from ILT-negative chickens, even when chickens were 30 weeks old. ILT antibody production after ILT vaccination could be detected by LAB-ELISA. A values peaked 5 weeks postinoculation and were maintained for 17 weeks.  相似文献

Rabbit syphilis was experimentally transmitted from clinical cases to healthy rabbits. The purpose was to evaluate changes in RPR titers during the course of infection and to detect the pathogenic organism. Two of three littermate rabbits were inoculated topically. One rabbit became symptomatic, with typical clinical signs on its genitalia about 8 weeks after inoculation and a marked rise in RPR titers, another remained asymptomatic with a moderate rise in titers, and the control rabbit remained negative. These results supported the specific relationship between clinical signs and RPR titers. Histopathological examination of the skin lesion from the symptomatic rabbit revealed spirochetes.  相似文献

Of the Capsicum peppers (Capsicum spp.), cultivated C. annuum is the most commercially important, but has lacked an intraspecific linkage map based on sequence-specific PCR markers in accord with haploid chromosome numbers. We constructed a linkage map of pepper using a doubled haploid (DH) population derived from a cross between two C. annuum genotypes, a bell-type cultivar ‘California Wonder’ and a Malaysian small-fruited cultivar ‘LS2341 (JP187992)’, which is used as a source of resistance to bacterial wilt (Ralstonia solanacearum). A set of 253 markers (151 SSRs, 90 AFLPs, 10 CAPSs and 2 sequence-tagged sites) was on the map which we constructed, spanning 1,336 cM. This is the first SSR-based map to consist of 12 linkage groups, corresponding to the haploid chromosome number in an intraspecific cross of C. annuum. As this map has a lot of PCR-based anchor markers, it is easy to compare it to other pepper genetic maps. Therefore, this map and the newly developed markers will be useful for cultivated C. annuum breeding.  相似文献

Tea cultivars have been bred by individual selection of landraces and by crossbreeding, but the validation of the parentage is limited. In this study, we performed parentage analysis of 79 tea cultivars in Japan based on SSR markers to confirm or identify the parent-offspring relationships among them. The effectiveness of nine SSR markers for parentage analysis was validated by comparing them to the existing cleaved amplified polymorphic sequence markers. The former markers were detectable more alleles than the latter. Simulation of parentage analysis of the tea cultivars predicted biparental origins for 12 cultivars (‘Houshun’, ‘Mie ryokuhou no. 1’, ‘Surugawase’, ‘Tenmyo’, ‘Yamanoibuki’, ‘Harumidori’, ‘Koushun’, ‘Minekaori’, ‘Okumusashi’, ‘Saemidori’, ‘Sofu’, and ‘Toyoka’), in the first five of which candidate parents of yet-to-be-defined pedigree were newly identified. Comparisons of a total of 41 SSR genotypes confirmed the newly-identified parentages of ‘Asahi’ for ‘Tenmyo’, ‘Rokurou’ for ‘Houshun’, ‘Surugawase’, and ‘Yamanoibuki’, and ‘Yamatomidori’ for ‘Mie ryokuhou no. 1’. The maternity of seven cultivars out of the 12 was also confirmed with chloroplast DNA sequences. Uniparental origins were confirmed for 25 cultivars. This parentage analysis has improved our knowledge of tea pedigrees and will aid in the development of new cultivars.  相似文献

Pattern discrimination by dewinged walking flies (Boettcherisca peregrina) was tested in behavioral experiments. After emergence, the flies were deprived of light or visual patterns. Deprivation impaired the normal development of visual pattern discrimination without impairing phototaxis. Flies kept in a lighted, white, unpatterned environment could not discriminate visual patterns, nor could flies kept in continuous darkness. These results indicate that there is considerable plasticity in the structure of the visual system of these flies.  相似文献

Background

The rice PLASTOCHRON (PLA) genes PLA1 and PLA2 regulate leaf maturation and the temporal pattern of leaf initiation. Although the function of PLA genes in the leaf initiation process has been analyzed, little is known about how they affect leaf growth. Previously, we suggested that PLA1 and PLA2 function downstream of the gibberellin (GA) signal transduction pathway. In the present study, we examined the phenotype of a double mutant of pla and slender rice 1 (slr1), which is a constitutive GA response mutant. By analyzing these double mutants, we discuss the relationship between PLA-related and GA-dependent pathways and the possible function of PLA genes in leaf growth.

Findings

Single slr1 and pla mutants exhibited elongated and dwarf phenotypes in the vegetative stage, respectively. The stature and leaf size of the pla1/slr1 and pla2/slr1 double mutants were intermediate between those of the pla and slr1 single mutants. However, the effects of slr1 on leaf elongation were markedly suppressed in the pla1 and pla2 mutant backgrounds. On the other hand, the change in cell length in the double mutants was almost the same as that in the single mutants. An expression analysis of genes involved in GA biosynthesis and catabolism indicated that feedback regulation functioned normally in the pla/slr1 double mutants.

Conclusions

Our genetic results confirm that PLA genes regulate leaf growth downstream of the GA pathway. Our findings also suggest that PLA1 and PLA2 are partly required for GA-dependent leaf elongation, mainly by affecting cellular proliferation.  相似文献

Rapid plasma reagin (RPR) titers were periodically examined during and after treatment in three rabbits clinically diagnosed with rabbit syphilis. RPR titers remained positive after clinical recovery and then gradually declined. Of the two rabbits inoculated experimentally, one showed clinical signs of the disease, while the other did not. RPR titers were also periodically evaluated before and after inoculation in these two rabbits. The trends in RPR titers reflected the course of infection, both in the spontaneous and in the experimental cases. An inapparent case and cases without clinical signs after clinical recovery showed low titers for long period of time. Useful information for interpretation of RPR titers measured clinically was obtained by this survey.  相似文献

Edamame [Glycine max (L.) Merr.] is a type of soybean selected for fresh or frozen vegetable use at an immature stage. Since edamame has a similar protein content, milder flavor, nuttier texture, and is easier to cook when compared to grain soybean, it is being promoted as a new vegetable for global consumption. Global production will require breeding programs for local adaptation; however, limited research has been published on genetic diversity of edamame varieties for the assessment of genetic resources. Simple sequence repeats (SSRs) were used to study the genetic diversity among 130 accessions, including edamame cultivars and landraces from Japan, China and the US, and also the new breeding lines in the US. Although it is assumed that elite edamame cultivars would have narrow genetic diversity, seventeen SSRs detected polymorphism to distinguish 99 of the 130 accessions. The cluster analysis generated nine clusters and 18 outliers. Genetic diversity within Japanese edamame was lower than that within Chinese vegetable soybean accessions (maodou), even though only 10 Chinese maodou were analyzed compared to 107 Japanese edamame. Cluster analysis revealed that the patterns of SSR diversity in edamame can generally distinguish maturity classes and testa color. We concluded that Japanese edamame have a narrow genetic base different from others and that SSRs can describe the patterns of genetic diversity among the elite vegetable soybean.  相似文献
