Cytolytic and noncytolytic strains of the porcine reproductive and respiratory syndrome virus (PRRSV) were isolated in primary cultures of porcine alveolar macrophages (PAM) from lung homogenates of stillborn fetuses or blood samples of dyspneic piglets collected from Quebec pig farms having experienced acute or chronic outbreaks of PRRS. Serological identification of the virus was confirmed by indirect immunofluorescence and indirect protein A-gold immunoelectron microscopy using reference antiserum prepared from experimentally-infected specific pathogen free (SPF) piglets and monoclonal antibodies (MoAbs) directed against the p15 nucleocapsid (N) protein of the reference ATCC-VR2332 isolate. Intracytoplasmic enveloped viral particles that tended to accumulate into cytoplasmic vesicles were observed in the infected PAM; no budding was demonstrated at the level of the cytoplasmic membrane. The extracellular virions appeared as pleomorphic but mostly spherical enveloped particles, 50-72 nm in diameter (averaged diameter of 50 particles was 58.3 nm), with an isometric core about 25-30 nm. Buoyant density of the virus in CsCL density gradients was estimated to 1.18-1.20 g/mL. No hemagglutinating activity was demonstrated. Analysis of semipurified virions of isolate IAF-exp91 by radioimmunoprecipitation (RIPA) and Western immunoblotting experiments, using reference rabbit and porcine hyperimmune sera, revealed four major viral proteins, a predominant 15 kD N protein and three other proteins with predicted M(r_ of 19, 26 and 42 kD. Progeny viral particles produced in PRRSV-infected PAM in the presence of tunicamycin lacked the 42 kD protein, thus confirming its N-glycosylated nature. Immunoprecipitation experiments using the anti-ATCC-VR2332 MoAbs confirmed the close antigenic relationships between Quebec and American reference isolates of PRRSV. 相似文献

ABSTRACT: Common silver biddy Gerres oyena were collected monthly between November 2002 and November 2005 in Okinawa Island of southern Japan. The seasonal reproductive cycle of this species was investigated using the gonadosomatic index (GSI) and through histological observation of gonads. Temporal variation in the GSI reflected gonadal development from histological observations; ovary development occurred from March to September and testes development occurred between March and August. Maximal development was observed in April and May in both sexes. While mean values for hepatosomatic and lipidosomatic indices decreased markedly in females relative to males during gonadal development, mean condition factors decreased with gonadal recrudescence in both sexes. The minimum standard length (SL) at sexual maturity was 89.7 mm for females and 81.4 mm for males and the size at which 50% of individuals were sexually mature was 104 and 92 mm SL in females and males, respectively. The power relationships of batch fecundity (BF) versus standard length (SL, mm) and body weight (BW, g) of females can be expressed as BF = 46.7 × e1.64 SL and BF = 19 697 × e0.49 BW , respectively. 相似文献

Phosphate rock (PR) was activated via acidulation with HCl, EDTA, and oxalic acid to enhance its reactivity. The release, lateral transport, and uptake of phosphorus (P) along with trace metals from pristine and activated PRs were investigated in a soil micro-block system over a period of 27 days, using wheat (Triticum aestivum L.) plants. Significantly (p < 0.05) higher amounts of available soil P, Fe, Mn, and Zn were released from all the PRs after application to soil within first 9 days of seedling transplantation, while the release of other trace metals (Cd, Co, Cr, Cu, Ni, and Pb) was minimal (<1.2 mg kg?1). On cumulative basis, APR-O (oxalic acid activated PR) was the most efficient amendment releasing 164% more available P, followed by APR-E (EDTA activated PR) releasing 130% more available P, compared to the pristine PR. Similar results were also observed in the release of available Fe, Mn, Zn, and other trace metals. The highest diffusive mass fluxes for available P, Mn, Fe, and Zn in soil were observed after 3 days of seedling transplantation, which reduced subsequently. The uptake of P, Fe, Mn, and Zn by wheat plants was increased by 394%, 715%, 92%, and 91%, respectively, in APR-O application compared to the pristine PR, while it was increased by 280%, 188%, 16%, and 27%, respectively, in APR-E application compared to the pristine PR. Subsequently, APR-O and APR-E amendments resulted in enhanced shoot lengths, root lengths, shoot dry matter, and root dry matter contents of wheat plants. Hence, it was concluded that activation of PR with oxalic acid and EDTA prior to direct soil application may enhance the reactivity of PR and could serve as a cost-effect fertilization strategy for higher wheat crop production. 相似文献

Systemicity of agrochemicals is an advantageous property for controlling phloem sucking insects, as well as pathogens and pests not accessible to contact products. After the penetration of the cuticle, the plasma membrane constitutes the main barrier to the entry of an agrochemical into the sap flow. The current strategy for developing systemic agrochemicals is to optimize the physicochemical properties of the molecules so that they can cross the plasma membrane by simple diffusion or ion trapping mechanisms. The main problem with current systemic compounds is that they move everywhere within the plant, and this non‐controlled mobility results in the contamination of the plant parts consumed by vertebrates and pollinators. To achieve the site‐targeted distribution of agrochemicals, a carrier‐mediated propesticide strategy is proposed in this review. After conjugating a non‐systemic agrochemical with a nutrient (α‐amino acids or sugars), the resulting conjugate may be actively transported across the plasma membrane by nutrient‐specific carriers. By applying this strategy, non‐systemic active ingredients are expected to be delivered into the target organs of young plants, thus avoiding or minimizing subsequent undesirable redistribution. The development of this innovative strategy presents many challenges, but opens up a wide range of exciting possibilities. © 2018 Society of Chemical Industry 相似文献

Gorgostane steroids are isolated from marine organisms and consist of 30 carbon atoms with a characteristic cyclopropane moiety. From the pioneering results to the end of 2021, isolation, biosynthesis, and structural elucidation using 13C-NMR will be used. Overall, 75 compounds are categorized into five major groups: gorgost-5-ene, 5,6-epoxygorgostane, 5,6-dihydroxygorgostane, 9,11-secogorgostane, and 23-demethylgorgostane, in addition to miscellaneous gorgostane. Of the numerous fractions obtained from the crude plant extracts of Chenopodium ambrosioides, Conyza dioscordisis and Convolvulus arvensis, 11 were found to be active against stored grain pests. In this investigation, we have evaluated the effect of sodium fluoride (NaF) on hepatic function in pregnant and lactating mice and their suckling pups. Experiments were carried out on female Wistar mice given 500 ppm sodium fluoride (226 ppm fluoride ion) in their drinking water from the 15th day of pregnancy until day 14 after delivery. All mice were sacrificed on day 14 after parturition. Our results showed a significant decrease in serum levels of total protein and albumin, a marked hypoglycaemia and a significant decline in serum cholesterol and triglyceride levels in fluoride-treated mice and their pups. Whereas globulin and biluribin levels in serum were not significantly changed by NaF treatment. On the other hand, serum transaminase activities (aspartate transaminase; alanine transaminase), which well known as markers of liver function, were elevated indicating hepatic cells’ damage after treatment with fluoride. Lipid peroxidation increased in NaF-treated mice and pups, as revealed by high liver malondialdehyde levels, while serum total antioxidant status showed a significant decline. These biochemical modifications in NaF-treated mice also correspond histologically with extensive ballooning, hepatocellular necrosis and infiltration of mononuclear cells.
