In the United States, yellow starthistle (Centaurea solstitialis) is an annual invasive weed with Mediterranean origins. Malformed plants displaying witches' broom, fasciations, abortion of buds and flower virescence symptoms were observed in central Italy. Attempts to transmit the causal agent from the natural yellow starthistle host to periwinkle by grafting, resulted in typical symptoms of a phytoplasma, i.e. yellowing and shortening of internodes. The detection of phytoplasmas was obtained from both symptomatic yellow starthistle and periwinkle by the specific amplification of their 16S-23S rRNA genes. PCR amplification of extracted DNA from symptomatic plant samples gave a product of expected size. Asymptomatic plants did not give positive results. An amplicon obtained by direct PCR with universal primers P1/P7 was cloned and sequenced. The homology search using CLUSTALW program showed more than 99% similarity with Illinois elm yellows (ILEY) phytoplasma from Illinois (United States) and 97% with Brinjal little leaf (BLL) phytoplasma from India. Digestion of the nested-PCR products with restriction enzymes led to restriction fragment length polymorphism patterns referable to those described for phytoplasmas belonging to the clover proliferation (16S-VI) group. Since this is a previously undescribed disease, the name Centaurea solstitialis virescence has been tentatively assigned to it. This is a new phytoplasma with closest relationships to ILEY and BLL, but distinguishable from them on the basis of 16S rDNA homology, the different associated plant hosts and their geographical origin.  相似文献

This study compared the productive and nutritional parameters of milk from crossbred lactating cows managed on Panicum maximum Jacq. cv. Tanzania and with a diet supplemented with different pressed oilseed cakes. The supplements used were as follows: peanut cake, sunflower cake and palm kernel cake for replacement of soybean meal. Sixteen cows with an average weight of 544 ± 57 kg and producing 8 ± 1.4 L of milk per day were used in this study. The animals were randomly assigned to the treatments according to a Latin square design repeated over time, with four treatments, 16 animals and four experimental periods. Supplementation of the diet with peanut cake, sunflower cake and palm kernel cake compared with soybean meal in the diet of cows did not affect the average daily production or composition of the milk. The palm kernel cake promoted an increase in lauric fatty acids (C12:0) and palmitoleic acids (C16:1) (5.02 and 1.65%, respectively) compared with peanut cake and sunflower cake (4.13 and 4.01%, respectively). The levels of oleic fatty acids (C18:1) were higher for the sunflower cake and palm kernel cake supplements (26.01 and 25.01%, respectively) compared with peanut cake (23.11%). The replacement of soybean meal with sunflower cake and palm kernel cake improved the nutritional quality of the milk, with lower concentrations of saturated fatty acids and higher concentrations of unsaturated fatty acids, without compromising the production or nutritional composition of the milk. © 2015 Japanese Society of Animal Science  相似文献

Application of urban refuse compost to agricultural soil could help to solve municipalities' problems related to the increasing production of waste only if soil property improvement and environmental conservation can be demonstrated. The use of low-pressure tractor tyres is another proposal in modern agriculture for reducing soil compaction. This study thus aimed to detect the effects of both compost and low-pressure tractor tyres on soil loss, runoff, aggregate stability, bulk density, penetrometer resistance and maize (Zea mays L.) yield. A 3-year field experiment was carried out on a hilly (15% slope) clay loam soil in central Italy. Twelve plots (200 m2 each) were monitored with tipping-pot devices for runoff and soil erosion measurement. Treatments were: compost addition (64 Mg ha−1), mineral fertilisation, use of low-pressure tyres, use of traditional tyres, with three replicates, in a fully randomised block design. Compost was applied once at the beginning of the experiment. Runoff reduction due to compost ranged between 7 and 399 m3 ha−1 during seasons, while soil erosion was reduced between 0.2 and 2.4 Mg ha−1. Mean weight diameter (MWD) of stable aggregates, measured on wheel tracks, increased by 2.19 mm, then progressively decreased. Compost significantly increased bulk density by 0.08 Mg m−3 due to its inert fraction content. This effect was less evident in the second and third year, probably due to harrowing. Maize yields were slightly, but significantly, reduced in composted plots by 1.72 Mg ha−1 in the third year. Low-pressure tyres significantly reduced soil loss in the third year by 1 Mg ha−1. Furthermore, they did not significantly influence runoff volumes and soil structural stability. Low-pressure tyres or compost addition were singly able to prevent an increase in penetrometer resistance due to agricultural machinery traffic. Low-pressure tyres increased the maize yield during the 3 years and the difference (0.4 Mg ha−1) became significant in the third year. In conclusion, results show the positive lasting effect of compost in ameliorating soil physical properties and reducing runoff and soil erosion. Low-pressure tyres appear justifiable both for the observed increase of grain production and reduction of soil compaction. This latter effect is, nevertheless, masked by compost addition which is also able to reduce penetrometer resistance. Further research is required to explain the causes of the slight inhibition of grain yield observed when compost was compared with mineral fertilisation.  相似文献

The aim of this study was to evaluate the compacting effect of rubber tracked tractors in comparison to that of the traditional wheeled tractors. Macroporosity, pore shape and size distribution, bulk density, penetration resistance and saturated hydraulic conductivity were analysed in a clay soil (Vertic Cambisol) near Rome (Italy) following one and four passes on the same track of rubber tracked and wheeled tractors of medium power. The soil structure attributes were evaluated by characterising porosity by means of image analysis of soil thin sections prepared from undisturbed samples. Macroporosity decreased in the 0–10 cm layer of compacted soil, particularly after four tractor passes, due to a large reduction in the proportion of elongated pores and in their vertical continuity. The rubber tracked tractor had a more pronounced compaction effect in the surface layer (0–10 cm) than the wheeled tractor both after one and four passes; the latter treatment producing the lowest soil porosity. The same trend was observed for hydraulic conductivity, which showed a highly significant correlation with elongated pores. In the 10–20 cm layer the porosity was significantly decreased following traffic, apart from in the soil under one pass of the rubber tracked tractor. Again in this layer, the lowest values of porosity were found in soil after four passes of the rubber tracked tractor. Single and multiple passes made by the two tractors induced different effects regarding soil penetration resistance and bulk density. Increment ratio of penetration resistance after tractor passes with respect to the control was: 12.5 and 49.9% with the wheeled and 34.4 and 39.8% with the tracked after one and four passes, respectively. Increment ratio of dry bulk density values after tractor passes with respect to the control was 7.9 and 11.7% with the wheeled and 7.5 and 8.3% with the tracked after one and four passes, respectively. The tractor passes transformed the initial subangular blocky structure into a massive structure with sometimes a platy structure in the upper few centimetres. The results indicated that soil compaction following traffic with the rubber tracked tractor was generally the more pronounced. However the compacting effect of this tractor after one pass seemed to be limited to the surface layer only.  相似文献

There is increased use of rubber-tracked tractors for ploughing on clay soil (Vertic Cambisol) in central, south and insular Italy instead of metal-tracked tractors, because they allow travel on public roads. Field tests were carried out on arable soil previously ploughed and harrowed to compare two types of tractors, one rubber-tracked (CAT Challenger Ch 45) and one wheeled (New Holland 8770) in order to establish the compacting effects resulting from 1 and 4 passes of the tractors in the same track. The following parameters were studied: soil penetration resistance, bulk density and its increment ratio, soil shear strength, soil macroporosity and hydraulic conductivity. Multiple passes made by the two tractors induced very similar effects on the soil in regards to soil penetration resistance. Mean values of penetration resistance (0–0.20 m depth) were 1.15 MPa for the rubber-tracked tractor and 1.11 MPa for the wheeled tractor; mean values of penetration resistance (0.21–0.40 m depth) were 1.07 MPa for the rubber-tracked tractor and 1.17 MPa for the wheeled tractor. The decrease in macroporosity, in particular that of elongated pores in the soil surface layer (0–0.10 m depth) was greater in treatments involving the rubber-tracked tractor (from 20.2 to 2.7%) than for the wheeled tractor (from 20.2 to 10.3%). Following traffic of the two tractors, hydraulic conductivity decreased and the following values were found for the five treatments: control, 18.48 mm h−1; wheeled tractor 1 and 4 passes, 11.15 and 7.45 mm h−1, respectively; rubber-tracked tractor 1 and 4 passes, 3.25 and 1.1 mm h−1, respectively. Highly significant correlations between shear strength and dry bulk density, and between hydraulic conductivity and elongated pores and total macroporosity were found. Significant linear relationships between macroporosity and penetration resistance for 1 and 4 passes of both tractors were found in the soil layers (0–0.10 m). A significant difference was found between tractors and for correlations of penetration resistance values above control values. However, in the soil layer (0–0.20 m depth), with respect to the higher degree of macroporosity and low values of penetration resistance, treatments involving wheeled tractor (1 pass) showed a lower degree of soil compaction than was observed after 1 pass of the rubber-tracked tractor.  相似文献

A study was conducted to evaluate a non-destructive method able to predict Magness–Taylor (MT) penetrometric firmness of blood oranges, potentially applicable for on-line selection. The method consisted of multivariate modelling of non-destructive penetrometric measurements performed on intact oranges with a 11.3 mm diameter probe by digital dynamometric equipment. The tests were carried out on 366 blood oranges (Citrus sinensis (L.) Osbeck cv. Tarocco); 60 were used as a control group to verify the non-destructive nature of the tests. Four test factors were used on the fruit: two different levels of non-destructive deformation (1 and 3 mm), at two different deformation speeds (5 and 30 mm s?1). In order to obtain the reference parameter (MT firmness), a destructive penetrometric test followed each non-destructive deformation on the same fruit (deformation of 20 mm at 1 mm s?1). The force–deformation curves of the non-destructive tests were elaborated to calculate different texture parameters (max force, elasticity, work). These parameters with raw curve data (force values for constant deformation steps) were used, after autoscaling pre-processing, as X-block datasets in supervised multivariate modelling (partial least squares) to predict MT firmness values. Among the test factors used, the 30 mm s?1 speed and 3 mm deformation provided the best overall prediction of MT firmness. This comprised an r value of 0.76 and standard error of prevision (SEP) of 2.99 N for a MT firmness mean value of 35.65 N (8.4%). The time needed to apply the selected deformation may lead to a sorting speed of about 10 fruit per second, representing an acceptable value for commercial application.  相似文献

Artificial crystal lattices can be used to tune repulsive Coulomb interactions between electrons. We trapped electrons, confined as a two-dimensional gas in a gallium arsenide quantum well, in a nanofabricated lattice with honeycomb geometry. We probed the excitation spectrum in a magnetic field, identifying collective modes that emerged from the Coulomb interaction in the artificial lattice, as predicted by the Mott-Hubbard model. These observations allow us to determine the Hubbard gap and suggest the existence of a Coulomb-driven ground state.  相似文献
