Summary Aluminum toxicity due to the cation Al+3 is a major factor limiting yields in acid soils. Wide genetic variability to aluminum tolerance is found in oat genotypes. The objectives of this study were to determine the number of genes controlling aluminum tolerance in oats and to verify if any detrimental effects were present of the aluminum tolerance genes on grain yield and grain quality in Al+3free soils. Aluminum tolerance was estimated as the average regrowth of the main root after exposure to toxic levels of Al+3 in a hydroponic solution under controlled conditions. The number of genes controlling that trait was estimated from the distribution of the average root regrowth frequencies in a population of 333 recombinant inbred lines (RIL's) in generations F5:6 and F5:7. The effects on grain yield and grain quality were assessed in a subpopulation of 162 RIL's chosen based on their aluminum tolerance response. Aluminum tolerance in the evaluated population was controlled by one dominant major gene with the tolerant genotypes carying Al a Al a and the sensitive ones al a al a alleles. Avena sativa L. subsp. nudisativa has the ability to produce naked grains. Genetic studies on the naked trait of oat began over a century ago, but the genetic and molecular factors associated with the expression of this trait have not been fully clarified. The objectives of this study were to evaluate the naked trait in two oat populations of recombinant inbred lines (RILs), to determine the number of genes, to estimate the heritability, and to map genomic regions associated with the naked trait in hexaploid oat. Parental lines and RILs of each population were screened for the naked trait from plants grown in the field over a 2 year period. Based on the phenotypic data, the oat RILs were classed as naked, partially naked, partially hulled and hulled. In both populations and years, a great number of RILs showed variable expressivity for the naked trait. The genetic analysis indicated the action of a major gene (N1) with the action of modifying genes controlling the formation of naked grains. The results of the estimate of heritability show that environmental conditions do not have a great influence in determining the naked trait. The quantitative trait loci analysis detected a genomic region with a large effect on the naked trait that explained more than 50% of the phenotypic variation. Further studies are needed to validate the use of these molecular markers to assist breeding programs to select high quality and stable naked oat cultivars.  相似文献

Anthropogenic conversion of primary forest to pasture for cattle production is still frequent in the Amazon Basin. Practices adopted by ranchers to restore productivity to degraded pasture have the potential to alter soil N availability and N gas losses from soils. We examined short-term (35 days) effects of tillage prior to pasture re-establishment on soil N availability, CO2, NO and N2O fluxes and microbial biomass C and N under degraded pasture at Nova Vida ranch, Rondônia, Brazilian Amazon. We collected soil samples and measured gas fluxes in tilled and control (non tilled pasture) 12 times at equally spaced intervals during October 2001 to quantify the effect of tillage. Maximum soil NH4+ and NO3 pools were 13.2 and 6.3 kg N ha−1 respectively after tillage compared to 0.24 and 6.3 kg N ha−1 in the control. Carbon dioxide flux ranged from 118 to 181 mg C–CO2 m2 h−1 in the control (non-tilled) and from 110 to 235 mg C–CO2 m2 h−1 when tilled. Microbial biomass C varied from 365 to 461 μg g−1 in the control and from 248 to 535 μg g−1 when tilled. The values for N2O fluxes ranged from 1.22 to 96.9 μg N m−2 h−1 in the tilled plots with a maximum 3 days after the second tilling. Variability in NO flux in the control and when tilled was consistent with previous measures of NO emissions from pasture at Nova Vida. When tilled, the NO/N2O ratio remained <1 after the first tilling suggesting that denitrification dominated N cycling. The effects of tilling on microbial parameters were less clear, except for a decrease in qCO2 and an increase in microbial biomass C/N immediately after tilling. Our results suggest that restoration of degraded pastures with tillage will lead to less C matter, at least initially. Further long-term research is needed.  相似文献

The aim of this study was to define the differences between the Brazilian states that export and do not export meat to the European Union (EU) and to identify the variables that are important to meet the export requirements. Infrastructure and computerization of the control of animal transit in Brazil that impact on regional health status were evaluated and linked to other variables such as status for foot and mouth disease (FMD) and qualification to export meat to EU. Variables related to transit control of bovines implemented by the state agencies of animal health and inspection in each Brazilian state were evaluated. Using a discriminant analysis, four variables were selected that explained the variation between Brazilian states that were “free” and “not free” of FMD while another four were selected to explain the variation between the zones “approved” and “not approved” to export meat to the EU, including number of official veterinarians, total transit of bovines and buffaloes, total number of animal transit certificates issued for bovine and buffaloes at the state or zone level, and total number of municipalities in the state or zone. It was possible to correctly discriminate between “free” and “not free” FMD states or zones. Variables related to animal transit are important in assessing the state for the classification of animal health situation and for EU approval for the exportation of meat.  相似文献

Understanding spatial and temporal patterns in land susceptibility to wind erosion is essential to design effective management strategies to control land degradation. The knowledge about the land surface susceptible to wind erosion in European contexts shows significant gaps. The lack of researches, particularly at the landscape to regional scales, prevents national and European institutions from taking actions aimed at an effective mitigating of land degradation. This study provides a preliminary pan‐European assessment that delineates the spatial patterns of land susceptibility to wind erosion and lays the groundwork for future modelling activities. An Index of Land Susceptibility to Wind Erosion (ILSWE) was created by combining spatiotemporal variations of the most influential wind erosion factors (i.e. climatic erosivity, soil erodibility, vegetation cover and landscape roughness). The sensitivity of each input factor was ranked according to fuzzy logic techniques. State‐of‐the‐art findings within the literature on soil erodibility and land susceptibility were used to evaluate the outcomes of the proposed modelling activity. Results show that the approach is suitable for integrating wind erosion information and environmental factors. Within the 34 European countries under investigation, moderate and high levels of land susceptibility to wind erosion were predicted, ranging from 25·8 to 13·0 M ha, respectively (corresponding to 5·3 and 2·9% of total area). New insights into the geography of wind erosion susceptibility in Europe were obtained and provide a solid basis for further investigations into the spatial variability and susceptibility of land to wind erosion across Europe. © 2014 The Authors. Chromosome 17 deletions and p53 gene mutations in colorectal carcinomas   总被引:175,自引:0,他引:175  
Previous studies have demonstrated that allelic deletions of the short arm of chromosome 17 occur in over 75% of colorectal carcinomas. Twenty chromosome 17p markers were used to localize the common region of deletion in these tumors to a region contained within bands 17p12 to 17p13.3. This region contains the gene for the transformation-associated protein p53. Southern and Northern blot hybridization experiments provided no evidence for gross alterations of the p53 gene or surrounding sequences. As a more rigorous test of the possibility that p53 was a target of the deletions, the p53 coding regions from two tumors were analyzed; these two tumors, like most colorectal carcinomas, had allelic deletions of chromosome 17p and expressed considerable amounts of p53 messenger RNA from the remaining allele. The remaining p53 allele was mutated in both tumors, with an alanine substituted for valine at codon 143 of one tumor and a histidine substituted for arginine at codon 175 of the second tumor. Both mutations occurred in a highly conserved region of the p53 gene that was previously found to be mutated in murine p53 oncogenes. The data suggest that p53 gene mutations may be involved in colorectal neoplasia, perhaps through inactivation of a tumor suppressor function of the wild-type p53 gene.  相似文献

Two primers, specific for Phytophthora nicotianae (Pn6) and P. citrophthora (Pc2B), were modified to obtain Scorpion primers for real-time identification and detection of both pathogens in citrus nursery soils and roots. Multiplex PCR with dual-labelled fluorogenic probes allowed concurrent identification of both species ofPhytophthora among 150 fungal isolates, including 14 species of Phytophthora. Using P. nicotianaespecific primers a delayed and lower fluorescence increase was also obtained from P. cactorumDNA. However, in separate real-time amplifications, the aspecific increase of fluorescence from P. cactorum was avoided by increasing the annealing temperature. In multiplex PCR, with a series of 10-fold DNA dilutions, the detection limit was 10 pg l-1 for P. nicotianaeand 100 pg l–1 for P. citrophthora, whereas in separate reaction DNA up to 1 pg l-1 was detected for both pathogens.Simple and rapid procedures for direct DNA extraction from soil and roots were utilised to yield DNA whose purity and quality was suitable for PCR assays. By combining these protocols with a double amplification (nested Scorpion-PCR) using primers Ph2-ITS4 amplifying DNA from the main Phytophthora species (first round) and PnB5-Pn6 Scorpion and Pc2B Scorpion-Pc7 (second round), it was possible to achieve real-time detection of P. nicotianaeand P. citrophthora from roots and soil. The degree of sensitivity was similar to that of traditional detection methods based on the use of selective media. The analyses of artificially and naturally infested soil showed a high and significant correlation between the concentration of pathogen propagules and the real-time PCR cycle threshold.  相似文献
