The relationship of passage-induced mutant genes 1 and 71 of an attenuated equine herpesvirus 1 (EHV-1) with virulence was analysed by constructing nine recombinant EHV-1 viruses by homologous recombination. Gene 1 or/and gene 71 of a virulent EHV-1 strain, HH1, was replaced by a mutant gene 1 or/and 71 of an attenuated HH1 strain, BK343, respectively. The beta-galactosidase gene of Escherichia coli was inserted within the gene 1 or 71 coding sequence of HH1 to inactivate the genes. Virus replications of these recombinant viruses in cell cultures were similar, but release of the gene 71-inactivated virus from infected cells was delayed compared to that of the other viruses. Plaque sizes of the recombinant viruses were similar to those of HH1, but those of BK343 were significantly smaller, indicating an effect of some unknown factor(s) on viral cell-to-cell spread. The growth abilities of the recombinant viruses with a mutant gene 1 or/and 71 in lungs of mice were similar to those of HH1, but those of gene 71-inactivated viruses were reduced to the level of BK343 and the titers were about 100-times lower than those of the other recombinant viruses. These results indicate that the mutant genes 1 and 71 of BK343 might not confer an attenuated nature to EHV-1.  相似文献

The objectives of this study were to estimate genetic parameters and to perform a genome‐wide association study (GWAS) for predicted methane‐related traits in Japanese Black steers. The methane production and yield traits were predicted using on‐farm measurable traits, such as dry matter intake and average daily gain. A total of 4,578 Japanese Black steers, which were progenies of 362 sires genotyped with imputed 551,995 single nucleotide polymorphisms (SNPs), had phenotypes of predicted methane‐related traits during the total fattening period (52 weeks). For the estimation of genetic parameters, the estimated heritabilities were moderate (ranged from 0.57 to 0.60). In addition, the estimated genetic correlations of methane production traits with most of carcass traits and feed‐efficiency traits were unfavorable, but those of methane yield traits were favorable or low. For the GWAS, no genome‐wide significant SNP was detected, but a total of four quantitative trait locus (QTL) regions that explained more than 5.0% of genetic variance were localized on the genome, and some candidate genes associated with growth and feed‐efficiency traits were located on the regions. Our results suggest that the predicted methane‐related traits are heritable and some QTL regions for the traits are localized on the genome in Japanese Black steers.  相似文献

We examined the relationship between the annual escapement of salmon and the δ 15N of willow (Salix spp.) leaves to evaluate the contribution of marine-derived nutrients (MDN) to riparian vegetation around the Pacific Northwest and Northeast regions. Foliar δ 15N values ranged from −3.42‰ to 4.65‰. The value increased with increasing density of carcasses up to 500 fish/km and 1500 fish/km. δ 15N values were variable at carcass densities below 500 fish/km. Possible factors affecting the fluctuation of δ 15N at reference sites are: (1) denitrification; (2) the presence of N2-fixing trees, such as alder; and (3) agricultural runoff. δ 15N values at the sites with carcass densities over 500 fish/km were consistently high, while a value of δ 15N below zero was observed at only one site (Rusha River; δ 15N = −1.87‰). At this site, most adult pink salmon returned to limited locations near the estuary because steeper channel gradients acted as a migration barrier, resulting in the negative δ 15N value. Nevertheless, we concluded that our results showed evidence of the feedback of MDN to terrestrial vegetation, although the use of the δ 15N value as a terrestrial end member at spawning sites is limited. If the relationship between the enrichment index, which is expressed as the values using a mixing model, and salmon abundance was estimated, the availability of MDN in riparian ecosystems could possibly be evaluated and will lead to the establishment of escapement goals. The purpose of this study was to find whether the concentration of N-acetylglutamate and ornithine transport into mitochondria would regulate urea synthesis when the dietary protein quality was manipulated. Experiments were done on three groups of rats given diets containing 10 g of gluten, 10 g of casein, or 10 g of whole egg protein/100 g for 10 days. The plasma concentration and urinary excretion of urea, the liver concentration and synthesis of N-acetylglutamate, the liver concentrations of glutamate and lysine, and the liver ornithine transport into mitochondria increased with the decrease in quality of dietary protein. A reverse correlation was observed between the activities of urea cycle enzymes, the plasma concentration of arginine, and urinary excretion of urea under these conditions. N-Acetylglutamate concentration and ornithine transport into mitochondria in the liver were closely correlated with the excretion of urea. These results suggest that greater N-acetylglutamate concentration and ornithine transport into isolated mitochondria in the liver of rats, given the lower quality of protein, stimulate urea synthesis and that the concentrations of glutamate and lysine in the liver are at least partly related to the hepatic N-acetylglutamate synthesis and ornithine transport, respectively.  相似文献

The objectives of this study were to better understand the genetic architecture and the possibility of genomic evaluation for feed efficiency traits by (i) performing genome‐wide association studies (GWAS), and (ii) assessing the accuracy of genomic evaluation for feed efficiency traits, using single‐step genomic best linear unbiased prediction (ssGBLUP)‐based methods. The analyses were performed in residual feed intake (RFI), residual body weight gain (RG), and residual intake and body weight gain (RIG) during three different fattening periods. The phenotypes from 4,578 Japanese Black steers, which were progenies of 362 progeny‐tested bulls and the genotypes from the bulls were used in this study. The results of GWAS showed that a total of 16, 8, and 12 gene ontology terms were related to RFI, RG, and RIG, respectively, and the candidate genes identified in RFI and RG were involved in olfactory transduction and the phosphatidylinositol signaling system, respectively. The realized reliabilities of genomic estimated breeding values were low to moderate in the feed efficiency traits. In conclusion, ssGBLUP‐based method can lead to understand some biological functions related to feed efficiency traits, even with small population with genotypes, however, an alternative strategy will be needed to enhance the reliability of genomic evaluation.  相似文献

Nematodes of the family Heligmonellidae (Heligmosomoidea; Trichostrongylina) reside in the digestive tracts of rodents and lagomorphs. Although this family contains large numbers of genera and species, genetic information on the Heligmonellidae is very limited. We collected and isolated adult worms of three species in Japan that belong to the family Heligmonellidae, namely Heligmonoides speciosus (Konno, 1963) Durette-Desset, 1970 (Hs) from Apodemus argenteus, Orientostrongylus ezoensis Tada, 1975 (Oe) from Rattus norvegicus and Lagostrongylus leporis (Schulz, 1931) (Ll) from Pentalagus furnessi, and sequenced the entire internal transcribed spacer regions, ITS-1 and ITS-2 of ribosomal DNA. ITS-1 of Hs, Oe and Ll was 426, 468 and 449 bp in length, and had a G+C content of about 41, 41 and 37 %, respectively. ITS-2 of Hs, Oe and Ll was 297, 319 and 276 bp in length and had a G+C content of about 38, 40 and 28%, respectively. The data of Hs, Oe and Ll were compared with those of two other known species within the family Heligmonellidae, Calorinensis minutus (Dujardin, 1845) (Cm) and Nippostrogylus brasiliensis (Travassos, 1914) (Nb), and with those of two species of Heligmosomidae (Heligmosomoidea), Heligmosomoides polygyrus bakeri and Ohbayashinema erbaevae. Phylogenetic analysis placed Hs, Oe and Ll in the same clade with Cm and Nb, forming a Heligmonellidae branch in both ITS-1 and ITS-2, separate from the Heligmosomoidea branch. These results demonstrated that the ITS-1 and ITS-2 sequences are useful for differentiating the Heligmonellidae nematode species. This study is the first to describe the ITS-1 and ITS-2 sequences of Hs, Oe and Ll.  相似文献

Since 2011, high mortality rates and symptoms consistent with vibriosis have been observed in farmed amberjack (Seriola dumerili) in Japan. To identify 41 strains isolated from diseased amberjack, a multilocus sequence analysis using nine concatenated genes (ftsZ, gapA, gyrB, mreB, pyrH, recA, rpoA, topA and 16S rRNA) was conducted. Twenty‐seven strains were identified as Vibrio harveyi, suggesting an epidemic of V. harveyi infection in amberjack farms. Other strains were identified as Vibrio anguillarum, Vibrio owensii and Photobacterium damselae subsp. damselae. To develop an efficient diagnostic method for vibriosis in amberjack, a multiplex PCR system was developed to identify V. anguillarum, V. harveyi and P. damselae subsp. damselae. The method successfully discriminated between these three bacterial species, with amplification products of 350 bp for V. anguillarum, 545 bp for V. harveyi and 887 bp for Pdamselae subsp. damselae and can be used for diagnosis in aquaculture farms.  相似文献

An indwelling ruminal pH system has been used for the continuous recording of ruminal pH to evaluate subacute ruminal acidosis (SARA) in dairy cows. However this system does not allow the field application. The objective of this study was to develop a new radio transmission pH measurement system, and to assess its performance and usefulness in a continuous evaluation of ruminal pH for use on commercial dairy farms. The radio transmission pH measurement system consists of a wireless pH sensor, a data measurement receiver, a relay unit, and a personal computer installed special software. The pH sensor is housed in a bullet shaped bolus, which also encloses a pH amplifier circuit, a central processing unit (CPU) circuit, a radio frequency (RF) circuit, and a battery. The mean variations of the measurements by the glass pH electrode were +0.20 (n = 10) after 2 months of continuous recording, compared to the values confirmed by standard pH solutions for pH 4 and pH 7 at the start of the recording. The mean lifetime of the internal battery was 2.5 months (n = 10) when measurements were continuously transmitted every 10 min. Ruminal pH recorded by our new system was compared to that of the spot sampling of ruminal fluid. The mean pH for spot sampling was 6.36 ± 0.55 (n = 96), and the mean pH of continuous recording was 6.22 ± 0.54 (n = 96). There was a good correlation between continuous recording and spot sampling (r = 0.986, P < 0.01). We also examined whether our new pH system was able to detect experimentally induced ruminal acidosis in cows and to record long-term changes in ruminal pH. In the cows fed acidosis-inducing diets, the ruminal pH dropped markedly during the first 2 h following the morning feeding, and decreased moreover following the evening feeding, with many pulse-like pH changes. The pH of the cows showed the lowest values of 5.3-5.2 in the midnight time period and it recovered to the normal value by the next morning feeding. In one healthy periparturient cow, the circadian changes in ruminal pH were observed as a constant pattern in the pre-parturient period, however that pattern became variable in the post-partum period. The frequency of the ruminal pH lower than 5.5 increased markedly 3 and 4 days after parturition. We demonstrated the possible application of a radio transmission pH measurement system for the assessment and monitoring of the ruminal pH of cows. Our new system might contribute to accurate assessment and prevention of SARA.  相似文献

The target antigen of autoantibodies in human pemphigus foliaceus (PF) is a desmosomal cadherin, desmoglein 1 (Dsg1). It was demonstrated by immunoelectron microscopy (IEM) that the location of the binding sites of PF autoantibodies in the human epidermis was the extracellular regions of the desmosomes. Only a limited number of canine PF sera were shown to react with canine Dsg1, and the target proteins have not yet been identified. The purpose of this study was to demonstrate the ultrastructural binding site of canine PF autoantibodies to the canine skin by two kinds of IEM methods. Three canine PF sera, which were shown to react with the keratinocyte cell surface by immunofluorescence, were tested in this study. Using a technique of immunoprecipitation-immunoblotting, one out of the three canine PF sera were shown to react with recombinant canine Dsg1. By post-embedding IEM using cryofixation technique, one serum, which did not react with canine Dsg1 by immunoprecipitation-immunoblotting, bound broadly to the extra- and intracellular regions of the desmosomes of normal canine skin. By pre-embedding IEM using canine cultured keratinocytes (MCA-B1 cells), the autoantibodies of all three canine PF sera were identified to be bound to the cell-cell contact area of the adjacent cytoplasmic projections. When double stained with human PF serum and canine PF sera, the binding sites of both human and canine autoantibodies were co-localized on the MCA-B1 cells where the cytoplasmic projections contacted each other. Therefore, it may be concluded that the serum antibodies of canine PF targeted desmosomal proteins, regardless of whether or not they react with canine Dsg1 by immunoprecipitation-immunoblotting method.  相似文献
