Context
Since the 2003 drought and heat wave, Douglas-fir dieback has been reported in France in trees older than 30 years. Consequently, it is questioned whether selected Douglas-fir provenances are suited to the frequent and severe drought events which are forecast due to climate change.Aims
Our objective was to contribute to the screening of variability in productivity and growth response to soil-water deficit of mature trees from provenances not currently used for plantation in France.Methods
We sampled 22 provenances, including coastal and interior Douglas-fir, covering a wide part of its natural distribution, from Oregon to California for coastal provenances and from British Columbia to New Mexico for interior provenances. These provenances were planted at the mid 1970s in two provenance trials located in the south-west area of France. Variability of productivity, of wood density, and of radial growth in response to drought episodes among provenances was quantified and related to soil-water deficit computed by daily water balance calculations.Results
Whatever the provenance, annual radial growth is highly dependent on local soil-water deficit (Felines R 2 = 0.57, Le Treps R2 = 0.49). Radial growth and wood properties exhibit large differences between provenances at 30 years old. Variability between provenances for all wood characteristics studied is mainly structured geographically. Coastal provenances perform best for productivity at 30 years old (619 cm2?±?59), and exhibit a small growth reduction in 2004, the second successive year of drought (?10.7 %?±?3.8). Surprisingly, the southern interior provenances from the driest environments in the natural range show a large growth reduction in 2004 (?30.5 %?±?5.2).Conclusions
The provenances tested exhibited significant differences in growth performance and growth reduction induced by the soil-water deficit. The approach coupling retrospective analysis of radial growth on mature trees and water balance modelling is relevant for evaluating provenance adaptation to more frequent or severe drought episodes. 相似文献2.

Botrytis cinerea is a major crop pathogen infesting >220 hosts worldwide. A cryptic species has been identified in some French populations but the new species, B. pseudocinerea, has not been fully delimited and established. The aim of this study was to distinguish between the two species, using phylogenetic, biological, morphological, and ecological criteria. Multiple gene genealogies confirmed that the two species belonged to different, well-supported phylogenetic clades. None of the morphological criteria tested (spore size, germination rate, or mycelial growth) was able to discriminate between these two species. Sexual crosses between individuals from the same species and different species were carried out. Only crosses between individuals from the same species were successful. Moreover, population genetics analysis revealed a high level of diversity within each species and a lack of gene flow between them. Finally, a population survey over time showed that B. cinerea was the predominant species but that B. pseudocinerea was more abundant in spring, on floral debris. This observation could not be explained by temperature adaptation in tests carried out in vitro or by aggressiveness on tomato or bean leaves. This study clearly establishes that B. cinerea and B. pseudocinerea constitute a complex of two cryptic species living in sympatry on several hosts, including grapevine and blackberry. We propose several biological or molecular tools for unambiguous differentiation between the two species. B. pseudocinerea probably makes a negligible contribution to gray mold epidemics on grapevine. This new species has been deposited in the MycoBank international database. 相似文献

A new method for encapsulation of micronutrients was successfully developed. The encapsulation matrix consisted of two polymers (alginate and chitosan), which were functionalized by acylation with palmitoyl chloride. The structural modifications of polymers were confirmed by Fourier transform infrared (FTIR) spectroscopy. Beads were formed by ionic gelation, and their mechanical and physical characteristics (puncture strength and deformation, viscoelasticity, water vapor permeability, and rate of gel swelling) were evaluated using beads or films made of bead-forming solutions. Functionalization increased elasticity and water impermeability of polymer films. Stability of selected encapsulated micronutrients (ferrous fumarate, ascorbic acid, and beta-carotene) was also evaluated under two levels of temperature (23 and 45 degrees C) and relative humidity (56 and 100%) for 6 months. Encapsulation strongly increased the stability of micronutrients. No difference was observed in the encapsulated micronutrients' stability between nonfunctionalized and functionalized beads. Finally, a release study in gastrointestinal media was conducted. Results showed that beads were not susceptible to enzymatic and acidic attacks during stomach transit. This research demonstrates the potential of a new encapsulation method to protect bioactive molecules from temperature, moisture, and acidic conditions. 相似文献

Objective
To develop a modified ultrasound-guided parasacral approach to the sciatic nerve and compare the effects of a volume of 0.2 mL kg–1 of 0.5% levobupivacaine with an equivalent volume of 0.9% saline injected near the sciatic nerve.Study design
Cadaveric and experimental, blinded, randomized study.Animals
Seven canine cadavers and seven experimental Beagle dogs.Methods
Both sciatic nerves of seven cadavers were identified using a modified in-plane ultrasound-guided approach. Methylene blue solution (0.2 mL) was injected perineurally, and success was evaluated through dissection. The same approach was repeated in seven Beagle dogs sedated with dexmedetomidine (50 μg kg–1) injected intramuscularly (IM). After randomization, 0.2 mL kg–1 of 0.5% levobupivacaine (limb L) and 0.2 mL kg–1 of 0.9% saline (limb C) were injected perineurally on either right or left limb. Block success was determined by sensory deficits every hour for 8 hours after an atipamezole injection (0.2 mg kg–1) IM. Reaction to pinprick (binary score) over the course of the sciatic nerve (four locations) and locomotion were assessed.Results
The overall sciatic nerve block success was 93% in cadavers and 86% in sedated dogs. It was impossible to localize the sciatic nerves in one obese sedated dog. Significant differences between limb L and limb C were observed for pinprick at great trochanter, caudal thigh and lateral tarsal joint (p < 0.0001). Reaction to pinprick was absent in all dogs at great trochanter and caudal thigh up to at least 3 hours on limb L. Locomotion was impaired in all but one dog for 60 (30–210) minutes (median; interquartile range). No complications were observed.Conclusions and clinical relevance
A volume of ≥0.2 mL kg–1 and a concentration of 0.5% levobupivacaine can be recommended when using a modified ultrasound-guided parasacral approach to the sciatic nerve in dogs. 相似文献6.

Background
In order to maintain high yields while saving water and preserving non-renewable resources and thus limiting the use of chemical fertilizer, it is crucial to select plants with more efficient root systems. This could be achieved through an optimization of both root architecture and root uptake ability and/or through the improvement of positive plant interactions with microorganisms in the rhizosphere. The development of devices suitable for high-throughput phenotyping of root structures remains a major bottleneck.Results
Rhizotrons suitable for plant growth in controlled conditions and non-invasive image acquisition of plant shoot and root systems (RhizoTubes) are described. These RhizoTubes allow growing one to six plants simultaneously, having a maximum height of 1.1 m, up to 8 weeks, depending on plant species. Both shoot and root compartment can be imaged automatically and non-destructively throughout the experiment thanks to an imaging cabin (RhizoCab). RhizoCab contains robots and imaging equipment for obtaining high-resolution pictures of plant roots. Using this versatile experimental setup, we illustrate how some morphometric root traits can be determined for various species including model (Medicago truncatula), crops (Pisum sativum, Brassica napus, Vitis vinifera, Triticum aestivum) and weed (Vulpia myuros) species grown under non-limiting conditions or submitted to various abiotic and biotic constraints. The measurement of the root phenotypic traits using this system was compared to that obtained using “classic” growth conditions in pots.Conclusions
This integrated system, to include 1200 Rhizotubes, will allow high-throughput phenotyping of plant shoots and roots under various abiotic and biotic environmental conditions. Our system allows an easy visualization or extraction of roots and measurement of root traits for high-throughput or kinetic analyses. The utility of this system for studying root system architecture will greatly facilitate the identification of genetic and environmental determinants of key root traits involved in crop responses to stresses, including interactions with soil microorganisms.7.

Botrytis bunch rot of grapes is mainly controlled by applying fungicides at three crop growth stages: the end of flowering (BBCH 68), bunch closure (BBCH 77) and the beginning of veraison (BBCH 81). The hydroxyanilide derivative fenhexamid is among the most effective fungicides registered to control Botrytis cinerea. Its effectiveness was examined in relation to spray timing, fungicide resistance and defense responses of grapevine. Overall, the earlier fenhexamid was applied, the more effective it was at controlling B. cinerea. Frequencies of B. cinerea strains which were resistant to fungicides were evaluated at harvest. The frequencies of resistant phenotypes were similar among treatments and years with the exception of a class of multidrug resistant strain (MDR 2) whose frequency appeared to increase after fenhexamid applications. If current spray programs including fenhexamid appear to control bunch rot at the current MDR frequency, a propagation of MDR 2 strains might lead to a decline in disease control. Finally, defense responses were studied in grapevine flowers/berries following fenhexamid application. None of the defense processes tested was induced in flowers/berries at stages 68 and 77. Only an increase in chitinase activity was observed in treated-berries at stage 81, suggesting that fenhexamid effectiveness was not related to a stimulation of defense responses. 相似文献

Current yeast interactome network maps contain several hundred molecular complexes with limited and somewhat controversial representation of direct binary interactions. We carried out a comparative quality assessment of current yeast interactome data sets, demonstrating that high-throughput yeast two-hybrid (Y2H) screening provides high-quality binary interaction information. Because a large fraction of the yeast binary interactome remains to be mapped, we developed an empirically controlled mapping framework to produce a "second-generation" high-quality, high-throughput Y2H data set covering approximately 20% of all yeast binary interactions. Both Y2H and affinity purification followed by mass spectrometry (AP/MS) data are of equally high quality but of a fundamentally different and complementary nature, resulting in networks with different topological and biological properties. Compared to co-complex interactome models, this binary map is enriched for transient signaling interactions and intercomplex connections with a highly significant clustering between essential proteins. Rather than correlating with essentiality, protein connectivity correlates with genetic pleiotropy. 相似文献
