Genetic diversity was estimated among 42 U.S. PlantIntroduction (PI) accessions of the genusCitrullus (of these, 34 PIs are reported tohave disease resistance), and 5 watermelon cultivars, using 30RAPD primers. These primers produced 662 RAPD markers that could berated with high confidence. Based on these markers, geneticsimilarity coefficients were calculated and a dendrogram wasconstructed using the unweighted pair-group method witharithmetic average (UPGMA). The analysis delineated threemajor clusters. The first cluster consisted of a group of fivewatermelon cultivars, a group of C.lanatus var. lanatusaccessions, and a group of C.lanatus var. lanatusaccessions that contained some C.lanatus var. citroidesgenes. The second cluster consisted of the C.lanatus var. citroidesaccessions, while the third cluster consisted of theC. colocynthis accessions.The two C. lanatus clustersdifferentiated from each other and from the C.colocynthis cluster at the level of 58.8%and 38.9% genetic similarity, respectively. Assessment ofgenetic diversity among accessions that have been reported to havedisease resistance indicated that resistance to either anthracnose,downy mildew, powdery mildew, or watermelon mosaic virus is foundamong all major groups of Citrullus PIs.Additionally, resistance to gummy stem blight or Fusarium wilt mayexist among C. lanatus var.citroides PIs. This study demonstrates thatmolecular markers can be useful in assessing genetic diversity, andin sorting Citrullus PIs into phylogeneticgroups prior to their evaluation for disease or pestresistance.  相似文献

A cucurbit species named Praecitrullus fistulosus (Stocks) Pangalo, which thrives in India, is considered to be a distant relative of watermelon. Recent experiments indicated that it has mild resistance to whiteflies (Bemisia tabaci). However, our attempts to cross various US plant introductions (PIs) of P. fistulosus with watermelon or other Citrullus PIs have not been successful. Thus, to determine genetic relatedness among those species, phylogenetic analysis [based on simple sequence repeat (SSR)–anchored (also termed ISSR), and randomly amplified polymorphic DNA (RAPD) markers] was conducted among PIs of P. fistulosus, Citrullus lanatus var. lanatus (watermelon), C. lanatus var. citroides and the wild Citrullus colocynthis. Phylogenetic relationships were also examined with Cucumis melo (melon), Cucumis sativus (cucumber), and wild Cucumis species including C. africanus, C. metuliferus, C. anguria, C. meeusei, and C. zeyheri. Wide genetic distance exists between Citrullus and Cucumis groups (8% genetic similarity). Phylogenetic relationships among Citrullus species and subspecies are closer (25–55% genetic similarity) as compared with those among most Cucumis species (14–68% genetic similarity). P. fistulosus appeared to be distant from both Cucumis and Citrullus species (genetic similarity between P. fistulosus and Cucumis or Citrullus groups is less than 3%). Although wide genetic differences and reproductive barriers exist among cucurbit species examined in this study, they are still considered as potential germplasm source for enhancing watermelon and melon crops using traditional breeding and biotechnology procedures.  相似文献

Inoculum of postharvest pathogens can accumulate inside storage rooms and contaminate new batches of fruit and vegetables, but this chain can be broken by disinfecting storage facilities between storage periods. Quaternary ammonium (QA) has been known for over 50 years as an efficient disinfectant against microorganisms, with wide applications in the food industry. The aim of this study was to determine the efficacy of didecyldimethylammonium chloride (Sporekill, designated here as QAsk), against development of Botrytis cinerea after direct exposure or by ultrasonic fogging. Following direct exposure to a concentration of QAsk below 5 mg L?1, a population of 104 conidia of B. cinerea was inactivated after 2 min; ultrasonic fogging with QAsk at 500 mg L?1 took 30 min to achieve consistent inactivation. Fumigation at 20 °C was considerably more effective than fumigation at 5 °C, and similar results were obtained for three other postharvest pathogens, Penicillium expansum, Colletotrichum gloeosporioides and Alternaria alternata. These results show that conidia of B. cinerea are highly sensitive to direct exposure to QAsk, but that effective sanitation of a storage facility by ultrasonic fumigation requires a QAsk concentration two orders of magnitude greater.  相似文献

Pharmacokinetic studies of soybean isoflavones have shown that following oral ingestion, the two major isoflavones, daidzin and genistin, are hydrolyzed in the intestine, rapidly absorbed into the peripheral circulation, and eliminated from the body with a terminal half-life of 7-8 h. These characteristics make maintenance of steady-state plasma isoflavone concentrations difficult to attain unless there is repeated daily ingestion of foods or supplements containing isoflavones. In an attempt to sustain more constant plasma isoflavone concentrations, a new slow-release formulation of a soybean isoflavone extract was prepared by microencapsulation with a mixture of hydroxypropylcellulose and ethylcellulose to alter its dissolution characteristics. In vitro experiments confirmed slow aqueous dissolution of isoflavones from this formulation when compared with the conventional isoflavone extract. The pharmacokinetics of this slow-release isoflavone extract was studied in 10 healthy postmenopausal women after oral administration of a single capsule containing the equivalent of 22.3 mg of genistein and 7.47 mg of daidzein expressed as aglycons. A comparison of the key pharmacokinetic parameters obtained in this study with those established in extensive studies performed previously in this laboratory indicated that the mean residence time of genistein and daidzein increased 2-fold with microencapsulation. These findings are indicative of a decreased rate of absorption, consistent with the observed slow in vitro dissolution rate. These findings show that it is feasible to employ polymer matrices that slow the aqueous dissolution for preparing sustained-release formulations of soy isoflavones. Further studies to optimize such formulations are warranted.  相似文献

Production of spring potato (Solanum tuberosum L. cv. Désirée) on a deep sandy soil in the central highland of the Negev desert of Israel under drip irrigation with saline water (up to 6.2 dS m−1) was studied in the years 1992–1997. The objective of the study was to determine the effects of saline water irrigation on potato production in an arid environment with special focus on the interactions with weather conditions. Although yields were often high, salinity effects were evident in some years. Thus 1992 and 1996 yields were 6–7 kg m−2 and showed no significant effect of salinity, while a pronounced drop in yield with increasing salinity was observed in 1993 and 1994. Analysis of weather data for 1993–1994 suggests that the decline in yield was due to interactions between saline irrigation and prolonged heat wave events occurring during crop development. Further experimental work (1997) revealed that tuber yield was most sensitive to combined salt and heat stress when heat waves occurred at 40–60 days after emergence. The combined stress apparently leads to the collapse of mechanisms for avoiding salt accumulation in young expanding leaves, resulting in failure of vegetative growth recovery and a consequent reduction in the leaf area index and canopy functioning. The relationship between tuber sink demand and available photoassimilate supply at certain stages of plant development is discussed with reference to the ability of the potato plant to recover from the combined stress.  相似文献

A Tuber melanosporum plantation established in 1994/1995 on Kibbutz Bar’am (in the Upper Galilee, Israel) gradually lost its T. melanosporum mycorrhiza. In 1999, T. aestivum inoculated seedlings were inadvertently introduced into the plantation to fill the gaps between trees. A single T. melanosporum fruit body was found in 1999. Although no truffles were found after 1999 and until 2009, in that year and in 2010, truffles were collected. Morphological and molecular analyses proved these to be T. aestivum. Thus, the intentionally introduced T. melanosporum mycorrhiza was replaced by that of another introduced mycorrhizal fungus, T. aestivum. Local oak species produced higher yields compared to introduced host species known to be good T. melanosporum plant symbionts. The yield was comparable to that reported for young commercial orchards, but the fruiting season was earlier than in Europe.  相似文献

The effect of various combinations of dinitro-o-cresol (DNOC) and narrow-distillation-range mineral oil (NRO) was tested during 4 consecutive years on breaking bud-rest and on fruiting of ‘Golden Delicious’ apple trees under mild winter conditions. Breaking of lateral leaf buds was directly correlated with oil concentration between 2.4 and 8.8%, while DNOC did not increase bud break at concentrations over 0.12% in the rest-breaking spray. Lower phytotoxicity of NRO compared with winter oil allowed safe use of higher concentrations of the former.Increased lateral leaf bud-break resulted in formation of more spurs and thus increased the yield in subsequent years. These higher yields were not accompanied by reductions in mean fruit weight or in trunk diameter, suggesting an increase in overall bearing potential of the tree by the best rest-breaking treatments.  相似文献

The mechanical behaviour of material bone can be completely described by a group of material properties. The mechanical behaviour of the entire bone organ, however, is much more difficult to predict; it is the result both of the properties of the material of which it is made, and of the geometric spatial architecture in which this is arranged. This review first describes material bone in terms of its complex, graded and hierarchical structure. Basic concepts used in the field of mechanics of materials are defined and explained and then used to describe the mechanical properties of whole bone. Some clinical implications of these properties are provided. Commonly used mechanical testing methods for the study of the mechanical behaviour of whole bone are reviewed and the technical difficulties associated with them are discussed.  相似文献

Classical mechanical methods for testing whole bone have been critically assessed in a previous review where their limitations in terms of precision, accuracy and the amount of data yielded were described. This article describes the use of optical metrology methods and their novel adaptation to the study of whole bone response to mechanical load. Such methods overcome many of the limitations of mechanical testing: they do not require contact with the tested sample, are non-destructive, can be conducted on wet samples, and results comprise deformation maps of entire surfaces. The concepts upon which each method is based are reviewed, and examples of their use in biomechanical studies of bone are presented. Potential future applications that are expected to make significant contributions to the understanding of whole bone mechanics are outlined.  相似文献
