Mastitis is the most common infectious disease affecting dairy cattle; in addition, it remains the most economically important disease of dairy industries around the world. Streptococcus agalactiae, a contagious pathogen associated with subclinical mastitis, is highly infectious. This bacterium can cause an increase in bulk tank bacterial counts (BTBC) and bulk tank somatic cell counts (BTSCC). The microbiological identification of S. agalactiae in samples from bulk tanks is an auxiliary method to control contagious mastitis. Thus, there are some limitations for time-consuming cultures or identification methods and additional concerns about the conservation and transport of samples. Bulk tank samples from 247 dairy farms were cultured and compared through polymerase chain reaction (PCR), directed to 16S rRNA genes of S. agalactiae, followed by BTBC and S. agalactiae isolation. The mean value of BTBC was 1.08×10(6) CFU mL(-1) and the bacterium was identified through the microbiological method in 98 (39.7%; CI(95%)=33.8-45.9%) and through PCR in 110 (44.5%; CI(95%)=38.5-50.8%) samples. Results indicated sensitivity of 0.8571±0.0353 (CI(95%)=0.7719-0.9196) and specificity of 0.8255±0.0311 (CI(95%)=0.7549-0.8827). The lack of significant difference between microbiological and molecular results (κ=0.6686±0.0477 and CI(95%)=0.5752-0.7620) indicated substantial agreement between the methods. This suggests that PCR can be used for bulk tank samples to detect contagious mastitis caused by S. agalactiae. 相似文献

Cork oak (Quercus suber) forests are acknowledged for their biodiversity and economic (mainly cork production) values. Wildfires are one of the main
threats contributing to cork oak decline in the Mediterranean Basin, and one major question that managers face after fire
in cork oak stands is whether the burned trees should be coppiced or not. This decision can be based on the degree of expected
crown regeneration assessed immediately after fire. In this study we carried out a post-fire assessment of the degree of crown
recovery in 858 trees being exploited for cork production in southern Portugal, 1.5 years after a wildfire. Using logistic
regression, we modelled good or poor crown recovery probability as a function of tree and stand variables. The main variables
influencing the likelihood of good or poor crown regeneration were bark thickness, charring height, aspect and tree diameter.
We also developed management models, including simpler but easier to measure variables, which had a lower predictive power
but can be used to help managers to identify, immediately after fire, trees that will likely show good crown regeneration,
and trees that will likely die or show poor regeneration (and thus, potential candidates for trunk coppicing). 相似文献

In the Iberian Peninsula Mediterranean oak forests have been transformed into a mosaic landscape of four main patch-types:
forests, savannas, shrublands and grasslands. We used aerial photographs over a period of 45 years (1958–2002) to quantify
the persistence and rates of transitions between vegetation patch-types in southern Portugal, where cork oak is the dominant
tree species. We used logistic regression to relate vegetation changes with topographical features and wildfire history. Over
the 45 years, shrublands have been the most persistent patch-type (59%), and have been expanding; forests are also persistent
(55%) but have been decreasing since 1985; savannas and grasslands were less persistent (33% and 15%, respectively). Shrublands
persistence was significantly correlated with wildfire occurrence, particularly on southern exposures after 1995. In contrast,
forest persistence decreased with wildfire occurrence, and forests were more likely to change into shrublands where wildfire

The impact of land‐use intensity is evaluated through changes in the soil properties in different areas of the traditional central Spanish landscape. Soil organic carbon (SOC) content, bulk density, aggregate stability and water‐holding capacity (WHC) in the topsoil of active and abandoned vineyards, livestock routes (LR) and young Quercus afforested areas were analysed. These different types of land use can be interpreted as having a gradient of progressively less impact on soil functions or conservation. As soil use intensity declines, there is an increase in SOC content (from 0.2 to 0.6%), WHC (from 0.2 to 0.3 g H2O per g soil) and aggregate stability (from 4 to 33 drop impacts). Soils beneath vines have lost their upper horizon (15 cm depth) because of centuries‐old tillage management of vineyards. Except for an increase in bulk density (from 1.2 to 1.4 g/cm3), there were no differences in soil characteristics 4 yr after the abandonment of vine management. LR can be considered sustainable uses of land, which preserve or improve soil characteristics, as there were no significant differences between topsoil from LR and that from a 40‐yr‐old Quercus afforested area. SOC content, one of the main indicators for soil conservation, is considered very low in every case analysed, even in the more conservative uses of land. These data can be useful in understanding the slow rate of recovery of soils, even after long‐term cessation of agricultural land use. 相似文献

The genetics of the prion protein gene (PRNP) play a crucial role in determining the relative susceptibility to transmissible spongiform encephalopathies (TSEs) in several mammalian species. To determine the PRNP gene variability in European red deer (Cervus elaphus), roe deer (Capreolus capreolus) and chamois (Rupicapra rupicapra), the PRNP open reading frame from 715 samples was analysed to reveal a total of ten single nucleotide polymorphisms (SNPs). In red deer, SNPs were found in codons 15, 21, 59, 78, 79, 98, 136, 168 and 226. These polymorphisms give rise to 12 haplotypes, and one of which is identical to the PRNP of American wapiti (Rocky Mountain elk, Cervus elaphus nelsoni). One silent mutation at codon 119 was detected in chamois and no SNPs were found in roe deer. This analysis confirmed that European wild ruminants have a PRNP genetic background that is compatible with TSE susceptibility, including chronic wasting disease. 相似文献

ObjectiveThe present study aimed to investigate the influence of methadone on cardiorespiratory parameters, electrocardiogram and clinical sedation in dogs. Further possible side effects are reported.Study designProspective experimental cross-over study.DogsEight, 1–4-year-old, various breeds of dogs of both genders weighing 9–36 kg.MethodsEach dog was treated three times: methadone 0.3 mg kg?1 (M0.3), 0.5 mg kg?1 (M0.5) and 1.0 mg kg?1 (M1.0) intramuscularly. Respiratory rate, heart rate and arterial blood pressure were recorded as well as electrocardiographic evaluation of lead II. Clinical sedation in each treatment received a score (0–3) after drug administration and at 30 minute intervals until scores and measurements returned to baseline values.ResultsA significant decrease in heart rate was seen with each dose of methadone and bradycardia (HR<60 bpm) was noted in a few dogs at each dose. A clinically significant arrhythmia occurred in one dog at 1 mg kg?1 that required reversal with butorphanol. There was no significant difference in SAP, MAP and DAP between treatments. Some side effects such as salivation, defecation, vocalization and panting, after administration of methadone were observed. There were no differences in mean values of heart rate, P-wave and QRS complex duration and QT interval between treatments.Conclusion and clinical relevanceMethadone administration was associated with panting and a decrease in heart rate at all doses tested in this study. The cardiac rhythm should be monitored carefully in dogs when methadone is administered on its own, especially at higher doses. 相似文献
