Development of new semen cryopreservation techniques improving sperm survival and ensuring availability of viable spermatozoa for a prolonged time‐period after AI is promising tools to reduce sensitivity of timing of AI and enhance overall fertility. The SpermVital® technology utilizes immobilization of bull spermatozoa in a solid network of alginate gel prior to freezing, which will provide a gradual release of spermatozoa after AI. The objective of this study was to compare post‐thaw sperm quality and in vitro sperm survival over time of Norwegian Red bull semen processed by the SpermVital® (SV) technology, the first commercialized production line of SpermVital® (C) and by conventional procedure applying Biladyl® extender (B). Post‐thaw sperm motility was not significantly different between SV, C and B semen (p > .05). However, sperm viability and acrosome intactness were higher for SV than C and B semen (p < .05). Small differences in DNA quality were observed (p < .05). Sperm viability after storage in uterus ex vivo was higher for SV than for C semen (p < .05). Furthermore, sperm survival in vitro over time at physiological temperature was significantly higher for SV semen than C semen as well as B semen during the incubation period of 48 hr (p < .05). In conclusion, the SpermVital® technology is improved and is more efficient in conserving post‐thaw sperm quality and results in higher sperm viability over time in vitro for SV than for C and B semen.  相似文献

 以大麦黄矮病毒二叉蚜和麦长管蚜专化株的病毒核酸为模板,以小牛胸腺DNA为引物,合成cDNA的第一条链,再用缺口翻译法合成第二条链,然后采用加装BamH1人工接头的方法将ds-cDNA插入到质粒载体pUC8中,重组质粒于大肠杆菌JM—83中进行克隆,以克隆的颜色变化选择含有外源DNA的克隆,再用病毒核酸制备的探针筛选真正病毒cDNA插入的克隆。重组质粒中ds-DNA的插入长度在300—1600bp之间。用缺口翻译法制备质粒DNA分子探针检测同源病毒液,反应灵敏度在100pg-1ng之间。应用cDNA探针检测不同病毒和病毒株系,从中筛选出黄矮病毒株系专化克隆系,黄矮病毒专化克隆系和黄矮病毒组专化克隆系.  相似文献

Management of prion diseases in livestock would benefit greatly from availability of a validated blood test. A promising immunocapillary electrophoresis technique (also known as capillary electrophoresis fluoroimmunoassay) to detect abnormal prion protein in blood from live sheep is evaluated here. Capillary electrophoresis fluoroimmunoassay was applied to analysis of extracted blood from scrapie-exposed sheep (n = 87; 347 samples) at various stages of incubation, and to control sheep (n = 194; 489 samples). Overall, test values for the control and test populations were not significantly different, and a similar proportion of control (7%) and test (10%) sheep were classified as positive. Over 2-3 month intervals from birth until clinical disease, test specificity and sensitivity ranged from 66.7% to 100% and 0% to 66.7%, respectively, indicating poor diagnostic performance at all stages of pathogenesis. In routine application, in its present form, the capillary electrophoresis fluoroimmunoassay procedure proved to be insufficiently robust for use as a blood test for scrapie diagnosis.  相似文献

Abstract— The daily riboflavin excretion of five adult cats has been studied for periods ranging from 7 to 11 days. The cats were housed in metabolism cages and were provided with a diet of cooked rabbit meat, wholemeal bread and water. The total amount of riboflavin excreted does not appear to be related to the dietary intake. The concentration of riboflavin in the urine does appear to be characteristic for a given cat under our experimental conditions.
Résumé— L'excrétion quotidienne de riboflavine chez cinq chats adultes a étéétudié pendant des périodes allant de 7 à 11 jours. Les chats étaient casés dans des cages à métabolisme et recevaient un régime de viande de lapin cuite, de pain entier et d'eau. La quantité totale de riboflavine excrétée ne semble pas être en rapport avec la quantité ingérée. Dans les conditions expérimentales que nous avons utilisées la concentration urinaire de riboflavine parait caractéristique pour un chat donné.
Zusammenfassung— In einer von 7 bis zu 11 Tagen reichenden Zeitspanne wurde untersucht, wieviel Laktoflavin fünf ausgewachsene Katzen täglich ausscheiden. Die Katzen wurden in Käfigen untergebracht, in denen der Stoffwechsel ständig überprüft werden kann, und sie wurden mit einer aus gekochtem Kaninchenfleisch, Vollkornbrot und Wasser bestehenden Diät gefüttert. Die Gesamt-menge des ausgeschiedenen Laktoflavins scheint in keiner Beziehung zu der Nahrungsaufnahme zu stehen. Die Konzentration des Laktoflavin im Urin scheint für eine unter unseren Versuchs-bedingungen stehende Katze charakteristisch zu sein.  相似文献

The occurrence of equine arteritis virus in Australia   总被引:2,自引:0,他引:2  
This paper reports the first isolation of equine arteritis virus (EAV) in Australia and serological evidence of exposure to EAV in Australian horses. Twelve Standardbred stallions imported from North America were found to shed EAV in semen. One hundred and seven stallions were tested for serum antibodies to EAV and 73% of Standardbred stallions tested were seropositive as compared to 8% of Thoroughbred stallions. Serum antibody was detected in 71% of Standardbred mares, 6% of Standardbred racehorses and 1% of Thoroughbred mares and racehorses. Examination of stored serums demonstrated that EAV had been present in Australia since at least 1975.  相似文献

The performance of amorphous organic photorefractive (PR) materials in applications such as optical data storage is generally limited by the concentration of active molecules (chromophores) that can be incorporated into the host without forming a crystalline material with poor optical quality. In polymeric PR systems described previously, performance has been limited by the necessity of devoting a large fraction of the material to inert polymer and plasticizing components in order to ensure compositional stability. A new class of organic PR materials composed of multifunctional glass-forming organic chromophores is described that have long-term stability and greatly improved PR properties.  相似文献

Late Cretaceous (Cenomanian) fossils discovered in the Kem Kem region of Morocco include large predatory dinosaurs that inhabited Africa as it drifted into geographic isolation. One, represented by a skull approximately 1.6 meters in length, is an advanced allosauroid referable to the African genus Carcharodontosaurus. Another, represented by a partial skeleton with slender proportions, is a new basal coelurosaur closely resembling the Egyptian genus Bahariasaurus. Comparisons with Cretaceous theropods from other continents reveal a previously unrecognized global radiation of carcharodontosaurid predators. Substantial geographic differentiation of dinosaurian faunas in response to continental drift appears to have arisen abruptly at the beginning of the Late Cretaceous.  相似文献

A method for quantification of tannins in wine was adapted to determine tannins added to turkey meat. Standard curves containing varying amounts of GSE [0, 0.5, 1.0, 1.5, 2.0, 2.5, and 5.0%, (w/w)] as a source of tannins were developed. The R(2) value of the mean standard curve was 0.9992. The overall percent recovery of GSE in meat was determined to be 54.78%. Results showed that estimation of GSE in four out of five of the spiked samples was less than or equal to 10%. It is unclear as to why spiked samples at 0.048 mg of GSE were always underestimated (25.0%). Overall, the method seems applicable for estimation of tannins in poultry meat and is probably applicable to estimation of tannins in other meat products.  相似文献
