A single dose of 5 x 10(8) bacilli of Pseudomonas pseudomallei by intratracheal injection resulted in acute (21 cases) or chronic (19 cases) melioidosis in 40 of 48 pigs. Fifteen (10 acute and 5 chronic) had been immunosuppressed by cyclophosphamide before inoculation. The major clinical signs were initial fever, marked neutrophilia and, in the acute cases, respiratory distress. There were no signs of the nasal and ocular discharge, paresis or diarrhoea seen in acute cases in south-east Asia. The cyclophosphamide treatment caused a significant decrease in the neutrophil count by 7 d after inoculation in all 15 immunosuppressed pigs, and all were culture positive at necropsy. Eight of the 33 non-treated pigs were culture negative at necropsy. Pigs overcoming the initial phase of infection had more abscess-like nodules that were bacteriologically sterile at necropsy than the pigs with acute cases of melioidosis. P. pseudomallei was isolated predominantly from the spleen, lungs and the injection site. Although only one strain was used in this study, it is likely that Australian strains of P. pseudomallei are not as virulent as the south-east Asian isolates.  相似文献

Two field trials, one with suckling Merino ewe lambs and the other with yearling Dohne Merino rams, are described. In these the anthelmintic efficacy of febantel (a benzimidazole), ivermectin, levamisole and morantel are compared, using the 1st stage larval reduction test. The mean natural log (+1 for zero values) of the post treatment larval counts of the treated groups was compared with that of the untreated controls and the percentage reduction used to assess anthelmintic efficacy. Febantel was only 87.4% effective against Teladorsagia in suckling lambs but the other anthelmintics were more than 99% effective against this genus. Efficacy against Haemonchus and Trichostrongylus ranged from 93.2%-100% for all 4 compounds. In the rams all compounds were 100% effective against Trichostrongylus, with the exception of morantel which was only 87.5% effective. None of the compounds were effective against Teladorsagia, particularly morantel, animals treated with which having more larvae than the controls. The interpretation of anthelmintic efficacy; the advantages of the first stage larval reduction test, compared with the faecal egg count reduction test; and the importance of incubating cultures at 30 degrees C for 24 h, in order to harvest first stage larvae, are discussed.  相似文献

Summary Although moisture requirements of Eisenia fetida are considered to be generally known, very few in-depth studies have been performed. The moisture preferences of juveniles as well as adults of this species were determined in moisture towers containing cow manure with a particle size bigger than 500 and smaller than 1000 m and an organic matter content of 54%. The clitellate worms showed a wider preference range than the juveniles but deposited most of their cocoons within the narrow range of 65%–70% moisture — a moisture range preferred by 80% of the juveniles. Growth studies of newly hatched worms at various moisture levels over a period of 90 days showed that growth rate and final biomass were influenced by moisture availability with a very low rate of growth at low as well as at high moisture levels. The change in biomass of clitellate specimens as well as their consecutively produced cocoons were monitored at different moisture levels. A clear relationship between mass of cocoons and biomass of the producing worms was established at presumably favourable moisture conditions. Furthermore, the relation between cocoon mass and the production rate of cocoons was also determined, which showed that cocoon mass increased concomitantly with increase in cocoon production. The study also showed that clitellum development was increased at moisture levels above 64%. The onset of cocoon production was, however, earliest at slightly lower moisture levels, from which it is concluded that the optimal moisture level for growth and development of clitella are not necessarily optimal for the production of cocoons.Dedicated to the late Prof. Dr. M.S. To understand the life cycle of an organism, it is important to understand the physiological processes that govern growth and reproduction. In this paper, we re-analyse a life-cycle data set for the earthworm Eisenia veneta, using a process-based model. The data set comprises measurements of body size and cocoon production over 200 days, at two temperatures (15-25 °C) and two densities (five and 10 worms per container, but with the same worm:soil weight ratio). The model consists of a set of simple equations, derived from Dynamic Energy Budget (DEB) theory. The dynamics of growth and reproduction are simultaneously described by the model, using very few parameters (five parameters for four curves). This supports the use of this model for efficient analysis of earthworm life-cycle data, and to interpret the effects of stressors. However, there was considerable inter-individual variation in the response, hampering the interpretation of the temperature and density effects. A temperature increase corresponded to an increase in the rate constants for growth and reproduction (with the same factor), without affecting the other parameters, as expected from DEB theory. Changing the earthworm density hardly affected the growth curves, but had an unexpected effect on reproduction: at higher densities, the worms start to produce cocoons at a larger body size and the maximum reproduction rate was lower. This study confirms the use of DEB as a reference model for earthworms, and using this model, we can recognise that temperature has a predictable effect on the life cycle of E. veneta. Furthermore, this analysis reveals that the effects of density are less clear and may involve a change in energy allocation that requires further study.  相似文献

The characterization of runoff-related pesticide input and the identification of areas of concern in the field are essential for a comprehensive assessment of risk and the planning of mitigation measures. To this end, the agriculturally-derived aqueous and particle-associated pesticide contamination of the Lourens River and its tributaries was measured in a comprehensive design. Sampling was performed in the beginning of April prior to the first rainfall of the wet season and in the middle of April during high water conditions following the first rainfall of 9.6 mm d-1. Pre-runoff samples indicated contamination with endosulfan at levels up to 0.06 μg L-1, while no other pesticides were detectable. Rainfall-induced runoff resulted in an increase in the levels of the pesticides endosulfan, deltamethrin, azinphos-methyl, chlorpyrifos, and procymidone up to levels of 0.35, 1.4, 0.6, 0.19 and 9 μg L-1, respectively in water samples. Endosulfan, azinphos-methyl, and chlorpyrifos were detected at maximum concentrations of 273, 152, and 245 μg kg-1 in suspended sediments. No increased pesticide levels were detected at a control site upstream of the agricultural area. The Lourens River received its contamination via the tributaries discharging the surrounding farming area. Contamination of the six tributaries depended on landuse and slope characteristics and enabled the identification of target sites for risk reduction approaches. Transient pesticide levels exceeded the target water quality limit proposed by the South African Department of Water Affairs and Forestry (DWAF). The Lourens River site downstream of the farming area has been identified as a site where potential toxic conditions could arise.  相似文献

Summary Observations in outdoor culture beds indicated that food availability and frequency of feeding influenced worm density. In order to study the role of feeding status on the growth and reproduction of Eisenia fetida, pre-clitellate specimens were kept for 180 days under controlled conditions. A control group was regularly provided with freshly produced cattle manure while the experimental groups were fed only sporadically and at some stage even starved by removing some of the substrate. Worm growth, maturation, and cocoon production were monitored. The role of food availability was highlighted by this study. Both worm growth and cocoon production were correlated closely with the feeding pattern that was followed. This study also showed that freshly produced, urine-free, cattle manure proved an excellent food source provided it was added in such a way as to prevent the development of anaerobic conditions. Regular feeding gave high growth and reproduction rates.  相似文献

A recently established Eragrostis curvula pasture 1,1 ha in size at Hennops River in the Transvaal Highveld was contaminated from 6 October 1976 until 20 May 1977 by 30 weaned Merinos, each infested with 7 000 infective larvae of H. contortus. On 2 November 1976 3 groups of weaned Merinos and again on 10 January 1977 a further group of Merinos were infested with 40 000 infective larvae of Trichostrongylus axei. From 19 November a single group of sheep, predosed with T. axei, and a control group grazed with the seeders for 8 weeks. After 6 weeks another group of sheep dosed with T. axei grazed with them, thus ensuring a 2-week overlap. This continued until autumn, when the last groups were removed. Efficacy against challenge reached Class C (greater than 50% effective against total worm burdens of H. contortus in greater than 50% of sheep) in early summer and autumn, but only reached significant levels of either P less than 0,01 or P less than 0,05 in summer. Sheep grazed on natural pastures heavily infested with infective larvae of Haemonchus contortus. Sixty-eight weaned Merinos were divided into 6 groups on Day 0 (23 November 1977), and on Day +14 (7 December) 79 Merinos were divided into 7 groups. There were 2 groups of undosed controls and other groups were either dosed with infective larvae of Trichostrongylus axei (bovine strain) only on Days 0 and +14, or in combination with H. contortus, or with subsequent doses of H. contortus, 28 days later. One group (Group 12) was dosed with T. axei and treated with a subcutaneous injection of di-iodonitrophenol (DNP) on Day +14. With the exception of 2 sheep, the sheep of the first 6 groups survived until slaughter in March and April 1978, while many sheep (43) of the latter 7 groups died or were killed in extremis from March-May. T. axei dosed on 23 November (Day 0) protected Group 2 by greater than 50% in greater than 50% of sheep. In the latter 7 groups the best results were achieved when DNP was combined with predosing with T. axei. The poor results were probably due either to delayed predosing with T. axei or a massive challenge in the wettest summer on record.  相似文献
