A collecting expedition has been carried out in the inland areas of the Basilicata Region, South Italy. Each of the 14 selected municipalities was subdivided in three strata based on the geographical distribution of population and orography: 1. city boundaries, 2. group of country houses, 3. isolated farm houses. Five sites were selected within each stratum/town for a total of 185 sites. In 101 sites crop species germplasm was collected and the farmer-maintainers were interviewed. Altogether 350 accessions were collected, the most common species were Capsicum annuum, Lycopersicon esculentum and Phaseolus vulgaris. A significant number of landraces are still preserved on farm, mostly in isolated farm houses (average age of farmers: 60?years) adopting traditional farming systems but a number of young farmers were also present. Perspectives for on farm conservation and management are discussed and public economic support for “farmer-maintainers” proposed.  相似文献

In a preliminary experiment on the induction of sexual maturity in Penaeus kerathurus by unilateral eyestalk ablation, spawning was obtained in 94% of the females treated. The time necessary to reach the spawning stage appeared to be closely related to the stage of natural physiological development of the gonads and the length of the photoperiod, the period between ablation and spawning decreasing from 69 days for females treated in November to about 10 days for females treated at the beginning of their natural reproductive period in May/June. Spawning after treatment was repeated up to eight times, sometimes at intervals of less than 10 days, without showing a decrease in the amount of fertile spawn. The mean number of fertile eggs per spawn was 78.400, which is not significantly different from the corresponding mean obtained previously after thermal stimulation of almost mature breeders; the hatching percentage, 67%, was also similar. Often only the anterior and median lobes of the ovaries are included in the spawning process. The discharged spawn is nearly always completely free of matrix material. It is anticipated that induction by eyestalk ablation could allow a considerable extension of the growing season and of the area suitable for culture.  相似文献

In this work an HPLC method for the determination of azadirachtin residues on olives was developed, and the field degradation kinetics of the pesticide was studied. In field trials the active ingredient (a.i.) decay had a half-life time of 0.8 days, which was too short to show a good efficacy of treatment. The mechanism of disappearance of the pesticide studied with model systems showed that it was unrelated to evaporation, thermodegradation, and co-distillation, but it was related to photodegradation. The high photodegradation rate of commercial formulations calls for the need to test different formulates in order to increase the persistence of the residue and thus the pesticide's efficacy.  相似文献

The aim of this work was to analyse factors which regulate sulphide mobility in the sediments of a dystrophic lagoon (Sacca di Goro, the southern lagoon of the Po river delta, Italy). In 1995-96, sediment oxygen demand and variations in organic matter content, redox potential, iron availability, inorganic sulphur concentrations and sulphate reduction rates were measured in sediment profiles at three stations (G, 4 and 17) representative of the main areas of the lagoon. Stations differed mainly in their salinity range and primary producer communities. High concentrations of reactive iron (110-275 µmol ml-1) and low sulphate reduction rates (0.8-16.1 mmol m-2 d-1) were measured in the sediment. Moreover, high concentrations of reactive ferric iron were detected in winter and spring at the stations closest to the freshwater inputs. Nevertheless, in summer, high concentrations of free sulphides were detected in the porewater, although most of the reactive ferrous iron was not sulphide-bound, indicating that not all of the reactive iron pool was available to buffer against sulphide release and thus measures of reactive iron pools may not be a good measure of the true buffering capacity of the sediment. Furthermore, a considerable production of sulphide may occur in the decaying Ulva biomass in the water column, where its concentration will be independent of the potential buffering capacity of iron in the sediment. Therefore when assessing the vulnerability of coastal lagoons to dystrophic events, both the size and availability of the reactive iron pool as well as the site of sulphide production must be taken into consideration.  相似文献

The essential oil obtained from the bark of Cinnamomum zeylanicum Blume (Lauraceae) and three of its main components, eugenol, (E)-cinnamaldehyde, and linalool (representing 82.5% of the total composition), were tested in two in vitro models of peroxynitrite-induced nitration and lipid peroxidation. The essential oil and eugenol showed very powerful activities, decreasing 3-nitrotyrosine formation with IC50 values of 18.4 microg/mL and 46.7 microM, respectively (reference compound, ascorbic acid, 71.3 microg/mL and 405.0 microM) and also inhibiting the peroxynitrite-induced lipid peroxidation showing an IC50 of 2.0 microg/mL and 13.1 microM, respectively, against 59.0 microg/mL (235.5 microM) of the reference compound Trolox. On the contrary, (E)-cinnamaldehyde and linalool were completely inactive.  相似文献

Cartap hydrolysis relative to its action at the insect nicotinic channel   总被引:2,自引:0,他引:2  
The insecticide cartap is the bis(thiocarbamate) derivative of 2-(dimethylamino)propane-1,3-dithiol, which on oxidation forms the natural toxicant nereistoxin (NTX) [4-(dimethylamino)-1,2-dithiolane]. Both cartap and NTX are ion channel blockers of the nicotinic acetylcholine receptor (nAChR). Cartap was originally proposed to act only after metabolic conversion to NTX and later suggested to block directly without activation. The present study uses a new approach to differentiate these hypotheses, that is, pH effects on channel-blocking activity and hydrolysis rates. As controls, mecamylamine (the classic channel blocker) and NTX are stable and similar in channel-blocking potency ([(3)H]thienylcyclohexylpiperidine binding assay, honeybee nAChR) at pH 6.1-8.4. In contrast, cartap is >200-fold more effective at pH 7.4 than at pH 6.1, indicating that it undergoes hydrolytic activation. Cartap slowly hydrolyzes to cartap monothiol at pH 6.1 but quickly forms the dithiol and some NTX at pH 7.4. The relationship between potency and hydrolysis products at various pH ranges suggests that cartap dithiol is the most plausible blocking agent.  相似文献

Penaeus japonicus Bate is absent on the Italian coasts, but it is considered a very promising species for national aquaculture. To develop the shrimp-culture on a commercial basis, a convenient method for producing eggs and larvae of Penaeus japonicus was studied. A stock of 156 females, age class 1+ and F3, underwent unilateral eyestalk ablation and they were stocked with males in three plastic tanks (bottom area 14 m2) at a density of 25 individuals/m2 and a sex ratio of 1:1. Conditions of environment and management consisted of: temperature 21±3°C; natural light and photoperiod; total exchange of sea water every day; feeding on mussels. A month after eyestalk ablation the females were inspected every 7–10 days; mature females were transferred to the spawning tanks. Females with well developed gonads averaged 25.0% of the population of females at every inspection; 51.3% of spawners were impregnated with spermatophores; others were inseminated artificially by the method of introducing spermatophores into the thelycum. In six inspections (carried out during a period of 89 days), 3,028,525 eggs were released; a female released 17,500 eggs on the average (maximum was 123,000 eggs, minimum 4,340). The mean rate of fertilized eggs was 52.7%—67.7% from naturally inseminated females and 7.5% from the artificially inseminated ones. The mean hatching rate was 30.9%—40.1% from natural insemination and 3.3% from artificial insemination. Total amount of viable nauplii was 820,550.  相似文献

Conifer bark beetles are often associated with fungal complexes whose components have different ecological roles. Some associated species are nutritionally obligate fungi, serving as nourishment to the larvae, whereas others are pathogenic blue-stain fungi known to be involved in the interaction with host defenses. In this study we characterized the local and systemic defense responses of Scots pine (Pinus sylvestris L.) against Ophiostoma brunneo-ciliatum Math. (a blue-stain pathogen) and Hyalorhinocladiella macrospora (Franke-Grosm.) Harr. (a nutritional fungus). These fungi are the principal associates of the pine engraver beetle, Ips acuminatus (Gyll.). Host responses were studied following inoculation with the fungi, singly and as a fungal complex, and by identifying and quantifying terpenoids, phenolic compounds and lignin. Although the length of the necrotic lesions differed between control (wound) and fungal treatments, only two compounds (pinosylvin monomethyl ether and (+)-α-pinene) were significantly affected by the presence of the fungi, indicating that Scots pine has a generic, rather than specific, induced response. The fact that both nutritional and blue-stain fungi triggered comparable induced defense responses suggests that even a non-pathogenic fungus may participate in exhausting host plant defenses, indirectly assisting in the beetle establishment process. Our findings contribute to the further development of current theory on the role of associated fungal complexes in bark beetle ecology.  相似文献

Diagnosis and control of dourine is strongly based on serological evidence, but knowledge of the humoral response of horses during infection is limited. In this study we developed a chemiluminescent immunoblotting (cIB) assay to characterise the Trypanosoma equiperdum antigen pattern recognised by IgGs from naturally or experimentally dourine-infected horses and analyse the kinetics of IgG humoral response following the infection. One compounding factor is that sera from uninfected animals often cross-react with T. equiperdum antigens. Development of the cIB assay was based on the hypothesis that serum IgGs from healthy and infected animals recognise different T. equiperdum antigen patterns. We used sera from 8 naturally infected horses which had recovered from Italian outbreaks and 2 experimentally infected mares. In addition, sera from 10 healthy control animals, eight of which were CFT positive but IFA negative for dourine, were collected from disease free regions. Sera were compared by the complement fixation test (CFT), indirect immune fluorescence (IFA) and the cIB assay.cIB analysis revealed that IgGs from infected horses, in contrast to IgGs from healhty horses, specifically recognise a T. equiperdum antigenic profile with low molecular weight bands ranging between 16 and 35 kDa. A time course experiment indicated that IgGs specific for the 16–35 kDa parasite protein fraction appear 17 days post-infection. The cIB assay confirmed all ten infected animals as positive and all controls as negative. This study demonstrated that analysis of IgGs by cIB can provide clear confirmation of trypanosome infection in horses, suggesting that this technique can be applied as a confirmatory serological test for dourine infection.  相似文献

The persistence and degradation of rotenone and its primary degradation product 12a beta-hydroxyrotenone in soils were determined under standardized laboratory conditions in the dark at 20 or 10 degrees C and at 40% of water holding capacity. Degradation experiments were carried out on two types of soil collected in southern Italy, a silt clay loam (SCL) and a loamy soil (L). A kinetic model was developed to describe degradation rates of rotenone, taking into account the production, retention, and degradation of the main metabolites. The DT50 values of rotenone and 12a beta-hydroxyrotenone, were 8 and 52 days in SCL soil, and 5 and 23 days in L soil at 20 degrees C, respectively. However, at 10 degrees C a tendency for slower degradation of rotenone and 12a beta-hydroxyrotenone was observed (25 and 118 days in SCL and 21 and 35 days in L soils, respectively). The differences were significant for most data sets. Temperature had a strong effect on degradation; a 10 degrees C increase in temperature resulted in a decrease in the DT50 value by a factor of 3.1 and 2.2 in SCL and of 4.2 and 1.4 in L soils for both rotenone and 12a beta-hydroxyrotenone, respectively. Results show that the degradation rates of both rotenone and 12a beta-hydroxyrotenone were greatly affected by temperature changes and soil physicochemical properties. The degradation reaction fits the two compartment or the multiple compartment model pathways better, which clearly indicates a rather complex rotenone degradation process in soils. Results provide further insights on the rates and the mechanisms of rotenone degradation in soils, aiming to more clearly describe the degradation pathway of chemical residues in the environment.  相似文献
