A study was conducted in the USA to determine whether transmissible gastroenteritis (TGE) virus could be transmitted from carcases of slaughtered pigs. Transmissible gastroenteritis virus was transmitted to 6-day-old piglets by dosing with homogenates of muscle and lymph node collected from 500 clinically normal pigs at the time of slaughter. All piglets in 2 separately housed litters showed clinical signs of TGE with 5 piglets dying within 10 d of oral dosing with homogenates. Transmissible gastroenteritis virus was isolated from 2 of these piglets and all piglets developed TGE antibody. Transmissible gastroenteritis virus was not isolated in tissue culture from muscle and lymph node homogenates, but was isolated from 4 (0.8%) of 500 tonsil samples collected from the same pigs. A survey of 250 serum samples provided an estimate of the prevalence of slaughtered pigs with TGE antibody of 34.8% in the sample population. The results indicate that carcases of some pigs from TGE endemic areas contain viable TGE virus, and that there would be a substantial risk of introducing TGE virus into Australia by the importation of uncooked pig meat from these areas.  相似文献

Equine skeletal muscle was histologically and histochemically analyzed in orderto measure fiber composition. Well-sampled open muscle biopsies were obtained from the semitendinosus muscle in select yearling Thoroughbreds. Each biopsy was sectioned and stained for the Harris Hematoxylin and Eosin and the modified Gomori Trichrome in order to demonstrate the basic muscle fiber morphology delineating intra and extracellular elements. A thirdstain, CA++ activated myosin Adenosine Triphosphatase at pH 9.4 was used to differentiate between specific types on the basis of enzyme activities. Subsequent differentiation of Type I slow twitch and Type II fast twitch muscle fibers was assessed on the basisof staining intensity. The high predominance of Type II fast twitch fibers ranged from 85.1 to 100 % with a mean of 90.5 %. Racing records, as two- and three-year-olds, were monitored in order to qualitatively compare histochemical results with performance achievements. While caution must be exercised, results of this investigation suggested that it may be possible, at an early age to partially predict the potential of each horse to adapt to training and to compete successfully in races of varying distances.  相似文献

Organic matter extracted with 9 : 1 formic acid : acetylacetone from twenty-five soils of widely varying organic content and precipitated with excess di-isopropyl ether was hydrolysed with 6N HC1. The relationships between the proportions of acid-soluble N, acid-insoluble N, amino-N, and ammonium-N thus produced, and the properties of the original soil were examined. The distribution of N in acid hydrolysates was similar to that reported in the literature for material isolated by neutral and alkaline extractants. The relatively large proportion of N released as ammonium-N from organic matter obtained from soils of high pH and low C : N ratio may derive from organically fixed ammonia. The low levels of amino-N in organic matter extracted by formic acid from soils of high clay content imply the existence of clay-amino complexes that resist extraction although formation of such complexes during extraction cannot be ruled out.  相似文献

A study was made to characterize the effect of the duration of regrowth on the pattern of changes in the major physiological processes involved in the net accumulation of herbage, and so to provide a rational basis for optimizing production under rotational grazing.
During regrowth following a severe defoliation, rates of canopy photosynthesis, and so the rate of production of new leaves, increased rapidly but there was a delay before there was a corresponding increase in the rate of leaf death. Although the amount available for harvest (crop live dry weight, W ) continued to increase as the duration of regrowth was extended from 'short' (12–13 days), through 'medium' (19–23 days) to 'long' (30-34 days), there was not a continued increase in the average growth rate – the increase in the weight of the crop, ( W – Wo ), divided by the duration of regrowth, t , In this study, the average growth rate (based on changes in the weight of lamina alone) increased as the duration of regrowth was extended from 12-13 to 19-23 days but changed little as the duration of regrowth was extended from 19-23 days to 30-34 days. In spring and summer, elongating stems increased the average growth rate (of lamina plus stem) up to 30-34 days but the accumulated stem material could not reliably be harvested by sheep.
A mathematical model was used to explain why the average growth rate is characteristically insensitive to the duration of regrowth beyond a given minimum duration. For practical purposes, we suggest from the results of this study that regrowths of at least 14 days but less than 28 days will be effective in achieving not only close to the maximum average growth rate of highly digestible material, but also in sustaining a densely tillered, leafy sward which regrows rapidly from severe defoliation and is more reliably harvested (utilized) by sheep.  相似文献

A low molecular weight poly(ethyleneoxide)-poly(butadiene) (PEO-PB) diblock copolymer containing 50 weight percent PEO forms gigantic wormlike micelles at low concentrations (<5 percent by weight) in water. Subsequent generation of free radicals with a conventional water-based redox reaction leads to chemical cross-linking of the PB cores without disruption of the cylindrical morphology, as evidenced by cryotransmission electron microscopy and small-angle neutron scattering experiments. These wormlike rubber micelles exhibit unusual viscoelastic properties in water.  相似文献

Phenylbutazone (PBZ) is a nonsteroidal anti‐inflammatory drug used in the treatment of chronic pain and arthritis. Topical and transdermal administration of PBZ would be beneficial in large animals in terms of minimizing gastro‐intestinal ulcerations and other side effects, easy administration to legs and joints and minimizing the dose to reduce systemic toxicity of the drug. A topical liposomal preparation with different concentrations of a mono‐substituted alkyl amide (MSA) and PBZ was formulated. The formulations were evaluated by in vitro skin‐permeation kinetics through deer skin using Franz diffusion cells. By increasing drug loading from 1% to 5% w/w, the steady‐state flux (μg/cm2/h) of PBZ was increased twofold (P < 0.001). Similarly, by increasing the MSA concentration from 0% to 4%, the steady‐state flux (μg/cm2/h) of PBZ was increased twofold (P < 0.001). Overall, by increasing the drug load and the use of an appropriate amount of the penetration enhancer, the steady‐state flux of PBZ through skin was increased fourfold (P < 0.001). MSA at both 2% and 4% w/w concentrations significantly increased the skin levels of PBZ as compared with control (P < 0.05). In conclusion, MSA served as an effective skin‐penetration enhancer in the liposomal gel of PBZ for deer.  相似文献

Natural clay minerals can play an important role in crude remediation of wastewater polluted with the heavy metals (HMs) Cu,Zn and Ni.The presence and timing of addition of natural dissolved organic matter (DOM) have a significant effect on the HM removal by clay mineral sorbents.However,the influence of the presence of DOM on the remediation of the used clay mineral sorbents once saturated with HMs is largely unknown.To resolve this,clay mineral-rich soil column of varying composition,loaded (i) with Cu,Zn and Ni only,(ii) first with DOM followed by Cu,Zn and Ni,or (iii) with DOM,Cu,Zn and Ni simultaneously,was used in a set of desorption experiments.The soil columns were leached with 0.001 mol L-1 CaCl2 dissolved in water as control eluent and 0.001 mol L-1 CaCl2 dissolved in DOM as treatment eluent.During the preceding loading phase of the sorbent,the timing of DOM addition (sequential or concurrent with HMs) was found to have a significant influence on the subsequent removal of the HMs.In particular when the column was loaded with DOM and HMs simultaneously,largely irreversible co-precipitation took place.Our results indicate that the regeneration potential of clay mineral sorbents in wastewater treatment will be significantly reduced when the treated water is rich in DOM.In contrast,in manured agricultural fields (where HMs enter together with DOM),HM mobility will be lower than expected from interaction dynamics of HMs and clay minerals.  相似文献

Melatonin, the major secretory product of the pineal gland, scavenges a variety of reactive oxygen and nitrogen species in vivo and in vitro, indicating that melatonin is a potent function as an antioxidant. The objective of this study was to investigate the effect of melatonin in the presence or absence of hydrogen peroxide (H2O2) on sperm characteristics (motility, viability, survival rate, membrane integrity, lipid peroxidation (LPO) and mitochondria activity) and also to examine the developmental rates to the blastocysts stage of porcine oocytes fertilized in vitro with semen treated with or without melatonin (100 nm ) in the presence or absence of H2O2 (250 μm ). The sperm were treated with melatonin in the presence or absence of H2O2 for 3, 6, 9 and 12 h at 37°C and then analysed for the sperm characteristics. The porcine embryos were produced by in vitro maturation and in vitro fertilization (IVM/IVF) using semen treated with or without melatonin (100 nm ) in the presence or absence of H2O2 (250 μm ) for 6 h. The semen characteristics, including motility, viability, survival rate, membrane integrity and mitochondria activity, were higher in the groups that were treated with melatonin in comparison to other groups, irrespective of incubation periods. Malondialdehyde levels in control, melatonin and melatonin + H2O2 groups were lower than H2O2 only group. A positive correlation was shown among motility, viability, survival rate and membrane integrity, but a negative correlation was observed between LPO and the other evaluation methods. The developmental rates to blastocysts of IVM/IVF porcine oocytes fertilized by semen treated with melatonin were significantly increased compared with any other groups, with the cell number of blastocysts shown to have a similar trend to the developmental rates. These results demonstrate that melatonin can improve the semen characteristics during in vitro storage and support the developmental ability of IVM/IVF embryos in pigs.  相似文献
